A study was conducted to determine the impact of exposure to dust in the cattle load-out area in feedyards on pathogen contamination of cattle hides. A total of 250 cattle hides were sampled during summer and fall months, which are associated with elevated prevalence of Escherichia coli O157 in West Texas. Animals were removed from their home pens and restrained in a chute and sampled prior to exposure to dust generated as a result of a simulated loading exercise. The cattle hides were sampled again after exposure to the loading dust to determine total numbers of pathogens on cattle hides on leaving their home pen (before loading) and on cattle hides after exposure to the dust in the loading area. Air and dirt samples from the home pens and the cattle load-out area were also collected. The presence of E. coli O157 and Salmonella was determined in all the samples, and when a positive sample was identified, the total numbers of these bacteria present were enumerated. The total numbers of pathogens increased after dust exposure; Salmonella counts increased from 1.09 log most probable number (MPN)/cm2 to 1.74 log MPN/cm2 after exposure, and E. coli O157 counts increased from 0.80 to 2.35 log MPN/cm2 after sampling. E. coli O157 and Salmonella were recovered from the air samples during dust generation at 6.66 and 11.1%, respectively. Salmonella and E. coli O157 prevalence was not changed and was not associated with the exposure to the dust. Results indicate airborne dust generated as a result of cattle movement and loading could be an important determining factor in total numbers of pathogens recovered on cattle hides.  相似文献

The effect of cow or goat milk-based diets, either normal or Fe-overloaded, on DNA stability was studied in control and anaemic rats for 30 or 50 days of chronic Fe repletion. DNA damage was assessed using the alkaline comet assay. Background DNA damage in lymphocytes of peripheral blood was much lower in control and anaemic rats, given the normal or Fe-overloaded goat milk-based diet after 30 and 50 days versus cow milk-based diet. Chronic Fe-overload had no adverse effect on DNA stability in control and anaemic fed the goat milk-based diet versus the same diet with normal-Fe content at the end of the study. The quality of goat milk fat, together with the high levels of bioavailable Mg and Zn may be responsible for its protective effect on DNA of peripheral blood lymphocytes under the different experimental conditions, even during chronic Fe-overload.  相似文献

A “constrained ” displacement finite element approach for studying the influence of transverse cracks and delaminations on the thermal response of laminated composites is presented. Typical results are given in the form of percent retention curves for the coefficient of thermal expansion as a function of crack density. Cross-ply and quasi-isotropic T300/5208 graphite-epoxy laminates are considered. It is shown that transverse cracks can have a significant influence on the coefficient of thermal expansion, but delaminations located symmetrically about the laminate mid-plane have no influence on thermal expansion.  相似文献

The goal of this project was to gain a better understanding of atrazine occurrence in the United States by surveying drinking water utilities' sources and finished water for atrazine on a weekly basis for seven months. Atrazine is a contaminant of interest because the United States Environmental Protection Agency (USEPA) has found short-term atrazine exposure above the drinking water maximum contaminant level (MCL) to potentially cause heart, lung, and kidney congestion, low blood pressure, muscle spasms, weight loss, and damage to the adrenal glands. Long-term exposure to atrazine concentrations above the drinking water MCL has been linked to weight loss, cardiovascular damage, retinal and muscle degeneration, and cancer. This survey effort improved upon previously conducted atrazine surveys through intensive, high frequency sampling (participating plants sampled their raw and finished water on a weekly basis for approximately seven months). Such an intensive effort allowed the authors to gain a better understanding of short-term atrazine occurrence and its variability in drinking water sources. This information can benefit the drinking water industry by facilitating (1) better atrazine occurrence management (i.e., awareness when plants may be more susceptible to atrazine), (2) more efficient atrazine control (e.g., effective treatment alternatives and more effective response to atrazine occurrence), and (3) treatment cost reduction (e.g., efficient atrazine control can result in substantial cost savings). Forty-seven drinking watertreatment plants located primarily in the Midwestern United States participated in the survey and sampled their raw and finished water on a weekly basis from March through October. Samples were analyzed using the Abraxis enzyme-linked immunosorbent assay (ELISA) test kit. Confirmation samples for quality assurance/quality control (QA/QC) purposes were analyzed using solid-phase extraction (SPE) followed by gas chromatography mass spectrophotometry (GC/MS). Several important conclusions can be drawn from this study including (1) surface waters were confirmed to be more vulnerable to atrazine contamination than groundwater sources, (2) peak atrazine concentrations corresponded well to precipitation/runoff events, and (3) atrazine occurrence tended to be uniform geographically when compared by river drainage basins. In addition, this project confirmed that the Abraxis atrazine ELISA test kit tended to have a positive bias (i.e., the measured ELISA concentration was higher than the actual concentration) in most measured samples. Finished samples tended to have more of a positive bias than raw water samples. Therefore, this bias may limit the effectiveness for ELISA for regulatory monitoring. There are many other applications for ELISA, however, including frequent monitoring for early detections of atrazine concentration changes that might trigger conventional analysis by GC/MS or be used for activated carbon dosing or other treatment operating controls.  相似文献

