The objective of this paper was to study the main chemical and microstructural changes in squid proteins during processing
as a frozen batter-coated product using different coatings. Protein bands of the squid and of the batters used for coating
were studied using SDS-PAGE. It is concluded that it is not only the proteins of the layer of batter, which are directly exposed
both to the frying oil and to freezing temperatures, that deteriorate during the preparation process, the protein fraction
of the squid substrate, mainly the myofibrillar proteins, is also degraded. At a microstructural level, big voids are generated
during freezing as a consequence of the packing of the fibres, although the size of these voids decreases after final frying.
After this step of the process, the central sarcoplasm is still visible but has been altered by coagulation of the sarcoplasmic
proteins and the sarcolemma is observed to be separate from the myofibrillar package. 相似文献

Legumes can be a source of mineral elements but also of antinutritional factors which can affect their absorption. An in vitro method including gastrointestinal digestion was used to estimate mineral bioavailability. Soluble (bioaccessible) and insoluble calcium, iron and zinc from white beans, chickpeas and lentils were determined after gastrointestinal digestion. The influence of the original sample weight on the soluble mineral fraction was also estimated. The results obtained show that white beans are the legumes with the highest bioaccessible calcium and iron contents. Lentils have a high iron content but its bioaccessibility is much lower than that of iron from white beans and chickpeas. An increase in sample weight increases the amount of bioaccessible element available for intake, but the increase is not always proportional. 相似文献

BACKGROUND: The polymerase chain reaction (PCR) is a powerful tool that is being increasingly used for detection of transgenic DNA. PCR requires only a minute quantity of template, but sensitive and accurate testing requires DNA of sufficient purity and free from inhibitors such as plant polysaccharides. Several standard protocols are available for this purpose, but they usually involve several steps, imply destruction of the maize kernel, or are time‐consuming. Our aim was to develop a fast and simple extraction method to isolate a raw DNA‐containing solution from maize tissues suitable for use as a template in a PCR‐based detection assay with specific oligonucleotides directed to the identification of event MON810. RESULTS: The NaOH‐based DNA extraction method we report here is time‐saving (5 min) and can be used to isolate DNA‐containing solutions from a small maize leaf portion (down to 1 mg) or from a single overnight‐germinated kernel. PCR performed with selected primers yielded reproducible detection of transgenic DNA. CONCLUSION: The main advantages of the procedure are the quick extraction step, the possibility of non‐destructive testing of maize kernels, and the robustness of the PCR‐based detection, a consequence of the selection of MON810‐matching oligonucleotides yielding intense and highly specific amplicons. This paper presents a new approach to information retrieval from non-structured attributes in databases, which involves the processing of text attributes. To make retrieval more effective, frequent text sequences are extracted and mathematically represented as intermediate forms which permit a clearer and more precise definition of operations on texts. These intermediate forms appear to users in the form of tag clouds to facilitate content identification, exploration, and querying. In this sense, tag cloud visualization is a simple, user-friendly visual interface to data. This paper proposes a theoretical model for the representation of frequent text sequences and their operations as well as a general procedure for generating tag clouds from text attributes in databases. The tag clouds thus obtained were compared with conventional tag clouds composed of single terms. Our study showed that automatically generated multi-term tag clouds provide better results than mono-term tag clouds. 相似文献

The article presents a method for learning the weights in one-layer feedforward neural networks minimizing either the sum of squared errors or the maximum absolute error, measured in the input scale. This leads to the existence of a global optimum that can be easily obtained solving linear systems of equations or linear programming problems, using much less computational power than the one associated with the standard methods. Another version of the method allows computing a large set of estimates for the weights, providing robust, mean or median, estimates for them, and the associated standard errors, which give a good measure for the quality of the fit. Later, the standard one-layer neural network algorithms are improved by learning the neural functions instead of assuming them known. A set of examples of applications is used to illustrate the methods. Finally, a comparison with other high-performance learning algorithms shows that the proposed methods are at least 10 times faster than the fastest standard algorithm used in the comparison. 相似文献

