A variety of polymerization methods for the assembly of elongated nanoribbons from rectangular DNA origami structures are investigated. The most efficient method utilizes single-stranded DNA oligonucleotides to bridge an intermolecular scaffold seam between origami monomers. This approach allows the fabrication of origami ribbons with lengths of several micrometers, which can be used for long-range ordered arrangement of proteins. It is quantitatively shown that the length distribution of origami ribbons obtained with this technique follows the theoretical prediction for a simple linear polymerization reaction. The design of flat single layer origami structures with constant crossover spacing inevitably results in local underwinding of the DNA helix, which leads to a global twist of the origami structures that also translates to the nanoribbons. 相似文献

A successful metal hydride application is closely related to an optimized design of the storage hydrogen system. In previous studies, Hardy and Anton developed scoping and numerical models describing phenomena occurring during the loading process in an alanate storage system having the configuration of a cylindrical shell, tube and fin heat exchanger. In this paper, the numerical tool is used to evaluate the influence of varying the fin thickness and the number of heat exchanger tubes on both the loading and discharging processes. The objective is to evaluate the influence of the geometric parameters of these heat exchangers on the management of heat to be removed/supplied during the sorption process and thus optimize the loading/discharging times; while having the maximum possible volume for containing the hydride and the lightest weight of the storage system. Results showed that equipping the storage system with fins fitted to the heat exchanger tubes is the best design for efficient use of the hydride bed. In the absence of fins, a number of optimal tubes is determined, however, the hydrogen uptake rate is still lower than one obtained for the finned case and there is a reduction of volumetric and gravimetric storage capacities by comparison to the finned system. 相似文献

The use of graphene electrodes with hydrogenated edges for solid‐state nanopore‐based DNA sequencing is proposed, and molecular dynamics simulations in conjunction with electronic transport calculations are performed to explore the potential merits of this idea. The results of the investigation show that, compared to the unhydrogenated system, edge‐hydrogenated graphene electrodes facilitate the temporary formation of H‐bonds with suitable atomic sites in the translocating DNA molecule. As a consequence, the average conductivity is drastically raised by about 3 orders of magnitude while exhibiting significantly reduced statistical variance. Furthermore, the effect of the distance between opposing electrodes is investigated and two regimes identified: for narrow electrode separation, the mere hindrance due to the presence of protruding hydrogen atoms in the nanopore is deemed more important, while for wider electrode separation, the formation of H‐bonds becomes the dominant effect. Based on these findings, it is concluded that hydrogenation of graphene electrode edges represents a promising approach to reduce the translocation speed of DNA through the nanopore and substantially improve the accuracy of the measurement process for whole‐genome sequencing. 相似文献

Quantitative phosphorylation analysis is essential to understanding cellular signal transductions. Here we present a novel technology for the highly efficient assay of protein phosphorylation in high-throughput format without the use of phospho-specific antibodies. The technique is based on a water-soluble, nanosize polymer, termed pIMAGO, that is multifunctionalized with titanium(IV) ions for specific binding to phosphoproteins and with biotin groups that allow for enzyme-linked spectrometric detection. The sensitivity, specificity, and quantitative nature of pIMAGO for phosphorylation assays were examined with standard phosphoproteins and with purified phosphoproteins from whole cell extracts. As low as 100 pg of phosphoprotein can be measured quantitatively with the pIMAGO chemiluminescence assay. The pIMAGO assay was applied to an in vitro kinase assay, kinase inhibitor screening, and measurement of endogenous phosphorylation events. The technique provides a universal, quantitative method for global phosphorylation analysis with high sensitivity and specificity. 相似文献

The narrow particle size distribution (PSD) of certain packing materials has been linked to a reduced eddy dispersion contribution to band broadening in chromatographic columns. It is unclear if the influence of the PSD acts mostly on the stage of the packing process or if a narrow PSD provides an additional, intrinsic advantage to the column performance. To investigate the latter proposition, we created narrow-PSD and wide-PSD random packings based on the experimental PSDs of sub-3 μm core-shell and sub-2 μm fully porous particles, respectively, as determined by scanning electron microscopy. Unconfined packings were computer-generated with a fixed packing protocol at bed porosities from random-close to random-loose packing to simulate fluid flow and advective-diffusive mass transport in the packings' interparticle void space. The comparison of wide-PSD, narrow-PSD, and monodisperse packings revealed no systematic differences in hydraulic permeability and only small differences in hydrodynamic dispersion, which originate from a slightly increased short-range interchannel contribution to eddy dispersion in wide-PSD packings. The demonstrated intrinsic influence of the PSD on dispersion in bulk packings is negligible compared with the influence of the bed porosity. Thus, the reduced eddy dispersion observed for experimental core-shell packings cannot be attributed to a narrow PSD per se. 相似文献