A range of physico‐chemical properties of 21 Californian red wines from 1 to 28 years old were correlated with wine age. The level of monomeric phenolics and low molecular‐weight phenolic oligomers decreased with age, while the level of high molecular‐weight phenolics increased. On the other hand, total phenols given by the Folin–Ciocalteau method, antioxidant capacity determined using a controlled chain reaction of methyl linoleate and the intensity of the anodic peak determined by cyclic voltammetry (CV) did not show any significant correlation with wine age. Total phenols and the shapes of CV traces were more dependent on the grape variety and the particular viticultural and wine‐making practices rather than on wine age. At the same time, young and aged wines showed a pronounced difference in the shape of [O2] traces recorded during the oxidation of methyl linoleate in a micellar solution inhibited by wine addition. Carbon gases (methane, CH4, and carbon dioxide, CO2) were measured for the first time in sediments of the Lobo‐Broa Reservoir, near São Carlos in São Paulo State, Brazil. It is believed these are the first measurements of this kind in any of the many reservoirs located in Brazil. Even though the Lobo‐Broa Reservoir is classified as oligotrophic, the sediment gas concentrations were exceedingly high, ranging from 0.4–3 mmol L?1 for CH4 and 1–9 mmol L?1 for CO2. Both gases exceeded their in situ gas saturation values at these shallow water depths (7 m in central basin; 11 m at dam), resulting in numerous sediment bubbles. Organic matter was highly concentrated in the reservoir sediments, averaging 25.5% loss on ignition (LOI) (dam) to 26.9% LOI (central basin) for the 0–12 cm depth interval, with values as high as 29–30% LOI (12% organic carbon) in the surface 0–5 mm layer. The theoretical flux of dissolved pore water carbon gases to the sediment–water interface (SWI) averaged 3.4 mmol L?1 m?2 day?1 CH4 and 7.3 mmol L?1 m?2 day?1 CO2 for the surface 0–10 mm. From gas emission measurements at the water surface, it was calculated that 90% of CH4 is consumed either at the SWI or in the water column, resulting in a loss of 0.31 mmol L?1 m?2 day?1 of CH4 to the atmosphere. However, only 20% of the total CO2 gas transported across the water–atmosphere interface (36.3 mmol L?1 m?2 day?1, or 1600 mg CO2 m?2 day?1) was produced in the sediments. The remaining 80% of CO2 probably comes from other carbon sources. With CH4 oxidation in the aerobic water column, close to 30% of the carbon gas flux to the atmosphere could be accounted for by gas production of CO2 and CH4 in the sediments and their diffuse transport to the water column.  相似文献

Intimate partner violence is a serious and pervasive social problem with deleterious consequences for survivors’ well-being. The current study involved interviewing 160 survivors 6 times over 2 years to examine the role of social support in explaining or buffering these negative psychological consequences. The authors examined both between- and within-persons variability to explore women’s trajectories regarding their experiences of abuse, social support, depression, and quality of life (QOL). Findings revealed the complex role of social support on women’s well-being. Evidence was found for main, mediating, and moderating effects of social support on women’s well-being. First, social support was positively related to QOL and negatively related to depression. Social support also partially explained the effect of baseline level and subsequent change in physical abuse on QOL and depression over time, partially mediated the effects of change in psychological abuse, and moderated the impact of abuse on QOL. The buffering effects of social support were strongest at lower levels of abuse. Implications for future research and intervention are discussed. Exponential increases in multiracial identities, expected over the next century, create a conundrum for perceivers accustomed to classifying people as their own- or other-race. The current research examines how perceivers resolve this dilemma with regard to the own-race bias. The authors hypothesized that perceivers are not motivated to include ambiguous-race individuals in the in-group and therefore have some difficulty remembering these individuals. Both racially ambiguous and other-race faces were misremembered more often than own-race faces (Study 1), though memory for ambiguous faces was improved among perceivers motivated to include biracial individuals in the in-group (Study 2). Racial labels assigned to racially ambiguous faces determined memory for these faces, suggesting that uncertainty provides the motivational context for discounting ambiguous faces in memory (Study 3). Finally, an inclusion motivation fostered cognitive associations between racially ambiguous faces and the in-group. Moreover, the extent to which perceivers associated racially ambiguous faces with the in-group predicted memory for ambiguous faces and accounted for the impact of motivation on memory (Study 4). We introduce a novel algorithm that decomposes a deformable shape into meaningful parts requiring only a single input pose. Using modal analysis, we are able to identify parts of the shape that tend to move rigidly. We define a deformation energy on the shape, enabling modal analysis to find the typical deformations of the shape. We then find a decomposition of the shape such that the typical deformations can be well approximated with deformation fields that are rigid in each part of the decomposition. We optimize for the best decomposition, which captures how the shape deforms. A hierarchical refinement scheme makes it possible to compute more detailed decompositions for some parts of the shape.
Although our algorithm does not require user intervention, it is possible to control the process by directly changing the deformation energy, or interactively refining the decomposition as necessary. Due to the construction of the energy function and the properties of modal analysis, the computed decompositions are robust to changes in pose as well as meshing, noise, and even imperfections such as small holes in the surface.  相似文献
