Design of a compact photonic-crystal-based polarizing beam splitter   总被引:5,自引:0,他引:5  
A compact polarizing beam splitter based on a photonic crystal (PC) directional coupler with a triangular lattice of air holes is designed and simulated. In the employed PC structure, transverse-electric (TE) light is confined with the photonic bandgap effect, while transverse-magnetic (TM) light is guided through an index-like effect. Due to the different guiding mechanisms, TM and TE light have strikingly different beat lengths, which is utilized to separate the two polarizations in a directional coupler no longer than 24.2 /spl mu/m. The two-dimensional finite-difference time-domain method of computation is used to evaluate the device performance. The extinction ratios are found to be around 20 dB for both TE and TM polarized light.  相似文献

South Africa has a decade of experience designing and implementing biodiversity offsets. In the absence of explicit national policy on biodiversity offsets, the country has relied on existing legal provisions in environmental law as the basis for offset requirements, supported by provincial guidelines. South Africa’s periodic national biodiversity surveys provide scientifically rigorous quantification and mapping for individual ecosystems and finer scale surveys identify biodiversity priority areas, primed as ‘offset receiving areas’. Yet despite enabling factors the use of offsets has frequently been inadequate to deliver intended biodiversity outcomes. Challenges include: (a) the absence of national policy to drive and shape offset implementation; (b) insufficient capacity to evaluate, design and implement offsets; (c) inconsistent decision-making; (d) problems establishing sustainable financing mechanisms; and (e) inadequate enforcement and monitoring, linked to poor drafting of licencing conditions and/or insufficient capacity to monitor implementation. South Africa’s experience provides valuable insights into the challenges and potential solutions for making offsets work for biodiversity conservation and offers important lessons for the development and implementation of biodiversity offsetting in other developing countries.  相似文献

Expression of recombinant proteins as translational fusionsis commonly employed to enhance stability, increase solubilityand facilitate purification of the desired protein. In general,such fusion proteins must be cleaved to release the mature proteinin its native form. The usefulness of the procedure dependson the efficiency and precision of cleavage and its cost perunit activity. We report here the development of a general procedurefor precise and highly efficient cleavage of recombinant fusionproteins using the protease chymosin. DNA encoding a modifiedpro-peptide from bovine chymosin was fused upstream of hirudin,carp growth hormone, thioredoxin and cystatin coding sequencesand expressed in a bacterial Escherichia coli host. Each ofthe resulting fusion proteins was efficiently cleaved at thejunction between the pro-peptide and the desired protein bythe addition of chymosin, as determined by activity, N-terminalsequencing and mass spectrometry of the recovered protein. Thesystem was tested further by cleavage of two fusion proteins,cystatin and thioredoxin, sequestered on oilbody particles obtainedfrom transgenic Arabidopsis seeds. Even when the fusion proteinwas sequestered and immobilized on oilbodies, precise and efficientcleavage was obtained. The precision, efficiency and low costof this procedure suggest that it could be used in larger scalemanufacturing of recombinant proteins which benefit from expressionas fusions in their host organism. The colour of bovine subcutaneous (sc) adipose tissue (carcass fat) depends on the age, gender and breed of cattle. Diet is the most important extrinsic factor but its influence depends on the duration of feeding. Cattle produced under extensive grass-based production systems generally have carcass fat which is more yellow than their intensively-reared, concentrate-fed counterparts and this is caused by carotenoids from green forage. Although yellow carcass fat is negatively regarded in many countries, evidence suggests it may be associated with a healthier fatty acid profile and antioxidant content in beef, synonymous with grass feeding. Nonetheless, management strategies to reduce fat colour of grass-fed cattle are sought after. Current research suggests that yellow colour of this tissue is reduced if pasture-fed cattle are converted to a grain-based diet, which results in accretion of adipose tissue and dilution of carotenoids. Colour changes may depend on the initial yellow colour, the carotene and utilisable energy in the finishing diet, the duration of finishing, the amount of fat accumulated during finishing and the rate of utilisation of carotene from body fat. Differences in nutritional strategies which cause differences in fatty acid composition may be reflected by differences in fat colour and carotenoid concentration. Fat colour and carotenoids are prominent among a panoply of measurements which can aid the authentication of the dietary history and thus to some extent, the origin of beef, although this potential utility is complicated by the simultaneous rather than discrete use of forages and concentrates in real production systems.  相似文献

