A method for the quantification of selected toxaphene congeners as well as "total toxaphene" was developed based on electron ionization (EI) tandem (MS/MS) ion trap mass spectrometry (MS) and a combination of fragment ion dissociations. Congeners were separated by high-resolution gas chromatography. Compared to conventional EI low-resolution MS, a 5-20-fold gain in sensitivity could be obtained for octa- or nonachlorinated compounds such as toxaphene #26 and #62 (according to Parlar nomenclature), allowing for their detection in the low picogram range in biota. In addition, response factors for important congeners such as #26, #32, #40/41, #42, #44, #50, and #62 deviated not more than a factor of 2, which is much less as compared to negative ion chemical ionization. This reduces the risk for systematic errors when determining total toxaphene on the basis of a limited number of reference compounds or the technical mixture. Furthermore, chlordanes and polychlorinated biphenyls did not interfere when applying the proposed MS/MS technique. The applicability of the method was tested by determining both total toxaphene and levels of selected congeners in six Arctic wildlife samples collected from Nunavut, Canada, as well as by repetitive analyses of the SRM 1588 certified reference material. 相似文献

采用水蒸气蒸馏法提取一年蓬花的挥发油,以DPPH自由基、ABTS自由基和亚硝酸钠的清除作用为指标评价一年蓬花挥发油的体外抗氧化活性,并与维生素C(VC)和2,6-二叔丁基-4-甲基苯酚(BHT)。研究结果表明:20%挥发油对DPPH自由基、ABTS自由基和亚硝酸钠有明显的清除作用,其IC50值分别为99.33、22.68、85.79μL。20%一年蓬花挥发油对DPPH自由基和亚硝酸钠的清除能力分别优于0.1mg/mL的维生素C和1mg/mL BHT;对ABTS自由基的清除能力略低于1mg/mL的维生素C。一年蓬花挥发油具有良好的体外抗氧化活性,样品量与清除率存在量效关系。 相似文献

植物乳杆菌KLDS1.0391能够合成细菌素,也是益生菌,在食品中既可作为益生菌使用,也可作为辅助发酵剂用于生物防控,该研究主要考察了KLDS1.0391菌株在酸奶体系中细菌素的产生特点。研究结果表明,在发酵的6 h期间,抑菌活性随发酵时间延长而增强,发酵结束时,添加植物乳杆菌KLDS1.0391酸奶组的抑菌活性显著高于仅使用酸奶发酵剂的对照组(P<0.01)。与对照组相比,加入辅助发酵剂的实验组的感官品质未发生明显的变化。植物乳杆菌KLDS1.0391具备开发益生酸奶的潜力。 相似文献

High-amylose cereal starch has a great benefit on human health. Acid modification is very helpful for application of high-amylose starch in food and non-food industries. In this study, the ordered structure of acid-modified high-amylose rice starch was investigated by GPC, HPAEC, 13C CP/MAS NMR and XRD. Acid preferentially degraded the amylose, then A chain and short B chain of amylopectin. Relative double helix content and crystallinity both initially increased sharply and then progressively with acid hydrolysis. The relative crystallinity of starches obtained from 13C CP/MAS NMR was higher than that from XRD. The onset gelatinisation temperature decreased, while the peak and conclusion temperatures increased with increasing hydrolysis time. The endothermic value initially increased and then decreased with acid hydrolysis. The swelling power decreased while solubility increased after acid hydrolysis. These results add to our understanding of the effect of acid hydrolysis on the high-amylose rice starch. 相似文献

This article describes a comparative study of enzyme and ultrasound techniques for the simultaneous extraction of vitamin C and phenolic compounds from acerola fruit. Ultrasound‐assisted extraction (UAE) took only 6 min to achieve the highest level of vitamin C and phenolic compounds as well as antioxidant activity of acerola juice, while enzyme‐assisted extraction (EAE) took up to 120 min to obtain the maximal values. On the basis of kinetic model of second‐order extraction, the extraction rate constant of vitamin C and phenolics in UAE increased approximately 3.1 and 2.7 times, respectively, in comparison with that in EAE. In addition, the maximal level of vitamin C, phenolics and the antioxidant activity evaluated by 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH), and 2,2′‐azinobis‐(3‐ethylbenzothiazoline‐6‐sulphonic acid) (ABTS) methods in UAE was 4.6%, 3.5%, 4.6% and 3.3%, respectively, higher than those in EAE. Klebsiella pneumoniae is an opportunistic and environmental mastitis-causing pathogen, with potential for contagious transmission. Repetitive element sequence-based PCR was used to determine genetic diversity and explore potential transmission and reservoirs for mastitis caused by K. pneumoniae on 2 large Chinese dairy farms. A total of 1,354 samples was collected from the 2 dairy farms, including milk samples from cows with subclinical and clinical mastitis, bedding, feces, feed, teat skin, and milking liners. Environmental samples were collected from all barns and milking parlors and extramammary samples from randomly selected dairy cows on both farms. In total, 272 and 93 K. pneumoniae isolates were obtained from Farms A and B, respectively (with ~8K and 2K lactating cows, respectively). Isolation rates from clinical mastitis (CM), subclinical mastitis (SCM), and environmental or extramammary samples were 34, 23 and 37%, respectively for Farm A and 42, 3, and 34% for Farm B. The K. pneumoniae isolated from CM milk and extramammary or environmental sources had high genetic diversity (index of diversity >90%) on the 2 farms and from SCM on Farm A. However, on Farm B, 9 SCM isolates were classified as 2 genotypes, resulting in a relatively low index of diversity (Simpson's index of diversity = 0.39; 95% CI = 0.08–0.70). Genotypes of K. pneumoniae causing mastitis were commonly detected in feces, bedding, and milking liners (Farm A), or from teat skin, sawdust bedding, and feed (Farm B). Based on its high level of genetic diversity, we inferred K. pneumoniae was an opportunistic and environmental pathogen causing outbreaks of CM on these 2 large Chinese dairy farms. Nevertheless, that only a few genotypes caused SCM implied some strains had increased udder adaptability and a contagious nature or a common extramammary source. Finally, control of intramammary infections caused by K. pneumoniae on large Chinese dairy farms must consider farm-level predictors, as the 2 outbreaks had distinct potential environmental sources of infection. 相似文献

This study aimed to predict the optimal carbon source for higher production of exopolysaccharides (EPS) by Lactobacillus paracasei TD 062, and to evaluate the effect of this carbon source on the production and monosaccharide composition of EPS. We evaluated the EPS production capacity of 20 strains of L. paracasei under the same conditions. We further investigated L. paracasei TD 062, which showed the highest EPS-producing activity (0.609 g/L), by examining the associated biosynthesis pathways for EPS. Genomics revealed that fructose, mannose, trehalose, glucose, galactose, and lactose were carbon sources that L. paracasei TD 062 could use to produce EPS. We identified an EPS synthesis gene cluster that could participate in transport, export, and sugar chain synthesis, and generate 6 sugar nucleotides. Experimental results showed that the sugar content of the EPS produced using fermentation with the optimized carbon source (fructose, mannose, trehalose, glucose, galactose, and lactose) increased by 115%. Furthermore, use of the optimized carbon source changed the monosaccharide content of the associated EPS. The results of enzyme activity measurements showed significant increases in the activity of 2 key enzymes involved in the glycoside synthesis pathway. Our study revealed that optimizing the carbon source provided for fermentation not only increased the production of EPS, but also affected the composition of the monosaccharides by increasing enzyme activity in the underlying synthesis pathways, suggesting an important role for carbon source in the production of EPS by L. paracasei TD 062. 相似文献
