A number of North American vegetable and animal fat shortenings were evaluated for their melting, crystallization, textural and polymorphic crystal characteristics and solid fat content (SFC). The majority of the dropping points and crystallization temperatures of the fats ranged from 42 to 46°C and from 27 to 31°C, respectively. Softening points of the products were higher than the dropping points of their fats, especially for the vegetable shortenings. Differential scanning calorimetry melting curves of the products were different for the various products. The animal fat shortenings were mainly in theβ-polymorphic form, while vegetable shortenings containing palm oil were in theβ′ form. Textural evaluation was carried out on the products with the cone penetrometer, constant speed penetration and constant speed compression. Constant speed compression supplied a measure of brittleness and a degree of viscosity. Lard and shortenings containing high levels of palm oil were able to withstand large deformations without breakage. The effect of tempering temperature of the fat in the SFC determination was evaluated and the values obtained were compared with the SFC of the actual product. SFC of fat and product were determined by pulse nuclear magnetic resonance. Correlation of values within textural methods was significant (P<..01), but were not significant between texture and SFC of the fat, indicating that the nature of the crystal network also plays a role in texture.  相似文献

Semiconductor nanoparticles or quantum dots are being increasingly utilized as fluorescent probes in cell biology both in live and fixed cell assays. Quantum dots possess an immense potential for use in multiplexing assays that can be run on high content screening analysers. Depending on the nature of the biological target under investigation, experiments are frequently required on cells retaining an intact cell membrane or also on those that have been fixed and permeabilized to expose intracellular antigens. Fixation of cell lines before or after the addition of quantum dots may affect their localization, emission properties and stability. Using a high content analysis platform we perform a quantitative comparative analysis of three common fixation techniques in two different cell lines exposed to carboxylic acid stabilized CdTe quantum dots. Our study demonstrates that in prefixed and permeabilized cells, quantum dots are readily internalized regardless of cell type, and their intracellular location is primarily determined by the properties of the quantum dots themselves. However, if the fixation procedures are preformed on live cells previously incubated with quantum dots, other important factors have to be considered. The choice of the fixative significantly influences the fluorescent characteristics of the quantum dots. Fixatives, regardless of their chemical nature, negatively affected quantum dots fluorescence intensity. Comparative analysis of gluteraldehyde, methanol and paraformaldehyde demonstrated that 2% paraformaldehyde was the fixative of choice. The presence of protein in the media did not significantly alter the quantum dot fluorescence. This study indicates that multiplexing assays utilizing quantum dots, despite being a cutting edge tool for high content cell imaging, still require careful consideration of the basic steps in biological sample processing.  相似文献

This paper presents a methodology for the assessment of the remaining load carrying capacity of thin‐walled components under tension containing highly strength undermatched welds and edge cracks. The analysis is based on the strength mismatch option of the fracture module, part of the newly developed European fitness‐for‐service (FFS) procedure FITNET. The mismatch option of the FITNET fracture module allows weld features such as weld tensile properties and weld geometry to be taken into account in the fracture analysis of cracked welded components. The methodology described was verified for centre cracked Al‐alloy large tensile panels containing undermatched welds in Ref. [ 1 ] and hence the present work provides validation with experimental results of the single edge cracked (SEC) and double edge cracked (DEC) panels. The material used is an age‐hardening aluminium alloy 6013 in T6 temper condition used in welded airframe components. The welds in the form of butt joints were produced using the CO2 laser beam welding process. The results show that by using the FITNET FFS methodology with an appropriate selection of the input parameters, safe acceptable predictions of the maximum load carrying capacity of the welded panels can be obtained. It should also be noted that one of the main difficulties that engineers encounter when applying mismatch analysis for first time is its apparent complexity. A step‐by‐step analysis is proposed here in order to provide guidance for this kind of assessments.  相似文献

Semiconductor heterojunctions are used in a wide range of applications including catalysis, sensors, and solar‐to‐chemical energy conversion devices. These materials can spatially separate photogenerated charge across the heterojunction boundary, inhibiting recombination processes and synergistically enhancing their performance beyond the individual components. In this work, the WO3/TiO2 heterojunction grown by chemical vapor deposition is investigated. This consists of a highly nanostructured WO3 layer of vertically aligned nanorods that is then coated with a conformal layer of TiO2. This heterojunction shows an unusual electron transfer process, where photogenerated electrons move from the WO3 layer into TiO2. State‐of‐the‐art hybrid density functional theory and hard X‐ray photoelectron spectroscopy are used to elucidate the electronic interaction at the WO3/TiO2 interface. Transient absorption spectroscopy shows that recombination is substantially reduced, extending both the lifetime and population of photogenerated charges into timescales relevant to most photocatalytic processes. This increases the photocatalytic efficiency of the material, which is among the highest ever reported for a thin film. In allying computational and experimental methods, this is believed to be an ideal strategy for determining the band alignment in metal oxide heterojunction systems.  相似文献