Frequently multivariate experimental data taken over multiple occasions is found to produce a multivariate multi-table experiment. Neither the separate analysis of each occasion, using MANOVA or canonical variate analysis, nor the joint analysis using the STATIS-ACT methodology for multiple tables, are adequate to capture the real structure of the data matrices, since the first account for group structure but not time evolution and the second confuses the “between” and “within” group variabilities. A method baptized Canonical STATIS, based on STATIS-ACT methodology, is proposed. The method permits to account for data group structure as well as time evolution on the various occasions by obtaining common or stable canonical variates across multiple occasions or data sets. A simultaneous representation of groups, variables and occasions (biplot) is obtained, thereby widening the capabilities of classical methods. 相似文献

BACKGROUND: In Citrus the inhibitory effect of fruit on flower formation is the main cause of alternate bearing. Although there are some studies reporting the effect on flowering of the time of fruit removal in a well‐defined stage of fruit development, few have investigated the effect throughout the entire fruit growth stage from early fruitlet growth to fruit maturity. The objective of this study was to determine the phenological fruit developmental stage at which the fruit begins its inhibitory effect on flowering in sweet orange by manual removal of fruits, and the role of carbohydrates and nitrogen in the process. RESULTS: Fruit exerted its inhibitory effect from the time it was close to reaching its maximum weight, namely 90% of its final size (November) in the present experiments, to bud sprouting (April). The reduction in flowering paralleled the reduction in bud sprouting. This reduction was due to a decrease in the number of generative sprouted buds, whereas mixed‐typed shoots were largely independent of the time of fruit removal, and vegetative shoots increased in frequency. The number of leaves and/or flowers per sprouted shoot was not significantly modified by fruit load. CONCLUSION: In ‘Valencia’ sweet orange, fruit inhibits flowering from the time it completes its growth. Neither soluble sugar content nor starch accumulation in leaves due to fruit removal was related to flowering intensity, but some kind of imbalance in nitrogen metabolism was observed in trees tending to flower scarcely. A spectrometric method for the determination of L-carnitine has
been developed based on the reaction of the 5,5′ dithiobis-(2-nitrobenzoic) acid (DTNB) and adapted to a Technicon RA-2000 automatic analyser Química Farmacéutica Bayer, S.A.). The detection limit of the method is 13.2 μmol/l, with a measurement interval ranging from 30 to 320 μmoll1. Imprecision and accuracy are good even at levels close to the detection limit (coeffcient of variation of 5.4% for within-run imprecision for a concentration of 35 μmol/l). A good correlation was observed between the method studied and the radiometric method. The method evaluated has suffcient analytical sensitivity to diagnose carnitine deficiencies. The short time period required for sample processing (30 samples in 40min), the simple methodology and apparatus, the ease of personnel training and the low cost of the reagents make this method a good alternative to the classical
radiometric method for evaluating serum L-carnitine in clinical laboratories without radioactive installations. 相似文献

Sardine (Sardina pilchardus) is a species that for its abundance assumes great importance in the Portuguese fishing sector. In order to contribute for a better utilisation of this species, the aim of this study was to investigate the effect of the pre‐treatment with soluble gas stabilisation (SGS) (100% CO2 at 2 bar, during 15 and 30 min) on the quality and shelf‐life of sardine fillets, packed in air (AP), vacuum (VP) and modified atmosphere (MAP: 5% O2/35% CO2/60% N2). During the chilled storage, the quality changes were evaluated by sensory evaluation, chemical and microbiological analysis. The total volatile basic nitrogen content remained almost constant, between 16 and 19 mg N/100 g muscle, during the storage period, for all samples. The TBARs values increased with storage time, for all batches and storage conditions. The application of SGS treatment to sardine fillets, resulted in a bacteriostatic effect, contributing to the improvement of the microbiological quality of fillets. Considering a sensory criteria, the shelf‐life of SGS pre‐treated sardine fillets was found to be 5 days in AP and MAP while in VP‐treated fillets a shelf‐life of 8 days was reported. At sensory rejection, sardine fillets presented a K‐value of 30% in AP and MAP batches and 40% in VP batch. 相似文献
