OBJECTIVES: Glycaemic control often deteriorates during puberty in girls with insulin dependent diabetes mellitus (IDDM). This may be due in part to the normal psychosocial changes associated with adolescence. Puberty is, however, also characterized by rapid somatic development, orchestrated by hormonal changes. Some of these hormones play a major role in glucose homeostasis. We have examined the insulin-GH-IGF-I axis in 11 adolescent girls with poorly controlled insulin dependent diabetes and compared the data with those of 10 non-diabetic girls matched for age, pubertal stage and body mass index (BMI). METHODS: Serum profiles of glucose, insulin, GH and IGF binding protein 1 (IGFBP1) were analysed in addition to IGF-I in serum and nocturnal urinary excretion of GH. MEASUREMENTS: Serum glucose, insulin and IGFBP1 were measured every hour for 24 h, whereas GH in serum was measured every 30 minutes during the same period. Nocturnal urinary GH was analysed as a mean of three consecutive nights. RESULTS: The insulin profiles of the IDDM patients were flat with low post-prandial peaks, corresponding to only one-third of the peaks of the non-diabetic girls. The integrated insulin levels, both during 24-h sampling and during daytime, were significantly lower in the diabetic group. There were no differences during night-time. The diabetic patients had elevated mean baseline levels of serum GH (IDDM 2.8 +/- 0.5 mU/l, controls 0.7 +/- 0.2; P < 0.001), a higher 24-h mean serum GH level (9.8 +/- 1.7 mU/l vs. 4.4 +/- 0.7; P < 0.001), significantly more peaks and a urinary GH excretion twice as high as in the non-diabetic group. An interesting observation was the finding of marked differences in daytime GH concentrations between the groups, both regarding overall integrated levels (GH AUC 103 +/- 15.8 and 35.9 +/- 7.1 mU/l x 12 h, respectively; P < 0.005) as well as baseline levels (3.8 +/- 0.6 mU/l vs. 0.7 +/- 0.2; P < 0.001). In contrast, during night-time only the mean basal levels of GH differed. The level of IGF-I was reduced in the diabetic group compared with the healthy controls (IDDM 233 +/- 19 micrograms/l vs. controls 327 +/- 21; P < 0.005). In addition, the IDDM patients had significantly increased concentrations of IGFBP 1, but kept a normal diurnal rhythm with a pronounced night peak. CONCLUSION: Hypoinsulinaemia in adolescent IDDM patients, particularly in the portal hepatic circulation, results in decreased IGF-I and increased IGFBP 1 production in the liver. High levels of IGFBP 1 may, in turn, reduce the bioactivity of IGF-I even further. Low levels of IGF-I will lead to increased GH secretion. Earlier studies on the relationship between GH and diabetic control have focused on elevated GH levels during the night. In this study we have observed markedly elevated levels of GH also during daytime in adolescent IDDM patients. This indicates increased insulin resistance and insulin demand also during the day in diabetic subjects. The increased insulin resistance may result in hyperglycaemia leading to additional insulin resistance. A vicious circle may thus be induced, accelerating metabolic impairment in poorly controlled adolescent IDDM girls.  相似文献

It is understood that defects of the atomic arrangement of the lattice in 2D molybdenum disulfide (MoS2) grown by chemical vapor deposition (CVD) can have a profound effect on the electronic and optical properties. Beyond these it is a major prerequisite to also understand the fundamental effect of such defects on phonon transport, to guarantee the successful integration of MoS2 into the solid‐state devices. A comprehensive joint experiment‐theory investigation to explore the effect of lattice defects on the thermal transport of the suspended MoS2 monolayer grown by CVD is presented. The measured room temperature thermal conductivity values are 30 ± 3.3 and 35.5 ± 3 W m?1 K?1 for two samples, which are more than two times smaller than that of their exfoliated counterpart. High‐resolution transmission electron microscopy shows that these CVD‐grown samples are polycrystalline in nature with low angle grain boundaries, which is primarily responsible for their reduced thermal conductivity. Higher degree of polycrystallinity and aging effects also result in smoother temperature dependency of thermal conductivity (κ) at temperatures below 100 K. First‐principles lattice dynamics simulations are carried out to understand the role of defects such as isotopes, vacancies, and grain boundaries on the phonon scattering rates of our CVD‐grown samples.  相似文献

