During the stationary phase of Campylobacter jejuni NCTC 11351 viable numbers fluctuate in a characteristic fashion. After reaching the maximum cell count (ca. 2×109 CFU/ml) in early stationary phase (denoted phase 1), viable numbers subsequently decrease to about 106 CFU/ml after 48 h and then increase again to about 108 CFU/ml (denoted phase 2) before decreasing once more to a value intermediate between the previous maximum and minimum values. To investigate whether the increase in viable numbers following the initial decline was due to the emergence of a new strain with a growth advantage in stationary phase analogous to the ‘GASP’ phenotype described in Escherichia coli [Science 259 (1993) 1757], we conducted mixed culture experiments with cells from the original culture and antibiotic-resistant marked organisms isolated from the re-growth phase. In many experiments of this type, strains isolated from phase 2 failed to out-compete the original strain and we have thus been unable to demonstrate a convincing GASP phenotype. However, strains isolated from phase 2 showed a much lower rate of viability loss in early stationary phase and a small increase in resistance to aeration, peroxide challenge and heat, indicating that the emergent strain was different from the parent. These results support the view that dynamic population changes occur during the stationary phase of C. jejuni that may play a role in the survival of this organism. 相似文献

INTRODUCTION: The use of MRI with iron-based magnetic nanoparticles for imaging cells is a rapidly growing field of research. We have recently reported that single iron-labeled cells could be detected, as signal voids, in vivo in mouse brains using a balanced steady-state free precession imaging sequence (b-SSFP) and a customized microimaging system at 1.5 T. METHODS: In the current study we assess the benefits, and challenges, of using a higher magnetic field strength for imaging iron-labeled cells with b-SSFP, using ex vivo mouse brain specimens imaged with near identical systems at 1.5 and 3.0 T. RESULTS: The substantial banding artifact that appears in 3 T b-SSFP images was readily minimized with RF phase cycling, allowing for banding-free b-SSFP images to be compared between the two field strengths. This study revealed that with an optimal 3 T b-SSFP imaging protocol, more than twice as many signal voids were detected as with 1.5 T. CONCLUSION: There are several factors that contributed to this important result. First, a greater-than-linear SNR gain was achieved in mouse brain images at 3 T. Second, a reduction in the bandwidth, and the associated increase in repetition time and SNR, produced a dramatic increase in the contrast generated by iron-labeled cells. 相似文献

The objective of the study was to assess the changes in the mechanical load on the low-back when shifting from a self-chosen to a recommended patient-handling technique. Nine female health care workers without formal education in patient-handling carried out 8 different tasks involving moving, turning and lifting situations. By means of a dynamic 3D biomechanical model of the lower part of the body, peak torque, compression and shear forces at the L4/L5 joint were compared using the two different patient-handling techniques. In 5 of the 8 tasks, a significant reduction was observed in spinal loading. Application of the recommended technique decreased the compression value significantly for all tasks with a mean value above 3000 N. For the two tasks with the highest compression values when using the self-chosen technique (4223, 4446 N), the loading was reduced with 36% and 25%, respectively. If the principles behind the recommended technique are implemented and maintained, a decrease in the risk of low-back disorders during patient-handling should thus be expected. 相似文献

Consider an algorithm whose time to convergence is unknown (because of some random element in the algorithm, such as a random initial weight choice for neural network training). Consider the following strategy. Run the algorithm for a specific time T. If it has not converged by time T, cut the run short and rerun it from the start (repeat the same strategy for every run). This so-called restart mechanism has been proposed by Fahlman (1988) in the context of backpropagation training. It is advantageous in problems that are prone to local minima or when there is a large variability in convergence time from run to run, and may lead to a speed-up in such cases. In this article, we analyze theoretically the restart mechanism, and obtain conditions on the probability density of the convergence time for which restart will improve the expected convergence time. We also derive the optimal restart time. We apply the derived formulas to several cases, including steepest-descent algorithms. 相似文献

Density functional calculations and minimization techniques have been employed to characterize the structural and electronic properties of [5,6]-heterofullerene-C58Si-C2v. Since it has odd number of atoms and a near planar tetracoordinate Si atom on the skeleton of the cage, it has odd number of atoms assembling a cage and is a novel molecule. Vibrational frequencies of the molecule have been calculated at the B3LYP/6-31G* level of theory. The absence of imaginary vibrational frequency confirms that the molecule corresponds to a true minimum on the potential energy hypersurface. Sixteen 13C nuclear magnetic resonance (NMR) spectral signals of C58Si are characterized, and its heat of formation was estimated in this work. 相似文献
