This paper extends the use of principal-component analysis in spectral quantification to the estimation of frequency and phase shifts in a single resonant peak across a series of spectra. The estimated parameters can be used to correct the spectra accordingly, resulting in more accurate peak-area estimation. Further, the removal of the variations in phase and frequency cause by instrumental and experimental fluctuations makes it possible to determine more accurately the remaining variations, which bear biological significance. The procedure is demonstrated on simulated data, a 3D chemical-shift-imaging dataset acquired from a cylinder of inorganic phosphate (Pi), and a set of 736 31P NMR in vivo spectra taken from a kinetic study of rate muscle energetics. In all cases, the procedure rapidly and automatically identifies the frequency and phase shifts present in the individual spectra. In the kinetic study, the procedure is used twice, first to adjust the phase and frequency of a reference peak (phosphocreatine) and then to determine the individual frequencies of the Pi peak in each of the spectra which further can be used for estimation of pH changes during the experiment.  相似文献

High energy intake in the dry period has reportedly had adverse effects on mobilization of body reserves, dry matter intake, and productivity of dairy cows. We investigated whether grass silage (GS) fed ad libitum (high energy intake, HEI; 141% of daily metabolizable energy requirements) in an 8-wk dry period affects metabolic adaptation—specifically, peripheral insulin resistance—compared with a total mixed ration consisting of GS, wheat straw, and rapeseed meal (55/40/5%; controlled energy intake, CEI; 108% of metabolizable energy/d) fed ad libitum. Multiparous Ayrshire dairy cows (n = 16) were used in a randomized complete block design until 8 wk after parturition. Commercial concentrates were fed 1 and 2 kg/d during the last 10 to 6 and 5 to 0 d before the expected calving date, respectively. Postpartum, a similar lactation diet with ad libitum access to GS and increasing concentrate allowance (maximum of 16 kg/d) was offered to all. The HEI group gained more body weight and had higher plasma insulin, glucose, and β-hydroxybutyrate concentrations than the CEI group prepartum. Postpartal plasma glucose tended to be higher and milk yield was greater from wk 5 onward for HEI compared with CEI cows. An intravenous glucose tolerance test (IVGTT) was performed at ?13 ± 5 d and 9 ± 1 d relative to calving. The HEI cows had greater insulin response to glucose load and smaller area under the response curve for glucose than CEI cows in prepartal IVGTT. Thus, compensatory insulin secretion adapted to changes in insulin sensitivity of the peripheral tissues, preserving glucose tolerance of HEI cows. Higher insulin levels were needed in HEI cows than in CEI cows to elicit a similar decrement of nonesterified fatty acid concentration in prepartal IVGTT, suggesting reduced inhibition of lipolysis by insulin in HEI cows before parturition. In conclusion, high energy intake of moderately digestible GS with low concentrate feeding in the close-up dry period did not have adverse effects on metabolic adaptation, insulin sensitivity, and body mobilization after parturition. Instead, this feeding regimen was more beneficial to early-lactation performance than GS-based total mixed ration diluted with wheat straw.  相似文献

Five rumen-cannulated Finnish Ayrshire cows were used in two 5 x 5 Latin square experiments designed to study the lactation and metabolic responses to increasing doses of DL-Met or L-Lys infused into the abomasum. The cows were fed grass silage ensiled with a formic acid additive for ad libitum intake. A supplement with barley and oats was given at a rate of 9 kg/d (Experiment 1) or 7 kg/d (Experiment 2). The experimental treatments were 0, 10, 20, 30, or 40 g of Met/d (Experiment 1) and 0, 15, 30, 45, or 60 g of Lys/d (Experiment 2). The infusion of Met did not significantly affect feed intake or daily milk yield, but increased milk fat content, ECM yield, and C4 to C14 and C18 to C20 fatty acid production in milk. The infusion of Met caused an increase in arterial plasma Met concentration and a decline in branched-chain amino acids (AA). Mammary gland uptake of Met was not related to plasma AA concentration. The infusion of Lys did not affect feed intake, milk yield, or milk composition, except for increases in milk urea and NPN contents. The infusion of Lys increased plasma Lys, BCAA, EAA, and the EAA to TAA ratio. Uptake of plasma BCAA and NEAA by the mammary gland decreased, which suggests that Lys was used as a substrate for milk NEAA synthesis. These data demonstrate that Met is important in the milk fat synthesis, and Lys is important in mammary gland AA metabolism. However, neither Met nor Lys is the first-limiting AA in the milk protein yield of cows fed a grass silage and cereal diet.  相似文献

The concurrent use of statistical process control and engineering process control involves monitoring manipulated and controlled variables. One multivariate control chart may handle the statistical monitoring of all variables, but observing the manipulated and controlled variables in separate control charts may improve understanding of how disturbances and the controller performance affect the process. In this article, we illustrate how step and ramp disturbances manifest themselves in a single-input–single-output system by studying their resulting signatures in the controlled and manipulated variables. The system is controlled by variations of the widely used proportional-integral-derivative(PID) control scheme. Implications for applying control charts for these scenarios are discussed.  相似文献

The increasing need for high-quality diesel fuel can be fulfilled by using environmental friendly, high-performance additives. We have synthesized novel fatty acid-derived multifunctional additives coupled with phosphazene moiety having the backbone of alternative nitrogen and phosphorous atoms which provided stability, while the additives possessed affinity for the metal surface to form a surface-complex film leading to the hindered metal contact and providing friction-reducing and antiwear properties. In addition, the applicability of the synthesized additives was investigated by testing the anti-oxidant and anticorrosion properties in polyethylene glycol (PEG200) which was taken as the reference base fluid. A rotating bomb oxidation test was used to evaluate the anti-oxidant characteristics, and a four-ball test for the tribological properties. Among the synthesized additives, phosphazene ester of erucic acid exhibited the best antiwear and antifriction properties; however, phosphazene ester of oleic acid was found to be the best anti-oxidant additive.  相似文献

