Exposure of Escherichia coli to a subminimal inhibitory concentration (25% below MIC) of benzalkonium chloride (BC), an antimicrobial membrane-active agent commonly used in medical and food-processing environments, resulted in cell death and changes in cell morphology (filamentation). A small subpopulation (1-5% of the initial population) survived and regained similar morphology and growth rate as non-exposed cells. This subpopulation maintained tolerance to BC after serial transfers in medium without BC. To withstand BC during regrowth the cells up regulated a drug efflux associated gene (the acrB gene, member of the AcrAB-TolC efflux system) and changed expression of outer membrane porin genes (ompFW) and several genes involved in protecting the cell from the osmotic- and oxidative stress. Cells pre-exposed to osmotic- and oxidative stress (sodium chloride, salicylic acid and methyl viologen) showed higher tolerance to BC. A control and two selected isolates showing increased BC-tolerance after regrowth in BC was genome sequenced. No common point mutations were found in the BC- isolates but one point mutation in gene rpsA (Ribosomal protein S1) was observed in one of the isolates. The observed tolerance can therefore not solely be explained by the observed point mutation. The results indicate that there are several different mechanisms responsible for the regrowth of a tolerant subpopulation in BC, both BC-specific and general stress responses, and that sub-MIC of BC may select for phenotypic variants in a sensitive E. coli culture.  相似文献

Abstract

When designed and built properly, hydrogen energy systems can enhance energy security through technological diversification and minimizing dependence on foreign imports of energy fuels. However, hydrogen can be produced from different feedstocks according to separate pathways, and these different pathways create particular consequences on a nation's overall energy security. The objective of this study is to investigate the superiorities and inferiorities of hydrogen pathways from the perspective of China and Denmark, and to determine which pathways best contribute to national energy security objectives. The results are useful for stakeholders and energy analysts so that they can correctly plan and research the most socially optimal portfolio of hydrogen technologies.  相似文献

The effect of modified atmosphere packaging (MAP) on the germination and growth of toxin producing psychrotolerant Bacillus spp is not well described. A model agar system mimicking a cooked meat product was used in initial experiments. Incubation at refrigeration temperature of 8 °C for 5 weeks of 26 Bacillus weihenstephanensis including two emetic toxin (cereulide) producing strains showed that B. weihenstephanensis is sensitive to MAP containing CO2. The sensitivity to 20% CO2 was dependent on strain and oxygen level, being increased when oxygen was excluded from the MAP. Growth from spores was observed at the earliest within 2 weeks when 20% CO2 was combined with 2% O2 and in 3 weeks when combined with “0”% O2 (the remaining atmosphere was made up from N2). Results were validated in a cooked meat sausage model for two non-emetic and one emetic B. weihenstephanensis strain. The packaging film oxygen transfer rates (OTR) were 1.3 and 40 ml/m2/24 h and the atmospheres were 2% O2/20% CO2 and “0”% O2/20% CO2. Oxygen availability had a large impact on the growth from spores in the MAP meat sausage, only the most oxygen restricted condition (OTR of 1.3 ml/m2/24 h and “0”% O2/20 % CO2) inhibited growth of the three strains during 4 weeks storage at 8 °C. Cereulide production was undetectable during storage at 8 °C irrespective of choice of the MAP (quantified by liquid chromatography mass spectrometry/mass spectrometry). MAP storage at 8 °C for 1 and 3 weeks followed by opening of packages and temperature abuse for 1.5 h daily at 20 °C during 1 week resulted in increased cell counts and variable cereulide production in the meat sausage. A pre-history at 8 °C for 1 week in MAP with OTR of 1.3 or 40 ml/m2/24 h and 2% O2 resulted in cereulide concentrations of 0.816 – 1.353 µg/g meat sausage, while a pre-history under the most oxygen restricted condition (OTR of 1.3 ml/m2/24 h, “0”% O2/20 % CO2) resulted in minimal cereulide production (0.004 µg/g meat sausage) at abuse condition. Extension of MAP storage at 8 °C for 3 weeks followed by abuse resulted in a substantially reduced cereulide production.Data demonstrates that MAP can be used to inhibit growth of a psychrotolerant toxin producing Bacillus spp. during chill storage at 8 °C, and substantially reduce the risk of emetic food poisoning at abuse condition. Results are of relevance for improving safety of ready to eat processed chilled foods of extended durability.  相似文献

The Welfare Quality (WQ) protocol for on-farm dairy cattle welfare assessment describes 27 measures and a stepwise method for integrating values for these measures into 11 criteria scores, grouped further into 4 principle scores and finally into an overall welfare categorization with 4 levels. We conducted an online survey to examine whether trained users' opinions of the WQ protocol for dairy cattle correspond with the integrated scores (criteria, principles, and overall categorization) calculated according to the WQ protocol. First, the trained users' scores (n = 8–15) for reliability and validity and their ranking of the importance of all measures for herd welfare were compared with the degree of actual effect of these measures on the WQ integrated scores. Logistic regression was applied to identify the measures that affected the WQ overall welfare categorization into the “not classified” or “enhanced” categories for a database of 491 European herds. The smallest multivariate model maintaining the highest percentage of both sensitivity and specificity for the “enhanced” category contained 6 measures, whereas the model for “not classified” contained 4 measures. Some of the measures that were ranked as least important by trained users (e.g., measures relating to drinkers) had the highest influence on the WQ overall welfare categorization. Conversely, measures rated as most important by the trained users (e.g., lameness and mortality) had a lower effect on the WQ overall category. In addition, trained users were asked to allocate criterion and overall welfare scores to 7 focal herds selected from the database (n = 491 herds). Data on all WQ measures for these focal herds relative to all other herds in the database were provided. The degree to which expert scores corresponded to each other, the systematic difference, and the correspondence between median trained-user opinion and the WQ criterion scores were then tested. The level of correspondence between expert scoring and WQ scoring for 6 of the 12 criteria and for the overall welfare score was low. The WQ scores of the protocol for dairy cattle thus lacked correspondence with trained users on the importance of several welfare measures.  相似文献

