We have recently demonstrated that hydroxytyrosol (3,4-DHPEA), the most representative olive oil phenol, induces apoptosis on HL60 cells through the production of considerable amount of extracellular hydrogen peroxide (H2O2). The aims of the present investigation were first to assess the ability of different phenolic compounds to both produce extracellular H2O2 and induce apoptosis on HL60 cells, and second to elucidate whether the pro-apoptotic activity was mediated by the production of H2O2 in the cell culture medium. Based on the results phenols can be classified as follows: (1) those which were not able to induce both apoptosis and H2O2 accumulation (tyrosol, homovanillic alcohol and protocatechuic, o-coumaric, vanillic, homovanillic, ferulic and syringic acids); (2) those which showed a pro-apoptotic activity mediated, at least in part, by the production of H2O2, as evidenced by the ability of catalase to inhibit apoptosis (3,4-DHPEA, dopamine, 3,4-dihydroxyphenylacetic, 3,4-dihydroxy-hydrocinnamic, caffeic and gallic acids); and (3) those which induced apoptosis without the involvement of H2O2 (the secoiridoid derivatives of both hydroxytyrosol and tyrosol). Oleuropein showed a peculiar behaviour since, and although it caused an abundant production of H2O2 in the cell culture medium, it exerted a weak pro-apoptotic effect. From these results we may conclude that the cathecol moiety of the phenol molecule is necessary for the H2O2 producing activity, and that the 3,4-DHPEA metabolism to homovanillic alcohol and homovanillic acid may significantly reduce its pro-apoptotic potential. The real in vivo meaning of the phenol-induced H2O2 production remains to be investigated.  相似文献

Fruit maturity indexes are crucial in harvest time determination and commercial context. The harvest time of apples, matching the desired commercial characteristics, is assessed through starch–iodine test in practice. Fruit halves are dipped into iodine solution and patterns are visually evaluated by experts comparing them to reference charts. Aim of the work was to study the relationships of near infrared (NIR) spectral images (1,000–1,700 nm), starch/starch-free patterns visually assessed and RGB color images. Spectral images of 88 Golden Delicious Klon B apples were sampled at seven different maturity stages. Partial least-squares discriminant analysis (PLSDA) technique was used on hyper-spectral NIR images to classify single pixels using its NIR reflectance spectrum. The response variable (i.e. the classification for each pixel) was identified through the matching of single pixel obtained with the color images, segmented in two classes (starch and starch-free), and the NIR hyper-spectral matrix. Mean hyper-spectral classification obtained through PLSDA modeling on individual apple correctly classified 80.81% of the total pixels, while the unique model, i.e. a single model including all the fruits, resulted in 66.33%. In the latter case, the relationship with the RGB classification showed high values (Pearson correlation coefficient r = 0.95). The present work shows the feasibility of NIR imaging spectroscopy as a tool for apple fruit maturity determination, avoiding expert’s subjective interpretation by traditional starch index assignments.  相似文献

An alkaline protease gene was amplified from genomic DNA and cDNA of the antagonistic yeast-like fungus Aureobasidium pullulans PL5, a biocontrol agent effective against Monilinia laxa on stone fruit and Botrytis cinerea and Penicillium expansum on pome fruits. An open reading frame of 1248 bp encoding a 415-amino acid (aa) protein with a calculated molecular weight (Mr) of 42.9 kDa and an isoelectric point (pI) of 4.5 was characterized. The cDNAALP5 gene had an 18-amino acid signal peptide, one N-gylcosylation, one histidine active site, and one serine active site. The ALP5 gene with a Mr of 1351 bp contained two introns. One intron was of 54 bp, while the other was of 50 bp. Protein BLAST and phylogenetic tree analysis of the deduced amino sequences from the cDNAALP5 gene showed that the encoded protein had 100% homology to a protease enzyme (ALP2) of a sea strain of A. pullulans, suggesting that the protein ALP5 was an alkaline serine protease. Expression of ALP5 in Escherichia coli BL21 (DE3), followed by identification with Western-blotting, purification with Ni-NTA and analysis of enzymatic activity, yielded an homogeneous recombinant ALP5 which hydrolysed the substrate casein and inhibited the mycelial growth of the pathogens. At its optimal pH of 10.0 and reaction temperature of 50 °C, the recombinant protease exhibited the highest activity towards the substrate casein, though the highest stability was at lower temperatures and pH between 7.0 and 9.0. This study provided the direct evidence that extracellular proteases secreted by the antagonist A. pullulans PL5 played a role in the biocontrol activities against some postharvest pathogens of apple and peach.  相似文献

A product study of the reaction between a number of aromatic amines substituted with widely different groups and paraformaldehyde in inert solvents was performed and found to yield 1,3,5-triaryl-1,3,5-hexahydrotriazines, 1,3,5,7-tetraaryl-1,3,5,7-tetrazocines and formaminals. It was not possible to correlate the product outcomes with the actual structure of the amine substrate. The X-ray diffraction structural determination of 1,3,5-tri-(t-butylphenyl)-( 1b ) and 1,3,5-tri-(m-fluorophenyl)-1,3,5-hexahydrotriazine ( 1c ) showed the diaxial arrangement of the N-substituents.  相似文献

A rapid and efficient preparation of 1,3,5-triphenylhexahydro-sym-triazine ( 10 ) using paraformaldehyde is reported. The trimeric nature of the compound and its structure in solid state, where a biaxial chair conformation is favoured, are conclusively shown by X-ray diffraction analysis. In the present work, the corrosion resistance of ferritic-martensitic EUROFER 97 and ODS-EUROFER steels was tested in solutions containing NaCl or H2SO4 and KSCN, both at 25 °C. The results were compared to those of AISI 430 ferritic and AISI 410 martensitic conventional stainless steels. The as-received samples were tested by electrochemical techniques, specifically, electrochemical impedance spectroscopy, potentiodynamic polarization curves, and double-loop electrochemical potentiokinetic reactivation tests. The surfaces were observed by scanning electron microscopy after exposure to corrosive media. The results showed that EUROFER 97 and ODS-EUROFER alloys present similar corrosion resistance but lower than ferritic AISI 430 and martensitic 410 stainless steels.  相似文献

Epigenetic regulation is an essential process for the normal functioning of genes. Therefore, targeting epigenetic dysregulation in cancer may be a valid therapeutic approach for the treatment of this severe disease. Histone deacetylases (HDACs) are enzymes involved in the regulation of epigenetic post-translational modifications; because they are overexpressed in many types of cancer, HDACs are valuable targets for the development of new anticancer agents. A large series of 2-aminobenzanilides linked at the 4'-position to α-amino acid amides, arenes, and heteroarenes through a methylene bridge were designed, synthesized, and tested as novel HDAC inhibitors. Several compounds showed IC(50) values in the two-digit nanomolar range in hrHDAC1 inhibition assays, lower than that of the reference compound MS-275. They also showed interesting selectivity profiles, as confirmed by western blot assays.  相似文献

The multi-scale Gibbs–Helmholtz constrained (GHC) equation is a new predictive cubic equation of state that constrains the energy parameter in the SRK equation to satisfy the Gibbs–Helmholtz equation. It makes use of internal energies of departure calculated from NTP Monte Carlo simulations at the molecular length scale and a novel up-scaling expression to determine the energy parameter at the bulk phase length scale.  相似文献
