SSAO酶抑制剂对脂肪细胞内GLUT4转运作用的研究

SSAO酶在脂肪细胞中的含量较多,位于细胞膜上的SSAO酶的活性和其蛋白质的免疫反应性都是最大的.对于体内葡萄糖的运输和葡萄糖转运蛋白的转运,此酶起到了一定的促进作用,当它与底物在膜上反应后会引起信号传导刺激细胞内葡萄糖转运蛋白(GLUT4)从内部囊泡转移到细胞表面程,从而促进葡萄糖的运输和细胞对葡萄糖的吸收.如果有SSAO酶抑制剂存在,如氨基脲,二溴乙胺等,这种对GLUT4转运的促进作用就会被削弱.本研究的目的在于建立一种新的定量方法-竞争ELISA法来测试脂肪细胞膜上GLUT4的含量,从而确定SSAO酶抑制剂对GLUT4转运的作用.实验结果说明当SSAO酶抑制剂和脂肪细胞预温育一段时间后,质膜上GLUT4的含量发生下降,钒盐使得这种下降的程度加大,证明GLUT4由细胞内囊泡向细胞质膜转运的易位过程受到了抑制.此种竞争性ELISA的测试方法优点在于只需要少量的细胞膜样品就可以确定GLUT4的含量,灵敏度较高.     

DA、Salsolinol及N-methyl-salsolinol的HPLC-ECD检测方法研究

采用高效液相色谱结合库仑阵列多电极检测器体系（HPLC-ECD）建立了一套操作简便、高效、快速测定多巴胺（DA）、6,7-二羟基-1-甲基-1,2,3,4-四氢异喹啉（Salsolinol）和6,7-二羟基-1,2-二甲基-1,2,3,4 -四氢异喹啉（N-methyl-salsolinol）的方法,并研究该检测方法的灵敏性和稳定性,检测物质浓度和响应因子之间的线性关系。DA、Salsolinol及N-methyl-salsolinol的相关系数分别为1、1和0.9997;DA的检测限低于0.1ng/mL,Sal和NMSal的检测限低于0.5ng/mL,各种物质的方法回收率为98.5%-104.7%。     

Anti-Hyperglycemic Activity of Shan Tang Ping an Ethanol Extract from Yew Tree in Diabetic Rats

The extracts of yew tree have showed unique anti-tumor and anti-leakematic activities. Taxol extracted from it is effective against a variety of cancers, such as ovarian cancer and breast cancer. However the hypoglycemic efficacy of the extract has not been reported yet, so the effects of oral administration of yew tree extracts on streptozotocin-induced diabetic rats were examined. Administration of shah tang ping （STP）, a fraction from the column separation of extract （450 mg/kg body weight）, for 14 days reduced the serum glucose concentrations of fasted diabetic rats by 52.4%. In addition, it also decreased the concentrations of triglyceride （TG）, total cholesterol （TC）, and low density lipoprotein cholesterol （LDL-C）. An oral glucose tolerance test （OGTT） was performed, a significant improvement in glucose tolerance was observed in rats treated with STP. These results suggest that STP has hypoglycemic properties and hypolipidemic action on streptozotocin-induced diabetic rats.     

发酵液中木糖酸的测定

本研究建立了发酵液中木糖酸的分析方法.分别采用反相高效液相色谱法和比色法对发酵液中的木糖酸进行分析.结果通过考察回收率和精密度等指标证明了高效液相色谱法对发酵液中木糖酸的分析优于比色法.高效液相色谱法对复杂的生物体系中的木糖酸含量测定具有快速,简便、灵敏、重现性好的特点,为木糖酸的生产、应用等研究打下了良好的基础.     