Division of labour between different specialized cell types is a central part of how we describe complexity in multicellular organisms. However, it is increasingly being recognized that division of labour also plays an important role in the lives of predominantly unicellular organisms. Saccharomyces cerevisiae displays several phenotypes that could be considered a division of labour, including quiescence, apoptosis and biofilm formation, but they have not been explicitly treated as such. We discuss each of these examples, using a definition of division of labour that involves phenotypic variation between cells within a population, cooperation between cells performing different tasks and maximization of the inclusive fitness of all cells involved. We then propose future research directions and possible experimental tests using S. cerevisiae as a model organism for understanding the genetic mechanisms and selective pressures that can lead to the evolution of the very first stages of a division of labour. In this paper a method for estimating the fuel cell impedance is presented, namely the current pulse injection (CPI) method, which is well suited for online implementation. This method estimates the fuel cell impedance and unlike electrochemical impedance spectroscopy (EIS), it is simple to implement at a low cost. This makes it appealing as a characterization method for on-line diagnostic algorithms. In this work a parameter estimation method for estimation of equivalent electrical circuit (EEC) parameters, which is suited for on-line use is proposed. Tests on a 10 cell high temperature PEM fuel cell show that the method yields consistent results in estimating EEC parameters for different current pulse at different current loads, with a low variance. A comparison with EIS shows that despite its simplicity the response of CPI can reproduce well the impedance response of the high and intermediate frequencies.  相似文献

Intraoral target (typing) and on-screen target (pointing/tracking) selection tasks were performed by 10 participants during 3 consecutive day sessions. Tasks were performed using 2 different intraoral sensor layouts. Reduction of undesired sensor activations while speaking as well as the influence of intraoral temperature variation on the signals of the intraoral interface was investigated. Results showed that intraoral target selection tasks were performed better when the respective sensor was located in the anterior area of the palate, reaching 78 and 16 activations per minute for repetitive and “unordered” sequences, respectively. Virtual target pointing and tracking tasks, of circles of 50, 70, and 100 pixels diameter, showed no significant difference in performance, reaching average pointing throughputs of 0.62 to 0.72 bits per second and relative time on target of 34% to 60%. Speaking tasks caused an average of 10 to 31 involuntary activations per minute in the anterior part of the palate. Intraoral temperature variation between 11.87 °C and 51.37 °C affected the sensor signal baseline in a range from –25.34% to 48.31%. Results from this study provide key design considerations to further increase the efficiency of tongue–computer interfaces for individuals with upper-limb mobility impairments.  相似文献

Poly(l ‐lactide) in its l ‐form has promising mechanical properties. Being a semicrystalline polymer, it can be subjected to strain‐induced crystallization at temperatures above Tg and can thereby become oriented. Following a simultaneous (SIM) biaxial strain process or a sequential (SEQ) biaxial strain process, the mechanical properties of biaxially strained tubes can be further improved. This study investigated these properties in relation to their morphology and crystal orientation. Both processes yield the same mechanical strength and modulus, yet exhibit different crystal orientation. Through further wide angle X‐ray scattering analysis it was found that the SEQ biaxial strain yields larger interplanar spacing and distorted crystals and looser packing of chains. However, this does not influence the mechanical properties negatively. A loss of orientation in SEQ biaxially strained samples at high degrees of strain was detected, but was not seen for SIM biaxial strain and did not correlate with mechanical performance in either case. However, post‐annealing reduced the orientation to the same level in both cases, and the modulus and strength decreased for both SIM and SEQ biaxial strain. It is therefore concluded that mechanical properties after biaxial strain are related to strain‐induced amorphous orientation and the packing of crystals, rather than strain‐induced crystallinity. © 2015 Society of Chemical Industry  相似文献

Sintered bulk samples of YBa2Cu3Ox of 95% to 99% theoretical density have been consolidated by a one-step densification process. Superconducting transitions determined by dc magnetization experiments are above 90 K. The metallurgical state of the material, both before and after consolidation, is characterized by X-ray diffraction and electron microscopy analysis as well as density and resistivity measurements. Improvement in critical current density (Jc) in the consolidated material is related to changes in the microstructure. Particular attention is focused on the granular nature of the compound and the influence of the consolidation process on the weak-link structure.  相似文献

Analyzed single-word and continuous association test behaviors of children at high (207 Ss) and low (104 Ss) risk for schizophrenia. A comparison was also made between test scores of those high-risk Ss who later became schizophrenic and scores of others who did not. Although high-risk Ss exhibited more deviant associations than low-risk Ss, premorbid associative disturbance did not characterize those who later became schizophrenic. These findings are not supportive of learning theories of associative disturbance. When combined with recent evidence showing that degree of associative disturbance at initial breakdown is not predictive of chronicity or outcome, these findings call for a reevaluation of the central character of associative disturbance in schizophrenia.
