We investigated the effect of auxin and acetylcholine on the expression of the tomato expansin gene LeEXPA2, a specific expansin gene expressed in elongating tomato hypocotyl segments. Since auxin interferes with clathrin-mediated endocytosis, in order to regulate cellular and developmental responses we produced protoplasts from tomato elongating hypocotyls and followed the endocytotic marker, FM4-64, internalization in response to treatments. Tomato protoplasts were observed during auxin and acetylcholine treatments after transient expression of chimerical markers of volume-control related compartments such as vacuoles. Here we describe the contribution of auxin and acetylcholine to LeEXPA2 expression regulation and we support the hypothesis that a possible subcellular target of acetylcholine signal is the vesicular transport, shedding some light on the characterization of this small molecule as local mediator in the plant physiological response.  相似文献

Silicon feedstock for production of solar-grade silicon should be as pure as possible to decrease the cost of manufacturing of solar cells. Impurities in quartz, carbonaceous materials, electrodes, and refractories are mostly present in the form of oxides. These oxides can be reduced to volatile gaseous compounds in presence of SiO(g) and CO(g) atmosphere and potentially leave the furnace or stay in the condensed reaction products, metal, and slag. This work investigates the conditions under which volatile impurities report to the gas phase in laboratory experiments with lumpy and pelletized mixtures of SiO2, SiC, and Si at 1923 K and 2123 K (1650 °C and 1850 °C), respectively, were carried out. The volatile compounds were generated by the reduction of quartz and collected in the form of condensate. The effects of the reaction temperature, quartz type, charge composition, pellets, and lumps on the composition of the condensate were studied. The trace elements in the charge input, reacting charge, and condensate were analyzed using inductively coupled plasma (ICP)-mass spectroscopy (MS) and X-ray diffraction (XRD). CO(g) and SiO(g), which are the major components in reduction reactions, formed four types of condensate: white, brown, green, and orange. The condensate constituents were amorphous SiO2, 3C:SiC, Si, and α-quartz. Each impurity present in the quartz charge entered the gas phase during quartz reduction and was detected in the condensate. Al and Fe show limited volatility. The volatility of Mn, P, and B depends on the charge mix: a higher PCO enhances the concentration of these elements in the gas phase. Fluid inclusions, common in hydrothermal quartz, enhance the distribution of the contaminants to the gas phase. Industrial campaigns on Si and Fe-Si production confirm the experimental results.  相似文献

A novel genetic variant at the αs1-casein locus of water buffalo (WB), 8-residue shorter than its wild-type has been found and sequenced. The internal deletion of the peptide E35KVNELsT42 was confirmed by the isolation of the junction peptide. The 8-residue deletion mutant has a molecular weight that is 919 Da less than that of the wild-type. The novel isoform with a unique f35-42 deletion could be the result of the skipping of exon 6, generating an exon 6-deleted variant of αs1-casein. The wild-type and its shortened αs1-casein forms were found to co-exist in many individual milk samples. In contrast, the 8-residue, internally deleted αs1-casein variant did not occur in water buffaloes of the Mediterranean breed reared in Italy. Wild-type αs1-casein has 6 to 8 phosphate groups (P) while the internally deleted form 6 and 7P per molecule.  相似文献

Incompatible land use is a major contributor to ecosystem degradation, and is often exacerbated by climate change impacts. We investigate Lake Basotu, Tanzania as a case study where natural lake variability has been affected by agricultural land use. Comparisons between a satellite-derived history of lake surface area, local precipitation records, and corresponding anthropogenic activity show the impacts of agricultural and historical practices. We argue that insufficient consideration to the wider ecological impacts of large agricultural projects has lasting implications. This is particularly true in semi-arid environments where food production demands need to be continuously met. In the future, major conservation strategies should be investigated to maintain the environmental integrity and sustainability of freshwater resources.  相似文献

A history of land clearance and riparian tree and wood management has resulted in limited riparian woodland and wood along British rivers. However, river management approaches are now being promoted that ‘work with natural processes’ to reduce flood risk through measures intended to restore, protect and emulate the natural function of catchments, rivers, estuaries and coasts. Wood has started to be reintroduced into river channels during river restoration and natural flood management activities, but this needs to be undertaken using appropriate quantities, locations and designs that mimic natural tree‐wood features. This paper reviews the knowledge that is needed to support wood reintroduction activities including (i) the characteristics of the riparian tree species and wood that are present; (ii) the importance of river size relative to that of trees and wood pieces; and (iii) the way trees, wood and geomorphic processes interact across rivers and floodplains of different energy and style.  相似文献

New Azoolefines and their Acidic cleavage to Aryldiimines 3-Amino-1-aryl-3′,3′-dimethyl-pyrazolin-4-spiro-2′-oxiran-5-ones ( 3a – c ) undergo ring opening with methoxide forming methyl 3-amino-3-arylazo-propenoates ( 5a – c ). 5a – c are cleaved under acidic conditions. The main products of the cleavage of 5c with methanolic hydrochloric acid are nitrogen, 2, 4, 6-trichloro-benzen ( 6 ), 2, 4, 6-trichloro-aniline ( 9 ) and 2, 4, 6-trichlorophenylhydrazine ( 10 ). Proenkephalin (PENK) and prodynorphin (PDYN) are endogenous opioid peptides mainly produced in the striatum and, to a lesser extent, in the cerebral cortex. Dysregulated metabolism and altered cerebrospinal fluid (CSF) levels of PENK and PDYN have been described in several neurodegenerative diseases. However, no study to date investigated these peptides in the CSF of sporadic Creutzfeldt–Jakob disease (sCJD). Using liquid chromatography-multiple reaction monitoring mass spectrometry, we evaluated the CSF PDYN- and PENK-derived peptide levels in 25 controls and 63 patients with sCJD belonging to the most prevalent molecular subtypes (MM(V)1, VV2 and MV2K). One of the PENK-derived peptides was significantly decreased in each sCJD subtype compared to the controls without a difference among subtypes. Conversely, PDYN-derived peptides were selectively decreased in the CSF of sCJD MV2K, a subtype with a more widespread overall pathology compared to the sCJD MM(V)1 and the VV2 subtypes, which we confirmed by semiquantitative analysis of cortical and striatal neuronal loss and astrocytosis. In sCJD CSF PENK and PDYN were associated with CSF biomarkers of neurodegeneration but not with clinical variables and showed a poor diagnostic performance. CSF PDYN and PENK-derived peptides had no significant diagnostic and prognostic values in sCJD; however, the distinct marker levels between molecular subtypes might help to better understand the basis of phenotypic heterogeneity determined by divergent neuronal targeting.  相似文献