Polycyclic aromatic hydrocarbon (PAH) air concentrations measured over the period 1992-2000 at the Canadian High Arctic station of Alert were subject to time-series analysis using dynamic harmonic regression (DHR). For most of the PAHs, the DHR model fit to the observed data was good, with DHR capable of interpolating over missing data points during periods when air concentrations were below detection limits. As expected, DHR identified seasonal increases in PAH air concentrations. However, it has also identified additional, subtler "seasonal" patterns as a series of harmonics with varying periodicity. For example, a regular summer high in air concentrations was apparent for many PAHs, particularly the lower molecular weight (two- to three-ringed) compounds, which may be attributed to summertime regional combustion events such as forestfires and/or revolatilization from surfaces (e.g., soil and oceans, as well as arctic surfaces). Comparison of wintertime PAH concentrations (where sigmaPAH ranged from 260 to 516 pg m(-3)) with an earlier arctic study did not reveal a reduction in PAH levels. However, removal of the seasonal components by DHR revealed a declining trend in PAH concentrations over the 1992-2000 period. For many lighter PAHs, this was typified by a linear decrease over the whole time series, although, for the higher molecular weight PAHs, a marked reduction was apparent in the first few years of sampling followed by a leveling off in concentrations by the mid/late-1990s. This behavior is similar to reported trends of other air pollutants in the Arctic, may be attributed to the decline in Soviet industry during the early 1990s, and has implications regarding the major PAH sources affecting the Arctic.  相似文献

Sudan I, II, III and IV dyes are banned for use as food colorants in the United States and European Union because they are toxic and carcinogenic. These dyes have been illegally used as food additives in products such as chilli spices and palm oil to enhance their red colour. From 2003 to 2005, the European Union made a series of decisions requiring chilli spices and palm oil imported to the European Union to contain analytical reports declaring them free of Sudan I–IV. In order for the USFDA to investigate the adulteration of palm oil and chilli spices with unapproved colour additives in the United States, a method was developed for the extraction and analysis of Sudan dyes in palm oil, and previous methods were validated for Sudan dyes in chilli spices. Both LC-DAD and LC-MS/MS methods were examined for their limitations and effectiveness in identifying adulterated samples. Method validation was performed for both chilli spices and palm oil by spiking samples known to be free of Sudan dyes at concentrations close to the limit of detection. Reproducibility, matrix effects, and selectivity of the method were also investigated. Additionally, for the first time a survey of palm oil and chilli spices was performed in the United States, specifically in the Washington, DC, area. Illegal dyes, primarily Sudan IV, were detected in palm oil at concentrations from 150 to 24 000 ng ml–1. Low concentrations (< 21 µg kg–1) of Sudan dyes were found in 11 out of 57 spices and are most likely a result of cross-contamination during preparation and storage and not intentional adulteration.  相似文献

Multiple stable isotope ratios (δ2H, δ13C, δ18O and δ34S) were measured in muscle, muscle lipids and lipid fractions collected from 28 lambs, subjected to a diet-switch and raised on two energy allowances (EAs), to determine tissue turnover and diet-tissue fractionation. The diet-muscle fractionations prior to the diet-switch were estimated to be −44.0‰, +1.9‰ and 0‰ for H, C and S, respectively, while the drinking water was demonstrated to be the main source of muscle O and thus δ18O variation. The diet-intra-muscular lipid fractionations prior to the diet-switch were estimated to be −172.7‰, −1.3‰ and −11.5‰ for H, C and O, respectively. The C half-lives of muscle were determined to be 75.7 and 91.6 days for animals receiving the high and low EA, respectively. Extracting temporally resolved pre-slaughter dietary information from meat by analysing bulk muscle, muscle lipids and muscle lipid fractions appeared to be not practicable due to possible incomplete turnover of lipids.  相似文献
