This paper presents the results of a three dimensional study for evaluating the temperature profiles and air flow movement in a model room with a numerical model based on the Euler equations. Numerical results obtained for two scenarios of ventilation and air conditioning are compared with the predictions of a Navier-Stokes model. These numerical results are validated by experimental results measured in the model room. A comparison of the local thermal comfort indices PMV and PPD obtained experimentally and numerically is also presented. Results show that the Euler model is able to predict adequately total thermal comfort in the model room. Furthermore, the use of Euler equations allows a reduction of computational time in the order of 50% compared to the Navier-Stokes modeling. 相似文献

Spreading of urban wastewater on agricultural land may lead to concomitant input of organic and inorganic pollutants. Such multiple pollution sites offer unique opportunities to study the fate of both heavy metals and pharmaceuticals. We examined the occurrence and fate of selected antibiotics in sandy-textured soils, sampled four years after cessation of 100 years irrigation with urban wastewater from the Paris agglomeration. Previous studies on heavy metal contamination of these soils guided our sampling strategy. Six antibiotics were studied, including quinolones, with a strong affinity for organic and mineral soil components, and sulfonamides, a group of more mobile molecules. Bulk samples were collected from surface horizons in different irrigation fields, but also in subsurface horizons in two selected profiles. In surface horizons, three quinolones (oxolinic acid, nalidixic acid, and flumequine) were present in eight samples out of nine. Their contents varied spatially, but were well-correlated one to another. Their distributions showed great similarities regarding spatial distribution of total organic carbon and heavy metal contents, consistent with a common origin by wastewater irrigation. Highest concentrations were observed for sampling sites close to irrigation water outlets, reaching 22 μg kg−1 for nalidixic acid. Within soil profiles, the two antibiotic groups demonstrated an opposite behavior: quinolones, found only in surface horizons; sulfamethoxazole, detected in clay-rich subsurface horizons, concomitant with Zn accumulation. Such distribution patterns are consistent with chemical adsorption properties of the two antibiotic groups: immobilization of quinolones in the surface horizons ascribed to strong affinity for organic matter (OM), migration of sulfamethoxazole due to a lower affinity for OM and its interception and retention in electronegative charged clay-rich horizons. Our work suggests that antibiotics may represent a durable contamination of soils, and risks for groundwater contamination, depending on the physicochemical characteristics both of the organic molecules and of soil constituents. 相似文献

Oenococcus oeni is responsible for the malolactic fermentation of wines. Genomic diversity has been recently established in the species and extensive attention is now being given to the genomic bases of strain-specific differences. We explored the role of insertion sequences (IS), which are considered as driving forces for novel genotypic and phenotypic variants in prokaryotes. The present study focuses on members of the IS30 family, which are widespread among lactic acid bacteria. An in silico analysis of the three available genomes of O. oeni in combination with the use of an inverse PCR strategy targeting conserved IS30-related sequences indicated the presence of seven IS30 copies in the pangenome of O. oeni. A primer designed to anneal to the conserved 3' end of the IS30 element was paired with each of the seven primers selected to bind to unique sequences upstream of each of the seven mobile elements identified. The study presents an overview of the abundance, and the genomic environment of IS30 elements in the O. oeni pangenome and shows that the two existing genetic sub-populations previously described in the species through multilocus sequence typing analysis (MLST) differ in their IS30 content. Possible IS30 impacts on bacterial adaptation are discussed. 相似文献