A methodology to improve the performance of a hybrid solid oxide fuel cell gas turbine (SOFC‐GT) system for the whole operating range is proposed. The method suggests a way to estimate the geometric parameters of the turbomachinery components for a hybrid SOFC‐GT system. It is based on the search of the compressor and turbine operating lines giving the optimum system efficiency both in design and part load operation. Turbomachinery models are used to calculate the geometry that produces the desired performance maps and the corresponding operating lines. Based on the new turbomachinery design, the hybrid system shows a clear efficiency advantage for the whole operating range. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

Glutathione transferases (GSTs; EC 2.5.1.18) form a group of multifunctional enzymes that are involved in phase II of the cellular detoxification mechanism and are associated with increased susceptibility to cancer development and resistance to anticancer drugs. The present study aims to evaluate the ligandability of the human GSTM1-1 isoenzyme (hGSTM1-1) using a broad range of structurally diverse pesticides as probes. The results revealed that hGSTM1-1, compared to other classes of GSTs, displays limited ligandability and ligand-binding promiscuity, as revealed by kinetic inhibition studies. Among all tested pesticides, the carbamate insecticide pirimicarb was identified as the strongest inhibitor towards hGSTM1-1. Kinetic inhibition analysis showed that pirimicarb behaved as a mixed-type inhibitor toward glutathione (GSH) and 1-chloro-2,4-dinitrobenzene (CDNB). To shine a light on the restricted hGSTM1-1 ligand-binding promiscuity, the ligand-free crystal structure of hGSTM1-1 was determined by X-ray crystallography at 1.59 Å-resolution. Comparative analysis of ligand-free structure with the available ligand-bound structures allowed for the study of the enzyme’s plasticity and the induced-fit mechanism operated by hGSTM1-1. The results revealed important structural features of the H-site that contribute to xenobiotic-ligand binding and specificity. It was concluded that hGSTM1-1 interacts preferentially with one-ring aromatic compounds that bind at a discrete site which partially overlaps with the xenobiotic substrate binding site (H-site). The results of the study form a basis for the rational design of new drugs targeting hGSTM1-1.  相似文献

This research investigated therapist interpretations and subsequent client actions in 3 therapeutic modalities--client-centered therapy, gestalt/existential therapies, and rational-emotive behavior therapy-by employing the coding system of the consensual qualitative research method (C. E. Hill, B. J. Thompson, & N. E. Williams, 1997). By allowing conceptual categories to emerge from these qualitatively analyzed data, the authors found that each approach had its own specific pattern of interpretation content as well as style of delivering the interpretations. Client reactions to interpretations were mostly positive. The results suggest that interpretation is a common therapeutic element and that different therapies nonetheless accentuate different aspects of interpretation. Implications for psychotherapy integration are offered, and the findings are discussed with reference to theory and clinical practice. This paper reports on the practises of bioinformatics research in South Africa using bibliometric techniques. The search strategy was designed to cover the common concepts in biological data organisation, retrieval and analysis; the development and application of tools and methodologies in biological computation; and related subjects in genomics and structural bioinformatics. The South African literature in bioinformatics has grown by 66.5% between 2001 and 2006. However, its share of world production is not on par with comparator countries, Brazil, India and Australia.  相似文献

Fluorine research has been identified as a priority area in South Africa and the South African Nuclear Energy Corporation (NECSA) is embarking in an effort to expand its hydrogen fluoride and aluminium trifluoride production capacity. On the eve of those efforts this article reports the findings of an effort to map and assess fluorine research in South Africa in comparison to four other countries i.e. Malaysia, Australia, Germany and Italy. The results of the assessment are aimed at guiding future directions for fluorine research in the country, at identifying centres of expertise nationally where new research chairs could be established, at identifying international centres of expertise to be utilised for collaboration and of course for inter-temporal benchmarking of fluorine research in South Africa. South Africa is identified to be producing a small number of fluorine research publications in comparison to other countries like Germany and Italy which produce orders of magnitude larger number of publications and in comparison to country’s total research effort. Furthermore the relevant research effort appears to be dispersed geographically and in disciplinary terms. Relevant recommendations are provided with particular emphasis on the pluralistic science policy approach followed in the country.  相似文献