Toxicological investigation of suspected cocaine-related deaths routinely involves the identification of cocaine (COC) and its metabolites including benzoylecgonine (BE) and ecgonine methyl ester (EME) in postmortem specimens. We utilized solid-phase extraction followed by gas chromatography/mass spectrometry for the qualitative and quantitative analysis of cocaine and eight cocaine-related analytes. These analytes included anhydroecgonine methyl ester (AEME), a unique product formed during cocaine smoking, and cocaethylene (CE), formed by transesterification of cocaine in the presence of ethanol. Thirteen pairs of postmortem heart blood and urine specimens were analyzed from cases of death due to acute cocaine intoxication, multiple drug intoxication, or other non-drug related causes. COC, EME, and BE were detected in all specimens. The range of concentrations in blood were: COC, 23-2088 ng/mL; BE, 215-9195 ng/mL; and EME, 220-7275 ng/ mL. AEME was identified in 2 blood and 10 urine specimens, and CE was identified in 1 blood specimen and 4 urine specimens. The identification of AEME in the specimens indicated that "crack" cocaine had been smoked, and the presence of CE indicated co-administration of cocaine and ethanol. The presence of these unique cocaine analysis in postmortem specimens provides valuable information regarding the cause and manner of death.  相似文献

BACKGROUND: Prevention efforts to reduce the morbidity and mortality disparity between blacks and whites in the United States present a national health priority. However, participation of blacks in prevention trials has been low. The present study reports successful recruitment processes from two economically diverse black populations. METHODS: The two studies were independent projects, but both were part of a nationally funded collaboration on cardiovascular health. "Hip Hop to Health" is a 4-year randomized controlled trial aimed at dietary fat reduction and increased exercise among inner-city black families. The Fat Reduction Intervention Trial in African-Americans project is a 5-year trial to reduce total fat, saturated fat, and cholesterol intake among black families from a working class community. RESULTS: The two populations differed on demographic characteristics. Direct presentation was the most effective recruitment strategy in the "Hip Hop to Health" program. In contrast, telephone recruitment and neighborhood canvassing were the more successful strategies for FRITAA. CONCLUSIONS: Although both populations were black and at comparable cardiovascular disease risk, the differing demographics between the groups made different recruitment strategies necessary. This study documented the labor-intensive quality of successful recruitment, and results suggest that successful recruitment requires strategies tailored to the needs, experiences, and environment of the target group.  相似文献

OBJECTIVE: The spread of nosocomial multiresistant microorganisms is affected by compliance with infection control measures and antibiotic use. We hypothesized that "colonization pressure" (ie, the proportion of other patients colonized) also is an important variable. We studied the effect of colonization pressure, compliance with infection control measures, antibiotic use, and other previously identified risk factors on acquisition of colonization with vancomycin-resistant enterococci (VRE). METHODS: Rectal colonization was studied daily for 19 weeks in 181 consecutive patients who were admitted to a single medical intensive care unit. A statistical model was created using a Cox proportional hazards regression model including length of stay in the medical intensive care unit until acquisition of VRE, colonization pressure, personnel compliance with infection control measures (hand washing and glove use), APACHE (Acute Physiology and Chronic Health Evaluation) 11 scores, and the proportion of days that a patient received vancomycin or third-generation cephalosporins, sucralfate, and enteral feeding. RESULTS: With survival until colonization with VRE as the end point, colonization pressure was the most important variable affecting acquisition of VRE (hazard ratio [HR], 1.032; 95% confidence interval [C1], 1.012-1.052; P=.002). In addition, enteral feeding was associated with acquisition of VRE (HR, 1.009; 95% CI, 1.000-1.017; P=.05), and there was a trend toward association of third-generation cephalosporin use with acquisition (HR, 1.007; 95% CI, 0.999-1.015; P=.11). The effects of enteral feeding and third-generation cephalosporin use were more important when colonization pressure was less than 50%. Once colonization pressure was 50% or higher, these other variables hardly affected acquisition of VRE. CONCLUSIONS: Acquisition of VRE was affected by colonization pressure, the use of antibiotics, and the use of enteral feeding. However, once colonization pressure was high, it became the major variable affecting acquisition of VRE.  相似文献

Surface‐enhanced Raman spectroscopy is a powerful and sensitive analytical tool that has found application in chemical and biomolecule analysis and environmental monitoring. Since its discovery in the early 1970s, a variety of materials ranging from noble metals to nanostructured materials have been employed as surface enhanced Raman scattering (SERS) substrates. In recent years, 2D inorganic materials have found wide use in the development of SERS‐based chemical sensors owing to their unique thickness dependent physico‐chemical properties with enhanced chemical‐based charge‐transfer processes. Here, recent advances in the application of various 2D inorganic nanomaterials, including graphene, boron nitride, semiconducting metal oxides, and transition metal chalcogenides, in chemical detection via SERS are presented. The background of the SERS concept, including its basic theory and sensing mechanism, along with the salient features of different nanomaterials used as substrates in SERS, extending from monometallic nanoparticles to nanometal oxides, is comprehensively discussed. The importance of 2D inorganic nanomaterials in SERS enhancement, along with their application toward chemical detection, is explained in detail with suitable examples and illustrations. In conclusion, some guidelines are presented for the development of this promising field in the future.  相似文献

The content and distribution of amathamide alkaloids within single colonies of the bryozoanAmathia wilsoni (Ctenostomata) varied depending on the location in the colony. Three colonies in all, collected from the same site at the same time, were analyzed and gave very similar results. The outermost, more exposed, tips of the colony had an alkaloid content of nearly 9% of dry weight, while basal parts were apparently devoid of alkaloids. Samples taken midway between tips and base yielded intermediate concentrations of about 1%. Very little variation in the proportions of individual amathamides A, B, C, E occurred between exposed tips of the colonies. However, some differences in ratios were found between tips from exposed and more protected regions.  相似文献