In inflammatory bone diseases such as periodontitis, the nucleotide-binding oligomerization domain, leucine-rich repeat, and pyrin domain-containing 3 (NLRP3) inflammasome accelerates bone resorption by promoting proinflammatory cytokine IL-1β production. However, the role of the NLRP3 inflammasome in physiological bone remodeling remains unclear. Here, we investigated its role in osteoclastogenesis in the presence and absence of lipopolysaccharide (LPS), a Gram-negative bacterial component. When bone marrow macrophages (BMMs) were treated with receptor activator of nuclear factor-κB ligand (RANKL) in the presence of NLRP3 inflammasome inhibitors, osteoclast formation was promoted in the absence of LPS but attenuated in its presence. BMMs treated with RANKL and LPS produced IL-1β, and IL-1 receptor antagonist inhibited osteoclastogenesis, indicating IL-1β involvement. BMMs treated with RANKL alone produced no IL-1β but increased reactive oxygen species (ROS) production. A ROS inhibitor suppressed apoptosis-associated speck-like protein containing a caspase-1 recruitment domain (ASC) speck formation and NLRP3 inflammasome inhibitors abrogated cytotoxicity in BMMs treated with RANKL, indicating that RANKL induces pyroptotic cell death in BMMs by activating the NLRP3 inflammasome via ROS. This suggests that the NLRP3 inflammasome promotes osteoclastogenesis via IL-1β production under infectious conditions, but suppresses osteoclastogenesis by inducing pyroptosis in osteoclast precursors under physiological conditions. 相似文献

Most antibacterial nanomaterials used in food packaging act by releasing reactive oxygen species (ROS), which cannot efficiently have an inhibitory effect by penetrating the cell wall of Gram-positive Staphylococcus aureus. In this work, we used the cruciform petal-like zeolite imidazole framework-8 (ZIF-8) synthesized in the water phase which can release active Zn compounds in aqueous solution and exert a stronger inhibitory effect on S. aureus. The experimental results demonstrated that the aqueous cruciform petal-like ZIF-8 has the same photocatalytic activity as traditional ZIF-8 and can be applied in photocatalytic bacterial inactivation. The cruciform petal-like ZIF-8 was also shown to release active Zn compounds in aqueous solution with a better antibacterial effect against S. aureus, reaching 95% inactivation efficiency. The antibacterial effect was therefore 70% higher than that of traditional ZIF-8. Based on its excellent antibacterial properties, we loaded petal-like ZIF-8, PDA and PVA onto ordinary fibers to prepare ZIF-8-Film. The results further showed that ZIF-8-Film has a high filtration capacity, which can be used in antibacterial packaging material with the required air permeability. Moreover, ZIF-8-Flim can clean the surface on its own and can maintain a sterile environment. Serotonin (5-hydroxytriptamine or 5-HT) is known to be a weak platelet agonist, and is involved in thrombus formation. While 5-HT cannot induce platelet aggregation on its own, when secreted from the alpha granules, it binds to its G-protein Coupled Receptor (GPCR; i.e., 5HT2AR), thereby acting to amplify platelet functional responses (e.g., aggregation). Thus, 5HT2AR-mediated responses are more involved in the secondary amplification of platelet aggregation in the growing thrombus. Therefore, even though 5-HT can be seen as a weak inducer of platelet activation, it is an important amplifier of aggregation triggered by agonists such as ADP, collagen, and epinephrine, thereby enhancing thrombogenesis. The 5HT2AR/5HT2A signaling pathway is of clinical interest to the scientific and medical communities as it has been implicated in the genesis of several forms of cardiovascular disorders. However, efforts to develop antagonists for 5HT2AR as therapeutic agents in cardiovascular diseases have thus far failed due to these reagents having deleterious side-effects, and/or to lack of selectivity, amongst other reasons. In light of research efforts that identified that the 5HT2AR ligand binding domain resides in the second extracellular loop (EL2; amino acids P209-N233), we developed an antibody, i.e., referred to as 5HT2ARAb, against the EL2 region, and characterized its pharmacological activity in the context of platelets. Thus, we utilized platelets from healthy human donors, as well as C57BL/6J mice (10–12 weeks old) to analyze the inhibitory effects of the 5HT2ARAb on platelet activation in vitro, ex vivo, and on thrombogenesis in vivo as well as on 5HT2AR ligand binding. Our results indicate that the 5HT2ARAb inhibits 5-HT-enhanced platelet activation in vitro and ex vivo, but has no apparent effects on that which is agonist-induced. The 5HT2ARAb was also found to prolong the thrombus occlusion time, and it did so without modulating the tail bleeding time, in mice unlike the P2Y12 antagonist clopidogrel and the 5HT2AR antagonist ketanserin. Moreover, it was found that the 5HT2ARAb does so by directly antagonizing the platelet 5HT2AR. Our findings document that the custom-made 5HT2ARAb exhibits platelet function blocking activity and protects against thrombogenesis without impairing normal hemostasis. 相似文献