Engineering process control and high-dimensional, time-dependent data present great methodological challenges when applying statistical process control (SPC) and design of experiments (DoE) in continuous industrial processes. Process simulators with an ability to mimic these challenges are instrumental in research and education. This article focuses on the revised Tennessee Eastman process simulator providing guidelines for its use as a testbed for SPC and DoE methods. We provide flowcharts that can support new users to get started in the Simulink/Matlab framework, and illustrate how to run stochastic simulations for SPC and DoE applications using the Tennessee Eastman process.  相似文献

Late pregnancy is associated with moderate insulin resistance in ruminants. Reduced suppression of lipolysis by insulin facilitates mobilization of nonesterified fatty acids (NEFA) from adipose tissue, resulting in elevated plasma NEFA concentrations. Decrease in dry matter intake (DMI) before parturition leads to accelerated lipomobilization and increases plasma NEFA, which may further impair insulin sensitivity. The aim of the study was to evaluate the effects of elevation of plasma NEFA concentration by abomasal infusions tallow (TAL) or camelina oil (CAM) on whole-body responses to exogenous glucose and insulin. We further assessed whether CAM, rich in C18:3n-3, enhances whole-body insulin sensitivity compared with TAL. Six late-pregnant, second-parity, rumen-cannulated dry Ayrshire dairy cows fed grass silage to meet 95% of metabolizable energy requirements were used in a replicated 3 × 3 Latin square with 5-d periods and 5 recovery days between each period. Treatments consisted of abomasal infusion of 500 mL/d (430 g of lipids/d) of water (control), TAL, or CAM administered in 10 equal doses daily. Intravenous glucose tolerance test (IVGTT) and i.v. insulin challenge (IC) were performed on d 5 after 98 and 108 h of treatment infusions, respectively. Infusion of lipids increased basal plasma NEFA concentrations on d 5 (CAM: 0.25; TAL: 0.28; control: 0.17 mmol/L). Following glucose injection, the rate of glucose clearance (CR) was lower in lipid-treated cows (CAM: 1.34; TAL: 1.48; control: 1.74%/min) and time to reach half-maximal glucose concentration (T(1/2)) was longer (CAM: 54; TAL: 47; control: 42 min). Similar responses were observed after insulin injection. Increased plasma NEFA concentration tended to decrease insulin secretion in IVGTT. Infusion of CAM increased plasma C18:3n-3 content (CAM: 26.4; TAL: 16.1; control: 20.9 g/100g of fatty acids). Data suggest that CAM had an insulin-sensitizing effect, because the disposition index and insulin sensitivity index, derived from minimal model analysis, were higher in CAM than in TAL during IVGTT, and lower insulin concentrations during IC led to similar glucose clearance in CAM as in TAL. These results indicate that elevated plasma NEFA concentration per se induces whole-body insulin resistance in late-pregnant dry cows.  相似文献

Dietary unsaturated fatty acids are extensively hydrogenated in the rumen, resulting in the formation of numerous intermediates that may exert physiological effects and alter the fat composition of ruminant-derived foods. A batch culture method was used to characterize the hydrogenation of linoleic acid (LeA) by strained rumen fluid in vitro. Incubations (n = 5) were performed in 100-mL flasks maintained at 39 °C containing 400mg of grass hay, 50 mL of buffered rumen fluid, and incremental amounts of LeA (0, 1.0, 2.5, 5.0, or 10.0mg) for 0, 1.5, 3.0, 4.5, 6.0, and 9.0 h. The fatty acid composition of flask contents was determined using complimentary silver-ion thin-layer chromatography, gas chromatography mass-spectrometry, and silver-ion high-performance liquid chromatography. Linoleic acid was extensively (98.1, 97.6, 98.0, and 89.8% for additions of 1.0, 2.5, 5.0, and 10.0mg of LeA, respectively) hydrogenated over time. Complete reduction of LeA to 18:0 was inhibited in direct relation to the amount of added substrate, the extent of which was greatest for the highest amount of LeA addition. Recoveries of 1.0, 2.5, 5.0, and 10.0mg of added LeA as 18:0 averaged 73.6, 65.0, 57.3, and 10.7%, respectively. Incubation of incremental amounts of LeA resulted in a time-dependent accumulation of geometric isomers of 9,11 and 10,12 conjugated linoleic acid, several nonconjugated 18:2 isomers, and a wide range of cis 18:1 and trans 18:1 intermediates. Several unusual intermediates including cis-6,cis-12 18:2; cis-7,cis-12 18:2; and cis-8,cis-12 18:2, were found to accumulate in direct relation to the amount of added LeA, providing the first indications that hydrogenation of LeA by ruminal bacteria may also involve mechanisms other than hydrogen abstraction or isomerization of the cis-12 double bond. Fitting of single-pool, first-order kinetic models to experimental data indicated that the rate of LeA disappearance decreased with increases in substrate availability. Reduction of 18:1 and 18:2 intermediates occurred at much lower rates compared with conjugated linoleic acid and nonconjugated 18:2 isomer formation. In conclusion, the extent of LeA biohydrogenation in vitro was shown to be time- and dose-dependent with evidence that LeA is hydrogenated by ruminal bacteria via several distinct metabolic pathways. The accumulation of several unusual 18:2 isomers indicates that biohydrogenation of LeA also proceeds via mechanisms other than isomerization of the cis-12 double bond.  相似文献