Quantification of CP4-EPSPS in genetically modified Nicotiana tabacum leaves by LC-MS/MS with 18O-labeling

The CP4-EPSPS gene is widely used in herbicide-tolerant plants/crops all over the world. In this study, a method was developed by coupling liquid chromatography with high sensitivity to tandem mass spectrometry to quantify the amount of CP4-EPSPS expression in Nicotiana tabacum leaves. The quantification of protein was converted to measure the unique peptide of CP4-EPSPS protein. One peptide unique to CP4-EPSPS was synthesized and labeled with H₂ ¹⁸O to get ¹⁸O stable isotope labeled peptide. The peptide served as the internal standard. The validated method had good specificity and linearity. The intra-and inter-day precisions and accuracy for all samples were satisfactory. The results demonstrated that the novel method was sensitive and selective to quantify CP4-EPSPS in the crude extract without time-consuming pre-separation or the purification procedures.     

Synthesis of Amino Acid Derived β-Cyclodextrins Used in Chiral Separation by Capillary Electrophoresis

Six new kinds of amino acid derived β-cyclodextrins were synthesized to improve their water solubility and chiral separation properties. They are heptakis {2,6-di-O-[3-L-(1-isopropyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Val-β-CD), heptakis {2,6-di-O-[3-L-(1-benzyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Phe-β-CD), heptakis{2,6-di-O-[3-(D, L-1-benzyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. D,L-Phe-β-CD), heptakis {2,6-di-O-[3-(L-1-hydrox-ymethyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Ser-β-CD), heptakis{2,6-di-O-[3-(L-1-carboxylmethyl carboxyl methyl amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Asp-β-CD), heptakis{2,6-di-O-[3-(L-2-carboxyl tetramethylene amino)-2-hydroxy propyl]}-β-cyclodextrin (i.e. L-Pro-β-CD).Their chemical structures were certified using FTIR and ^1H NMR. Except for L-Phe-β-CD and D,L-Phe-β-CD,that are in soluble in water, the other amino acid derived β-CDs all have good water solubility. D,L-tyrosine and promethazine were baselinely separated by L-Val-β-CD in capillary electrophoresis.     

N-炔丙基-R-2-庚胺的合成工艺研究

以2-庚酮和甲酸铵为原料合成了2-庚胺,收率76.8%,然后经D-二苯甲酰酒石酸拆分后得到R-2-庚胺,拆分率41.4%.最后R-2-庚胺和乙炔、甲醛在碘化亚铜催化下合成N-炔丙基-R-2-庚胺,收率82.29b.并对反应可能的机理进行了探讨.     

有机-无机杂化介孔材料包埋南极假丝酵母脂肪酶的研究

以四甲氧基硅烷(TMOS)和一定比例的甲基三甲氧基硅烷(MTMS)、二甲基二甲氧基硅烷(DMDMS)、三甲基甲氧基硅烷(TMMS)为硅源,采用酸催化的溶胶-凝胶法包埋南极假丝酵母脂肪酶。比较了有机-无机杂化介孔材料和纯硅基介孔材料固定化南极假丝酵母脂肪酶的酶活力和保留活力,同时还比较了有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶和游离南极假丝酵母脂肪酶的pH、温度以及储存稳定性。结果表明,有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶具有比纯硅基介孔材料固定化南极假丝酵母脂肪酶更高的酶活力和保留活力。其中三取代的有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶的酶活力最高,其保留活力可达到84.9%。此外与游离的南极假丝酵母脂肪酶相比,有机-无机杂化介孔材料固定化南极假丝酵母脂肪酶具有更好的pH、温度和储存稳定性,以及更宽的pH值和温度的酶活力范围。

空间特殊环境给生命科学研究带来了前所未有的机遇,空间生命科学技术已经成为世界科技强国的科技竞争制高点。科学载荷(仪器)是开展空间生命科学实验的基础工具,是航天生物医学研究的必要基础,决定了空间生命科学研究和医学保障能力。本文按照不同的研究对象,对国内外典型的生命科学载荷进行综述,同时,结合研究需求状况对部分关键技术的发展趋势进行了分析,指出了器官芯片技术在空间生命科学载荷技术发展中的关键作用,以及原位生命信息探测技术的持续需求。最后,对当前生命科学载荷技术的发展趋势进行了总结。
