Diabetes mellitus and related disorders significantly contribute to morbidity and mortality worldwide. Despite the advances in the current therapeutic methods, further development of anti-diabetic therapies is necessary. Mitochondrial dysfunction is known to be implicated in diabetes development. Moreover, specific types of mitochondrial diabetes have been discovered, such as MIDD (maternally inherited diabetes and deafness) and DAD (diabetes and Deafness). Hereditary mitochondrial disorders are caused by certain mutations in the mitochondrial DNA (mtDNA), which encodes for a substantial part of mitochondrial proteins and mitochondrial tRNA necessary for mitochondrial protein synthesis. Study of mtDNA mutations is challenging because the pathogenic phenotype associated with such mutations depends on the level of its heteroplasmy (proportion of mtDNA copies carrying the mutation) and can be tissue-specific. Nevertheless, modern sequencing methods have allowed describing and characterizing a number of mtDNA mutations associated with human disorders, and the list is constantly growing. In this review, we provide a list of mtDNA mutations associated with diabetes and related disorders and discuss the mechanisms of their involvement in the pathology development.  相似文献

Soluble poly(4-alkylstyrene) containing 4-methyl-, 4-tert-butyl, 4-dodecyl-, and 4-octadecylstyrene were formed either with monomers containing dyes-, ligands, or catalysts or with 5–10 mol% of 4-chloromethylstyrene and studied as supports for ligands or catalysts. Studies with dye-labeled polymers showed that polymers containing longer alkyl groups alone or at ca. 10 mol% loading are highly soluble in heptane can be used to separate and recycle organo- and transition metal catalysts.  相似文献

Self-healing coatings is a rapidly growing research area, where focus has mainly been on development of new approaches to the mechanism of self-healing. However, there is a growing need for investigation of practical issues related to formulation, application, and testing of true self-healing coatings. In this work, ways of reducing the size of poly(urea–formaldehyde) microcapsules, filled with linseed oil and intended for a microcapsule-based self-healing anticorrosive coating (above water exposure), are explored. The influence of microcapsules on epoxy coating performance is also studied. The actual self-healing effect was not part of this work. The synthesis parameters investigated are stirrer geometry, agitation rate, temperature, and stabilizer concentration. It was found that an increase in stirring rate, correct choice of temperature, and a high stabilizer concentration all caused a decrease in microcapsule size but were accompanied by excessive formation of nanoparticles. Thus, isolation of too large microcapsules has been performed by filtration utilizing a novel low-energy fluoropolymer-coated steel sieve. An estimation of the critical pigment (microcapsule) volume concentration (CPVC) was conducted using gloss measurements and a PVC ladder and found to be about 30 vol%. Due to the rather large capsules used (relative to the coating thickness), the low CPVC value can probably be ascribed to a fairly low packing efficiency in the coating, but this needs to be confirmed. Coating performance was evaluated using salt spray exposure and impact testing. Results of the impact testing showed that addition of microcapsules to a binder matrix did not compromise resistance of the coating to mechanical damage and led to formation of fewer and shorter cracks compared to a filler-containing coating. Flaking of the coating was also reduced. Results of the salt spray testing (3 weeks exposure) showed that with an increase of microcapsule content, in the interval 30–50 vol%, the extent of corrosion and potential coating delamination decreased and was identical to that of a full commercial anticorrosive coating.  相似文献

Poly(2-hydroxyethyl methacrylate) (PHEMA) embolization particles with enhanced haemostatic properties were prepared by bulk or suspension polymerisation of 2-hydroxyethyl methacrylate (HEMA) followed by particle soaking in ethamsylate solution. The particles accelerated thrombus formation as evidenced by blood analysis of rabbits with implanted emboli. Usefulness of both spherical and cylindrical PHEMA particles with enhanced haemostatic effect was demonstrated on the embolization of arterial anastomosis, fistulas of the lower extremity and abdominal cavity, haemangioma and arteriovenous malformation of the head of several children.  相似文献

We investigated the microwave absorbing properties of composite bulk samples with nanostructured and micron-sized fillers. As magnetic fillers we used magnetite powder (Fe3O4 with low magnetocrystalline anisotropy) and strontium hexaferrite (SrFe12O9 with high magnetocrystalline anisotropy). The dielectric matrix consisted of silicone rubber. The average particle size was 30 nm for the magnetite powder and 6 micro/m for the strontium hexaferrite powder. The micron-sized SrFe12O19 powder was prepared using a solid-state reaction. We investigated the influence of the filler concentration and the filler ratio (Fe3O4/SrFe12O19) in the polymer matrix on the microwave absorption in a large frequency range (1 / 18 GHz). The results obtained showed that the highly anisotropic particles become centers of clusterification and the small magnetite particles form magnetic balls with different diameter depending on the concentration. The effect of adding micron-sized SrFe12O19 to the nanosized Fe3O4 filler in composites absorbing structures has to do with the ferromagnetic resonance (FMR) shifting to the higher frequencies due to the changes in the ferrite filler's properties induced by the presence of a magnetic material with high magnetocrystalline anisotropy. The two-component filler possesses new values of the saturation magnetization and of the anisotropy constant, differing from those of both SrFe12O1919 and Fe3O4, which leads to a rise in the effective anisotropy field. The results demonstrate the possibility to vary the composite's absorption characteristics in a controlled manner by way of introducing a second magnetic material.  相似文献

