Perfluorooctane sulfonate (PFOS) and other perfluoroalkyl compounds (PFCs) were measured in liver samples from 29 marine tucuxi dolphins from Rio de Janeiro state (RJ), Brazil. PFC measurement combined liquid chromatography and mass spectrometry, using a CapLC system connected to a Quadrupole-LIT mass spectrometer. PFOS was the only PFC detected and it was so in all samples. PFOS concentrations (ng x g(-1) dw) of dolphins (n=23) from the highly contaminated Guanabara Bay (in RJ) varied between 43 and 2431 as well as between 76 and 427 from areas of RJ other than the quoted bay (n=6). Concentrations of three fetuses and one neonate varied between 664 and 1590. Fetus/mother ratios were calculated in two situations (2.75 and 2.62). It seems that mother-to-calf transference plays important role for relationships between PFOS and age. When a one-year-old male calf presenting 2431 ng x g(-1) dw was excluded from the test, significant correlations were observed between PFOS concentrations and both age and total length. Despite the placental transference, PFOS concentrations were not significantly lower in females than in males. PFOS levels in marine tucuxi dolphins from Guanabara Bay are among the highest detected to date in cetaceans, and this may represent a threat to the small population concerned.  相似文献

ABSTRACT:  Solid-state electron paramagnetic resonance (EPR) spectroscopy of commercial samples of isolated soy proteins (ISP) revealed a symmetrical free-radical signal typical of carbon-centered radicals ( g = 2.005) ranging from 2.96 × 1014 to 6.42 × 1014 spins/g. The level of free radicals in ISP was 14 times greater than similar radicals in sodium caseinate, 29 times greater than egg albumin, and about 100 times greater levels than casein. Nine soy protein powdered drink mixes contained similar types of free radicals up to 4.10 × 1015 spins/g of drink mix, or up to 6.4 times greater than the highest free-radical content found in commercial ISP. ISP samples prepared in the laboratory contained trapped radicals similar to the levels in commercial ISP samples. When ISP was hydrated in 2.3 mM sodium erythorbate or 8.3 mM L-cysteine, frozen and dried, the level of trapped free radicals increased by about 17- and 19-fold, respectively. The ESR spectrum of defatted soybean flakes contained overlapping signals from the primary free-radical peak ( g = 2.005) and a sextet pattern typical of manganese-II. The manganese signal was reduced in the laboratory ISP and very weak in the commercial ISP.  相似文献

A mechanical lung simulator can be used to simulate specific lung pathologies, to test lung-function equipment, and in instruction. A new approach to mechanical simulation of lung behavior is introduced that uses a computer-controlled active mechatronic system. The main advantage of this approach is that the static and dynamic properties of the simulator can easily be adjusted via the control software. A nonlinear single-compartment mathematical model of the artificially ventilated respiratory system has been derived and incorporated into the simulator control system. This model can capture both the static and dynamic compliance of the respiratory system as well as nonlinear flow-resistance properties. Parameters in this model can be estimated by using data from artificially ventilated patients. It is shown that the simulation model fits patient data well. This mathematical model of the respiratory system was then matched to a model of the available physical equipment (the simulator, actuators, and the interface electronics) in order to obtain the desired lung behavior. A significant time delay in the piston motion control loop has been identified, which can potentially cause oscillations or even instability for high compliance values. Therefore, a feedback controller based on the Smith-predictor scheme was developed to control the piston motion. The control system, implemented on a personal computer, also includes a user-friendly interface to allow easy parameter setting.  相似文献

In this work, poly(N-isopropylacrylamide) (PNIPAAm) was incorporated into previously oxidized PS and PET surfaces by grafting using two photo-initiation pathways. The incorporation of PNIPAAm was observed by drop water contact angle measurements, dyeing with Methylene Blue and AFM images analysis of the virgin and modified polymers. It was verified that the grafting process depends on the chemical surface environment. The grafted surfaces are hydrophilic below 32 °C and hydrophobic above this temperature. The transition is due to the incorporated PNIPAAm. This characteristic gives to the grafted materials potential to be applied as biomaterials.  相似文献

During the stationary phase of Campylobacter jejuni NCTC 11351 viable numbers fluctuate in a characteristic fashion. After reaching the maximum cell count (ca. 2×109 CFU/ml) in early stationary phase (denoted phase 1), viable numbers subsequently decrease to about 106 CFU/ml after 48 h and then increase again to about 108 CFU/ml (denoted phase 2) before decreasing once more to a value intermediate between the previous maximum and minimum values. To investigate whether the increase in viable numbers following the initial decline was due to the emergence of a new strain with a growth advantage in stationary phase analogous to the ‘GASP’ phenotype described in Escherichia coli [Science 259 (1993) 1757], we conducted mixed culture experiments with cells from the original culture and antibiotic-resistant marked organisms isolated from the re-growth phase. In many experiments of this type, strains isolated from phase 2 failed to out-compete the original strain and we have thus been unable to demonstrate a convincing GASP phenotype. However, strains isolated from phase 2 showed a much lower rate of viability loss in early stationary phase and a small increase in resistance to aeration, peroxide challenge and heat, indicating that the emergent strain was different from the parent. These results support the view that dynamic population changes occur during the stationary phase of C. jejuni that may play a role in the survival of this organism.  相似文献

Flow-mediated dilation (FMD) offers a mechanism to characterize endothelial function and, therefore, may play a role in the diagnosis of cardiovascular diseases. Computerized analysis techniques are very desirable to give accuracy and objectivity to the measurements. Virtually all methods proposed up to now to measure FMD rely on accurate edge detection of the arterial wall, and they are not always robust in the presence of poor image quality or image artifacts. A novel method for automatic dilation assessment based on a global image analysis strategy is presented. We model interframe arterial dilation as a superposition of a rigid motion and a scaling factor perpendicular to the artery. Rigid motion can be interpreted as a global compensation for patient and probe movements, an aspect that has not been sufficiently studied before. The scaling factor explains arterial dilation. The ultrasound sequence is analyzed in two phases using image registration to recover both transformation models. Temporal continuity in the registration parameters along the sequence is enforced with a Kalman filter since the dilation process is known to be a gradual physiological phenomenon. Comparing automated and gold standard measurements (average of manual measurements) we found a negligible bias (0.05%FMD) and a small standard deviation (SD) of the differences (1.05%FMD). These values are comparable with those obtained from manual measurements (bias = 0.23%FMD, SD(intra-obs) = 1.13%FMD, SD(inter-obs) 1.20%FMD). The proposed method offers also better reproducibility (CV = 0.40%) than the manual measurements (CV = 1.04%).  相似文献

The goal of this study was to evaluate methods of multidimensional wavelet denoising on restoring the fidelity of biological signals hidden within dynamic positron emission tomography (PET) images. A reduction of noise within pixels, between adjacent regions, and time-serial frames was achieved via redundant multiscale representations. In analyzing dynamic PET data of healthy volunteers, a multiscale method improved the estimate-to-error ratio of flows fivefold without loss of detail. This technique also maintained accuracy of flow estimates in comparison with the "gold standard," using dynamic PET with O15-water. In addition, in studies of coronary disease patients, flow patterns were preserved and infarcted regions were well differentiated from normal regions. The results show that a wavelet-based noise-suppression method produced reliable approximations of salient underlying signals and led to an accurate quantification of myocardial perfusion. The described protocol can be generalized to other temporal biomedical imaging modalities including functional magnetic resonance imaging and ultrasound.  相似文献