With the increasing usage of drugs to remedy different diseases, drug safety has become crucial over the past few years. Often medicine from several companies is offered for a single disease that involves the same/similar substances with slightly different formulae. Such diversification is both helpful and dangerous as such medicine proves to be more effective or shows side effects to different patients. Despite clinical trials, side effects are reported when the medicine is used by the mass public, of which several such experiences are shared on social media platforms. A system capable of analyzing such reviews could be very helpful to assist healthcare professionals and companies for evaluating the safety of drugs after it has been marketed. Sentiment analysis of drug reviews has a large potential for providing valuable insights into these cases. Therefore, this study proposes an approach to perform analysis on the drug safety reviews using lexicon-based and deep learning techniques. A dataset acquired from the ‘Drugs.Com’ containing reviews of drug-related side effects and reactions, is used for experiments. A lexicon-based approach, Textblob is used to extract the positive, negative or neutral sentiment from the review text. Review classification is achieved using a novel hybrid deep learning model of convolutional neural networks and long short-term memory (CNN-LSTM) network. The CNN is used at the first level to extract the appropriate features while LSTM is used at the second level. Several well-known machine learning models including logistic regression, random forest, decision tree, and AdaBoost are evaluated using term frequency-inverse document frequency (TF-IDF), a bag of words (BoW), feature union of (TF-IDF + BoW), and lexicon-based methods. Performance analysis with machine learning models, long short term memory and convolutional neural network models, and state-of-the-art approaches indicate that the proposed CNN-LSTM model shows superior performance with an 0.96 accuracy. We also performed a statistical significance T-test to show the significance of the proposed CNN-LSTM model in comparison with other approaches. 相似文献

Phosphatidylcholines (PCs) are the major components of biological membranes in animals and are a class of phospholipids that incorporate choline as a headgroup. Lysophosphatidylcholines (LPCs) are a class of lipid biomolecules derived from the cleavage of PCs, and are the main components of oxidized low-density lipoproteins (oxLDLs) that are involved in the pathogenesis of atherosclerosis. Since obesity is associated with a state of chronic low-grade inflammation, one can anticipate that the lipidomic profile changes in this context and both PCs and LPCs are gaining attention as hypothetically reliable biomarkers of obesity. Thus, a literature search is performed on PubMed, Latin American and Caribbean Health Science Literature (LILACS), and Excerpta Medica DataBASE (Embase) to obtain the findings of population studies to clarify this hypothesis. The search strategy resulted in a total of 2403 reports and 21 studies were included according to the eligibility criteria. Controversial data on the associations of PCs and LPCs with body mass index (BMI) and body fat parameters have been identified. There is an inverse relationship between BMI and most species of PCs, and a majority of studies exhibited negative associations between BMI and LPCs. Other findings regarding the differences between PCs and LPCs in obesity are presented, and the associated uncertainties are discussed in detail. 相似文献

For modern high-tech flexible energy storage devices, it becomes important to synthesize micro-/nanostructures as per the required shape and morphology with superior physical and electro-active characteristics. This work shares the fabrication and characterization of ZnSn(OH)6 (Zinc hydroxystannate [ZHS]) prepared by facile microwave-assisted technique and furthermore converted into flexible sheets by employing lignocelluloses (LC) known as natural fibers, collected from Carica papaya leaf petiole as a substrate to provide the flexible matrix. X-ray diffraction measurements confirm the successful crystalline structure of ZHS. Scanning electron microscopy and transmission electron microscopy showed the solid spherical structure of ZHS microspheres. Fourier transform infrared spectrometry and Raman spectroscopy confirmed the composite formation of ZHS and LC-based composite sheets (ZHS/LC sheets). Electrochemical measurements that is, cyclic voltammetry (CV), Galvanostatic charge/discharge, and electrochemical impedance (EIS) spectroscopy revealed the electroactive behavior of ZHS/LC paper sheets as working electrode for energy storage applications. CV measurements revealed the specific capacitance of 100 F/g and EIS measurements confirmed the decrease in the resistance of LC fiber after the growth of ZHS microspheres.