Nanosized gold catalysts supported on doped ceria were prepared by deposition–precipitation method. A deep characterization study by HRTEM/EDS, XRD, FT-Raman, TPR and FTIR was undergone in order to investigate the effect of ceria modification by various cations (Sm3+, La3+ and Zn2+) on structural and redox properties of gold catalysts. Doping of ceria affected in different way catalytic activity towards purification of H2 via preferential CO oxidation. The following activity order was observed: Au/Zn–CeO2 > Au/Sm–CeO2 > Au/CeO2 > Au/La–CeO2. The differences in CO oxidation rates were ascribed to different concentration of metallic gold particles on the surface of Au catalysts (as confirmed by the intensity of the band at 2103 cm−1 in the FTIR spectra collected during CO–O2 interaction). Gold catalysts on modified ceria showed improved tolerance towards the presence of CO2 and H2O in the PROX feed. The spectroscopic experiments evidence enhanced reactivity when PROX is performed in the presence of H2O already at 90 K.  相似文献

This study proposed and demonstrated the application of a new Raman microscopy-based method for metabolic state-based identification and quantification of functionally relevant populations, namely polyphosphate accumulating organisms (PAOs) and glycogen accumulating organisms (GAOs), in enhanced biological phosphorus removal (EBPR) system via simultaneous detection of multiple intracellular polymers including polyphosphate (polyP), glycogen, and polyhydroxybutyrate (PHB). The unique Raman spectrum of different combinations of intracellular polymers within a cell at a given stage of the EBPR cycle allowed for its identification as PAO, GAO, or neither. The abundance of total PAOs and GAOs determined by Raman method were consistent with those obtained with polyP staining and fluorescence in situ hybridization (FISH). Different combinations and quantities of intracellular polymer inclusions observed in single cells revealed the distribution of different sub-PAOs groups among the total PAO populations, which exhibit phenotypic and metabolic heterogeneity and diversity. These results also provided evidence for the hypothesis that different PAOs may employ different extents of combination of glycolysis and TCA cycle pathways for anaerobic reducing power and energy generation and it is possible that some PAOs may rely on TCA cycle solely without glycolysis. Sum of cellular level quantification of the internal polymers associated with different population groups showed differentiated and distributed trends of glycogen and PHB level between PAOs and GAOs, which could not be elucidated before with conventional bulk measurements of EBPR mixed cultures.  相似文献

Human urokinase-type plasminogen activator (uPA) is poorly secreted and aggregates in the endoplasmic reticulum of yeast cells due to inefficient folding. A screen for Hansenula polymorpha mutants with improved uPA secretion revealed a gene encoding a homologue of the Saccharomyces cerevisiae protein-O-mannosyltransferase Pmt1p. Expression of the H. polymorpha PMT1 gene (HpPMT1) abolished temperature sensitivity of the S. cerevisiae pmt1 pmt2 double mutant. As in S. cerevisiae, inactivation of the HpPMT1 gene affected electrophoretic mobility of the O-glycosylated protein, extracellular chitinase. In contrast to S. cerevisiae, disruption of HpPMT1 alone caused temperature sensitivity. Inactivation of the HpPMT1 gene decreased intracellular aggregation of uPA, suggesting that enhanced secretion of uPA was due to improvement of its folding in the endoplasmic reticulum. Unlike most of the endoplasmic reticulum membrane proteins, HpPmt1p possesses the C-terminal KDEL retention signal.  相似文献

Targeted DNA integration into known locations in the genome has potential advantages over the random insertional events typically achieved using conventional means of genetic modification. We studied the presence and extent of DNA rearrangements at the junction of plant and transgenic DNA in five lines of Arabidopsis thaliana suspension cells carrying a site-specific integration of target genes. Two types of templates were used to obtain knock-ins, differing in the presence or absence of flanking DNA homologous to the target site in the genome. For the targeted insertion, we selected the region of the histone H3.3 gene with a very high constitutive level of expression. Our studies showed that all five obtained knock-in cell lines have rearrangements at the borders of the integrated sequence. Significant rearrangements, about 100 or more bp from the side of the right flank, were found in all five plant lines. Reorganizations from the left flank at more than 17 bp were found in three out of five lines. The fact that rearrangements were detected for both variants of the knock-in template (with and without flanks) indicates that the presence of flanks does not affect the occurrence of mutations.  相似文献
