A method for detecting cadmium uptake in leaves of Saponaria officinalis doped with a solution of cadmium acetate is described. The technique based on the exposure of dried leaves to X-rays of a wavelength close to that of the metal K-edge could be useful for phytoremediation studies as it could reveal the bioaccumulation in plants due to the treatment either in vivo or in vitro with heavy metals. X-ray microradiography measurements are in agreement with those from peroxidase enzyme assay utilized to follow the oxidative damage induced by heavy metals. At present, as we will see in this report, microradiography has still poorer sensitivity in comparison with enzyme assay, but it has the advantage of being faster, not destructive, and usable even at very high doping levels, where the enzyme assay technique results are fully saturated. Further analysis of the optical density values could lead to a quantitative measurement of the heavy metal in the sample. Thus, the technology developed in this article could be useful for tracing the intake in phytoremediation studies. 相似文献

Breast cancer is the main cause of morbidity and mortality in women worldwide. However, the molecular pathogenesis of breast cancer remains poorly defined due to its heterogeneity. Several studies have reported that G Protein-Coupled Estrogen Receptor 1 (GPER1) plays a crucial role in breast cancer progression, by binding to estrogens or synthetic agonists, like G-1, thus modulating genes involved in diverse biological events, such as cell proliferation, migration, apoptosis, and metastasis. In addition, it has been established that the dysregulation of short sequences of non-coding RNA, named microRNAs (miRNAs), is involved in various pathophysiological conditions, including breast cancer. Recent evidence has indicated that estrogens may regulate miRNA expression and therefore modulate the levels of their target genes, not only through the classical estrogen receptors (ERs), but also activating GPER1 signalling, hence suggesting an alternative molecular pathway involved in breast tumor progression. Here, the current knowledge about GPER1 and miRNA action in breast cancer is recapitulated, reporting recent evidence on the liaison of these two players in triggering breast tumorogenic effects. Elucidating the role of GPER1 and miRNAs in breast cancer might provide new tools for innovative approaches in anti-cancer therapy. 相似文献

For the numerical solution of initial value problems a general procedure to determine global integration methods is derived and studied. They are collocation methods which can be easily implemented and provide a high order accuracy. They further provide globally continuous differentiable solutions. Computation of the integrals which appear in the coefficients are generated by a recurrence formula and no integrals are involved in the calculation. Corneal endothelial dystrophy is a relevant cause of vision loss and corneal transplantation worldwide. In the present study, we analyzed the effect of mesenchymal stem cell (MSC)-derived extracellular vesicles (MSC-EVs) in an in vitro model of corneal dystrophy, characterized by endoplasmic reticulum stress. The effects of MSC-EVs were compared with those of serum-derived EVs, reported to display a pro-angiogenic activity. MSC-EVs were able to induce a significant down-regulation of the large majority of endoplasmic reticulum stress-related genes in human corneal endothelial cells after exposure to serum deprivation and tunicamycin. In parallel, they upregulated the Akt pathway and limited caspase-3 activation and apoptosis. At variance, the effect of the serum EVs was mainly limited to Akt phosphorylation, with minimal or absent effects on endoplasmic reticulum stress modulation and apoptosis prevention. The effects of MSC-EVs were correlated to the transfer of numerous endoplasmic reticulum (ER)-stress targeting miRNAs to corneal endothelial cells. These data suggest a potential therapeutic effect of MSC-EVs for corneal endothelial endoplasmic reticulum stress, a major player in corneal endothelial dystrophy. 相似文献

A novel molecular beacon (a nanomachine) is constructed that can be actuated by a radio frequency (RF) field. The nanomachine consists of the following elements arranged in molecular beacon configuration: a gold nanoparticle that acts both as quencher for fluorescence and a localized heat source; one reporter fluorochrome, and; a piece of DNA as a hinge and recognition sequence. When the nanomachines are irradiated with a 3 GHz RF field the fluorescence signal increases due to melting of the stem of the molecular beacon. A control experiment, performed using molecular beacons synthesized by substituting the gold nanoparticle by an organic quencher, shows no increase in fluorescence signal when exposed to the RF field. It may therefore be concluded that the increased fluorescence for the gold nanoparticle‐conjugated nanomachines is not due to bulk heating of the solution, but is caused by the presence of the gold nanoparticles and their interaction with the RF field; however, existing models for heating of gold nanoparticles in a RF field are unable to explain the experimental results. Due to the biocompatibility of the construct and RF treatment, the nanomachines may possibly be used inside living cells. In a separate experiment a substantial increase in the dielectric losses can be detected in a RF waveguide setup coupled to a microfluidic channel when gold nanoparticles are added to a low RF loss liquid. User interactions are indispensable for any online network to thrive, especially for BitTorrent‐like and Web real‐time communication‐based distributed online networks that rely on users' collective contributions instead of the help of central servers. User interactions provide fine‐grained information for many applications, such as security enhancement and cooperation promotion. To date, several schemes for estimating user interaction strength in centralized online networks have been proposed. In contrast, we present design, deployment, and analysis of UISE for user interaction strength estimation in distributed online networks. Among the strong points of UISE is that it captures both direct and indirect user interactions, and that it scales with only partial information dissemination. We apply UISE to devise the first distributed scheme for online time estimation and we implement it into Tribler, a distributed online network for media and social applications like file sharing, streaming, and voting. We demonstrate the accuracy and the scalability of UISE with different information dissemination protocols and user behaviors using simulations, emulations, and a real‐world deployment. The day-night task requires saying "night" to a picture of the sun and "day" to a picture of the moon. In this investigation of why young children fail at this task, systematic variations of the task were administered to 96 children, half 4 years old and half 4 1/2 years old. Training children on the strategy of chunking the 2 rules into 1 ("say the opposite"), thus reducing memory load, did not help their performance. What helped was reducing the inhibitory demand by instructing them to say "dog" and "pig" (not "night" and "day") even though memory of 2 rules and inhibiting saying what the pictures represented were still required. Here the response to be activated and the response to be inhibited were unrelated. When the correct response was semantically related to, and the direct opposite of, the to-be-inhibited response, children performed poorly. Inserting a delay between stimulus and response helped even though that delay was filled with distraction. Young children apparently need several seconds to compute the answer on this task. Are spatial proximity (0.10-12.5 cm), temporal proximity (0-, 2-, and 5-s gaps), and/or perceived connectedness of stimulus and reward key to infants' ability to deduce an abstract nonmatching rule from reward feedback? In this investigation, 3 conditions of the delayed nonmatching to sample task were administered to infants 9, 12, and 15 months old, and 5 more conditions were administered just to 12-month-olds. Results showed that connectedness is key. In its presence, neither close spatial or temporal proximity was needed. In the absence of the perception that stimulus and reward were components of a single thing, even close spatial and temporal proximity were insufficient for infants in the 1st year to grasp the rule-based association between stimuli and rewards. The current study presents a Rasch-derived short form of the Center for Epidemiologic Studies-Depression scale (CES-D) for use as a depression screening tool in the general population. In contrast to short forms developed with reliance on classical measurement techniques, those developed using techniques based on item response theory produce a measure that offers true interval scaling, provide enhanced information about responders with extreme scores, and expand understanding of the underlying latent structure. Cross-validation of the Rasch-derived CES-D short form supported its utility and structural validity across samples. Tests of structural validity using latent variable modeling methodology indicated that a hierarchical, single-factor model of depression had the best fit for the original full form and the Rasch-derived short form of the CES-D. This finding challenges depression researchers and theorists to reconsider the interfactor relationships in the study and assessment of depression.
