The tumor-promoting agent 4β-phorbol-12-myristate-13-acetate (TPA) is shown to be a potent stimulator of fatty acid synthesis in isolated rat hepatocytes. The maximal effect of TPA is seen at 10−6 M, and the concentration for half-maximal effect is ca. 10−8 M. Stimulation of fatty acid synthesis by TPA is shown not to require the presence of extracellular Ca++. TPA produces a significant increase in lactate and pyruvate accumulation. The possible involvement of protein kinase C in short-term regulation of fatty acid synthesis in the liver is discussed.  相似文献

The body distribution and tumor accumulation of polymers were evaluated using poly(vinyl alcohol) (PVA) which has the simplest chemical structure among water-soluble polymers, similar to poly(ethylene glycol). To study the effect of polymer size on the tumor accumulation, we used not only water-soluble PVA with different molecular weights but also PVA microgels prepared through gamma-irradiation of aqueous PVA solutions. The PVA specimens in the aqueous solution were intravenously injected to mice carrying a tumor mass at their footpad. Both types of PVA (water-soluble and microgel) of larger size were retained in the blood circulation for longer time periods and excreted more slowly from the kidney than those of smaller size. The plasma half-life period of PVA became longer with increasing size both for the water-soluble and microgel PVA, indicating that the body fate of PVA is governed only by the size. Both the water-soluble PVA and PVA microgels were accumulated in tumor tissue to a significantly greater extent than in normal tissue. The size dependence of the plasma half-life period and tumor accumulation was similar between the water-soluble PVA and the PVA microgels and the tumor accumulation became maximum around the size of 60 nm both for water-soluble and microgel PVA. A pharmacokinetical study demonstrated that the tumor uptake rate index of PVA decreased with the increase in PVA size. On the other hand, the greater the size, the larger the value of the area under the blood concentration-time curve (AUC). In addition, the PVA around 60 nm in diameter showed the smallest liver clearance. It was concluded that the balance between the uptake rate and the AUC as well as the liver clearance resulted in the maximum accumulation of PVA with the size of 60 nm.  相似文献

Grafting of N,N-dimethylacrylamide (DMAA) onto polytetrafluoroethylene (PTFE) has been carried out by using gamma rays from a 60Co source. Degree of grafting was affected by grafting conditions. Especially, solvent for the grafting was found to be a very significant parameter to obtain a higher grafting yield. When ethylacetate or acetone was used as the solvent, grafting yield greater than 5% was obtainable while the graftcopolymerization scarcely occurred in the presence of other kinds of solvent such as ethanol or water. Apparent activation energy of the grafting in ethylacetate was calculated to be 7.90 Kcal/mol, and initial grafting rate of it was proportional to 0.55 power of dose rate.  相似文献

The DNA properties have been deliberated to comprehend new functional bio-devices. However there are few reports on DNA nano-patterning to make them. Therefore, we have tried DNA nano-patterning by using a nanoimprint process. A substrate coated with a poly-L-lysine was heated at 120 degrees C for 5 min in atmosphere, and nanoimprint carried out at 120 degrees C, 6 MPa, 5 min, and we applied 1 mg/ml DNA solution on the substrate and immobilized with the poly-L-lysine. Finally the substrate was washed at twice in water and at once in hot water intensely. DNA lines that consist of lines at about 700 nm as line width was obtain, and the very fine lines correspond to convex patterns of the mold surface. These results suggest that the imprint would make the poly-L-lysine reform by exposing it under high pressure and high temperature. Therefore since DNA is immobilized with the amino group in the poly-L-lysine, a lot of amino group would expose on the surface by the imprint from the balk, and would be patterned DNA.  相似文献

A 56-year-old Japanese woman was referred to us for the treatment of lung cancer. On admission, the patient showed multiple bone metastases, including the skull, without brain metastasis. During chemoradiotherapy for the primary tumor and bone metastasis involving the thoracic spine, she suffered a fatal intracerebral hemorrhage. Since the patient had no risk factors for intracerebral hemorrhage, the skull bone metastasis was thought to be responsible for this event. At autopsy, penetration of the metastatic tumor from the skull bone into the dura, with direct invasion of the brain tissue, was confirmed histologically. A hematoma also was identified at the same site adjacent to the skull bone metastasis. To our knowledge, direct tumor invasion to the brain from a skull metastasis of non-small cell lung cancer has not been previously reported.  相似文献

The activity of hepatic triglyceride lipase (H-TGL) of plasma membranes isolated from rat liver is shown to be reduced by fasting. Refeeding restores the enzyme activity. The suppressed activity of H-TGL in streptozotocin diabetic rats is restored by insulin treatment. The behavior of the enzyme activites in both situation coincides with that of plasma insulin levels. The results suggest that the H-TGL of rat liver plasma membranes is under hormonal regulation by insulin.  相似文献
