Two-dimensional map of Haemophilus influenzae following protein enrichment by heparin chromatography

Two-dimensional gel electrophoresis separates several hundred protein molecules in one single experiment and is efficiently used to study the products expressed by different genomes. Low-copy-number gene products are invisible on a stained two-dimensional map and must be enriched such that sufficient amounts are present for visualization and identification. We investigated the enrichment of proteins of the bacterium Haemophilus influenzae by chromatography on immobilized heparin which has affinity for growth and protein biosynthesis factors. Total soluble proteins of the microorganism were fractionated on Heparin-Actigel which resulted in enrichment of approximately 160 proteins. The eluates, representing about 40% of the applied proteins, were analyzed by two-dimensional gel electrophoresis and the protein spots were characterized by amino acid composition analysis and matrix-assisted laser desorption ionization mass spectrometry. The proteins enriched by chromatography on the heparin gel were not exclusively low-copy-number gene products and they did not exclusively belong to one single class of proteins. The proteins that bound to the heparin gel are indicated in a two-dimensional protein map which includes more than 110 newly identified proteins.     

Solder engineering for optoelectronic packaging

In addition to providing electrical connections, solder is useful in the formation of passive, precision alignments for optoelectronic packaging. Different designs have demonstrated micrometer-level alignment accuracies, and the aligned structures are becoming more and more complex. Solder engineering has been successfully introduced into the field of optoelectronics, and it is expected to be applied to many other areas demanding low-cost, precision alignments.     

Membranous duodenal stenosis

The experience of our 16 patients treated for membranous duodenal stenosis is reported. Their treatment and course was analysed in a retrospective study. Eight patients were operated on within the first 16 days of life and in the remaining group surgery was performed at 1 month to 4 y of age. The presenting symptom leading to diagnosis was, in all but one case, non-bile-stained vomiting. Associated malformations were found in all but four patients, mostly morbus Down. The operative procedure performed was a partial excision of the duodenal membrane and a duodenoplasty in 10 patients, a duodenojejunostomy in 5 patients, and a duodenoplasty only in 1 patient. The postoperative course was without lethal complications. One late stricture in an anastomosis occurred. We conclude that in its presentation, duodenal stenosis differs from duodenal atresia, and can often be misinterpreted, resulting in a late diagnosis, and should be reported as a separate entity.     

A simple determination of steroidal alkaloid glycosides by thin-layer chromatography immunostaining using monoclonal antibody against solamargine

A method of determination for solasodine glycosides by using thin-layer chromatography (TLC) immunostaining was investigated. Solasodine glycosides separated by silica gel TLC were transferred to a polyvinylidene difluoride membrane. The membrane was treated with sodium periodate solution followed by bovine serum albumin (BSA), resulting in a solasodine glycoside-BSA conjugate. Conjugation was confirmed by matrix-assisted laser desorption/ionization mass spectrometry. Individual spots were stained by monoclonal antibody against solamargine. Immunostaining of solasodine glycosides was more sensitive compared to other stainings. The newly established immunostaining method can be extended to analysis of the distribution of solasodine glycoside in the plant body.     

Mechanical stress evolution and the blech length: 2D simulation of early electromigration effects

Modeling of stress and electromigration at the microscopic level, in confined interconnect metallic lines with tungsten studs, can very well account for the resistance behaviour in time. The resistance change at saturation for a metallic line with blocking boundaries at both ends can be related, according to the model, to threshold product (jL)_c found by Blech [1].     

Effects of substrate temperature and texturing on the magnetic properties and crystallographic structures of CoCrTa/Cr thin film

Bi-layer CoCrTa/Cr films were deposited on textured aluminium or textured NiP-plated aluminium substrate by d.c. magnetron sputtering. The crystal anisotropy and thereby magnetic properties depending on substrate material, substrate temperature and texturing, were investigated. The magnetic crystal anisotropy induced by the mechanical texture on aluminium or NiP/Al substrates along the texture lines for the film deposited at high temperature, were clearly observed, while the film deposited at low temperature shows less prominent anisotropic behaviour. X-ray diffraction analysis indicates a change in the preferred orientation of the chromium and CoCrTa films sputtered on different substrates at different temperatures. It was found that a high substrate temperature was beneficial to the formation of Cr(002) and therefore epitaxial growth of Co(11¯20) on Cr(002) for either aluminium or NiP/Al substrates.     

Laterality, alternation, and perseveration relationships on the T-maze test.

The T-maze test has been used to study several entirely different issues: spontaneous alternation behavior (SAB), perseveration behavior (PB), and behavioral lateralization. Despite the fact that in this test the behavior studied is always the same one (i.e., side choice), the possible relationships among SAB, PB, and lateralization have not been previously evaluated. The present study investigated the relationships among these functions. The results demonstrated that (1) shock increases PB and lateralization decreases SAB, (2) practice increases lateralization and decreases SAB but does not modify PB, and (3) there are sex differences for alternation and SAB. Because these functions are expressed by the same behavioral pattern, they must be quantified simultaneously to avoid mistaken conclusions when the T-maze test is used.
