Desiccation of plants during drought can be detrimental to agricultural production. The phytohormone abscisic acid (ABA) reduces water loss by triggering stomatal pore closure in leaves, a process requiring ion-channel modulation by cytoplasmic proteins. Deletion of the Arabidopsis farnesyltransferase gene ERA1 or application of farnesyltransferase inhibitors resulted in ABA hypersensitivity of guard cell anion-channel activation and of stomatal closing. ERA1 was expressed in guard cells. Double-mutant analyses of era1 with the ABA-insensitive mutants abi1 and abi2 showed that era1 suppresses the ABA-insensitive phenotypes. Moreover, era1 plants exhibited a reduction in transpirational water loss during drought treatment.  相似文献

Adult American women as a group tend to gain weight with age, and many women report that their weight gain started around the time of their menopause. Moreover, as women age, there are changes in body composition that include losses in bone mineral and body cell mass, and increases in total body fat, visceral fat, and extracellular fluid. It appears as if these body composition changes begin or accelerate during the menopausal years. The importance of weight gain and changes in body composition are their associations with an increased risk of developing some malignancies, cardiovascular disease, osteoporosis, and several other clinical conditions. This overview describes selected studies of menopause and aging-associated weight gain, changes in body composition, and alterations of energy expenditure in women. Gaps in the present understanding of these changes are highlighted, and an emphasis is placed on new research methodologies for investigating body composition and energy expenditure in vivo. A concluding section of the report summarizes areas in need of future investigation.  相似文献

Genetic studies have suggested homogeneity between the Baltic-type and Mediterranean-type progressive myoclonic epilepsy. Magnetoencephalography was applied to elucidate the mechanism underlying the giant evoked responses in cortical reflex myoclonus. A new concept of negative myoclonus mediated by cortical reflex mechanism was proposed. Cortical myoclonus was demonstrated in various neurodegenerative or metabolic disorders, such as presenile or senile dementia, olivopontocerebellar atrophy, and myoclonus epilepsy associated with ragged-red fibres. Myoclonus in corticobasal degeneration is especially noteworthy because it has clinical and electrophysiological features of cortical reflex myoclonus, but its latency is shorter compared with the conventional cortical reflex myoclonus. Clinical features of 'palatal myoclonus' were reported by the name of 'palatal tremor'.  相似文献

The cyclin dependent kinase 2 (Cdk2) is required for initiation and progression of DNA replication. Activation of Cdk2 involves binding to cyclin E or cyclin A and dephosphorylation of Tyr15. The present studies demonstrate that treatment of U-937 cells with 1-beta-D-arabinofuranosylcytosine (ara-C) is associated with tyrosine phosphorylation of Cdk2 and inhibition of Cdk2 activity. The results also demonstrate that Cdk2 directly associates with the Src-like tyrosine kinase Lyn as a consequence of ara-C-treatment. Confocal microscopy studies show that Lyn is detectable in the nucleus and that it colocalises with Cdk2. Subcellular fractionation and coimmunoprecipitation studies further demonstrate nuclear binding of Lyn and Cdk2. We also show that Lyn phosphorylates Tyr15 of Cdk2 and that incubation of Lyn with Cdk2 results in inhibition of Cdk2 activity. These findings suggest that the association of Lyn and Cdk2 in ara-C-treated cells may contribute to regulation of Cdk2-dependent cell cycle checkpoints.  相似文献

Antimicrobial activity of some complicated space phenols (screened) was studied. The compounds had different activities against grampositive bacteria and were inactive against gramnegative microbes. Di-tertiary butyl derivatives of pyrocatechol and resorcin showed the highest activities. The MICs of such derivatives for the collection strains of Bacillus megaterium, Bacillus subtilis and Staphylococcus aureus were 8 to 30 micrograms/ml and exceeded 6-25 times those of the nonsubstituted analogs. The derivatives of pyrocatechol and resorcin impaired the membrane permeability in susceptible intact cells of B.megaterium and S.aureus 209P and had no effect on the membrane permeability of the Escherichia coli resistant cells. In concentrations up to 200 micrograms/ml the nonsubstituted analogs of pyrocatechol and resorcin did not impair the membrane permeability in the intact cells of the above bacteria. Di-tertiary butyl derivatives of pyrocatechol and resorcin had lytic activity with respect to cytoplasmic membranes (protoplasts) of B.megaterium and had no lytic action on E.coli spheroplasts. The antimicrobial spectrum correlated with the membranotropic properties of the compounds. It was suggested that the target of the antimicrobial action of the screened phenols was the bacterial cell cytoplasmic membrane.  相似文献

The c-Abl protein tyrosine kinase is activated by ionizing radiation (IR) and certain other DNA-damaging agents. The present studies demonstrate that c-Abl associates constitutively with protein kinase C delta (PKCdelta). The results show that the SH3 domain of c-Abl interacts directly with PKCdelta. c-Abl phosphorylates and activates PKCdelta in vitro. We also show that IR treatment of cells is associated with c-Abl-dependent phosphorylation of PKCdelta and translocation of PKCdelta to the nucleus. These findings support a functional interaction between c-Abl and PKCdelta in the cellular response to genotoxic stress.  相似文献

A 10 to 17 years endoscope follow-up was performed to 138 cases of chronic gastritis. The result showed that 118 cases still proved to be chronic gastritis, and the increase of chronic atrophic gastritis (CAG) in numbers. 15 cases developed into peptic ulcer. 5 cases to carcinoma (4 cases were early carcinoma). The time of cancerization differed from 2 to 12 years. The rate of cancerization of CAG reached 7.46%. The rate of cancerization of intestinal metaplasia (IM) were 8.20%. 13 cases of IM were mucus histo-chemical stained, and five of them contained sulfuric acid mucus, one of the 5 cases cancerized. 3 of 14 cases with atypical hyperplasia (ATP) turned into stomach cancer. We believe that chronic gastritis, especially CAG with ATP and IM, or with sulfuric acid mucus of IM had a high possibility of cancerization with the increase of age, and should be followed up for a long time.  相似文献
