BACKGROUND: In atherosclerosis, both reductions and elevations in plasma levels of antioxidants have been reported. This study investigated total antioxidant capacity of plasma from subjects with atherosclerotic disease. MATERIALS AND METHODS: The study population consisted of 48 men with or without carotid atherosclerosis. At baseline (1990) carotid arteries were evaluated by duplex sonography and plasma samples were obtained for testing antioxidant capacity by two different test systems. One assay system used neutrophils from healthy volunteers as a source of oxygen free radicals activating the non-fluorescent dichlorofluorescin diacetate in the presence of antioxidant containing plasma from study subjects. In the other test system, total plasma antioxidants were detected colorimetrically by using 2,2'-azino-di-(3-ethylbenzthiazoline sulphonate), metmyoglobin and superoxide in the presence of plasma. Carotid arteries were re-evaluated for the development of new plaques 5 years later (1995). RESULTS: Increased baseline total antioxidant capacity of plasma was significantly associated with the development of new atherosclerotic lesions during a period of 5 years. CONCLUSIONS: Endogenous antioxidant capacity of plasma is increased in patients with active atherosclerotic disease. As scavenging of oxygen free radicals is thought to protect from atherogenesis, elevated antioxidative capacity may represent an adaptive mechanism.  相似文献

Since pneumotachograph masks are commonly used in studies of speech breathing, the purpose of this study was to measure the differences in respiratory volumetric and frequency measures during speech under two conditions: with and without a circumferentially vented pneumotachograph mask coupled to the face. Thus we sought to identify whether changes in breathing patterns occur with the use of a specific face mask, because these patterns are accepted as representative of normal speech breathing. Subjects were 10 normal-speaking women, each of whom produced a syllable train and a connected speech task, both at comfortable intensity levels. Respiratory measures were made using linearized magnetometers during speech production. The measurements included lung volume, rib cage volume, and abdominal volume at utterance initiation and termination, volume excursions during the utterance, and the number of breath groups during the speech task. There were no significant differences between the mask-on and mask-off conditions in volumetric and frequency measures. A significant task difference for abdominal initiation was found. It was concluded that the use of a circumferentially vented pneumotachograph mask does not alter the reliability of respiratory volume and frequency measures for studies of voice.  相似文献

Cyclin D1 is part of a cell cycle control node consistently deregulated in most human cancers. However, studies with cyclin D1-null mice indicate that it is dispensable for normal mouse development as well as cell growth in culture. Here, we provide evidence that ras-mediated tumorigenesis depends on signaling pathways that act preferentially through cyclin D1. Cyclin D1 expression and the activity of its associated kinase are up-regulated in keratinocytes in response to oncogenic ras. Furthermore, cyclin D1 deficiency results in up to an 80% decrease in the development of squamous tumors generated through either grafting of retroviral ras-transduced keratinocytes, phorbol ester treatment of ras transgenic mice, or two-stage carcinogenesis.  相似文献

A case of clofazimine enteropathy is described. A young male received clofazimine 200 mg daily for four years. He was admitted in a pigmented, emaciated state with abdominal pain, diarrhoea and weight loss. At laparotomy his abdominal organs were stained with dark brown-black pigment due to heavy infiltration with clofazimine crystals. Despite withdrawal of clofazimine his symptoms failed to settle. He developed oedema and hypoalbuminaemia. He died following a cerebral infarction.  相似文献

As with other forms of stress, surgery is a time of fluctuating haemodynamics, physiological shifts, intense metabolic changes, and protein catabolism and anabolism. At times, these changes can occur within hours. There is a reasonable body of literature concerning the adverse effects of surgery on gastric emptying, but not on the effects of surgery on the distribution, metabolism and excretion of drugs. Gastric emptying is diminished or absent following major procedures, which prevent the delivery of orally administered drugs to their major site of absorption, the small bowel. Changes in the paracellular absorption of drugs may occur postoperatively, although transcellular absorption appears to be unimpaired. Distribution is affected by changes in blood volume, alterations in circulation, increases in the extracellular fluid and changes in the circulating plasma protein levels, such as albumin and alpha 1-acid glycoprotein. Little is known about alterations in drug metabolism following surgery. However, it has been shown that systemic hypoxia alters the function of some of the cytochrome P450 (CYP) system and increased levels of cytokines have an effect on the metabolism of at least 1 drug. In addition, the renal elimination of drugs is affected in patients postoperatively, although the effects of biliary clearance in this period are difficult to determine. Despite the lack of research into pharmacokinetics during the postoperative period, and given the immense and often sudden changes observed in patients post-surgery, it is reasonable to recommend vigilance with respect to drug therapy during this period.  相似文献

The dynamics of superparamagnetic particles subject to competing magnetic and viscous drag forces have been examined with a uniform, stationary, external magnetic field. In this approach, competing drag and magnetic forces were created in a fluid suspension of superparamagnetic particles that was confined in a capillary tube; competing viscous drag and magnetic forces were established by rotating the tube. A critical Mason number was determined for conditions under which the rotation of the capillary prevents the formation of chains from individual particles. The statistics of chain length was investigated by image analysis while varying parameters such as the rotation speed and the viscosity of the liquid. The measurements showed that the rate of particle chain formation was decreased with increased viscosity and rotation speed; the particle dynamics could be quantified by the same dimensionless Mason number that has been demonstrated for rotating magnetic fields. The potential for enhancement of mixing in a bioassay was assessed using a fast chemical reaction that was diffusion-limited. Reducing the Mason number below the critical value, so that chains were formed in the fluid, gave rise to a modest improvement in the time to completion of the reaction.  相似文献

Summary The hydrolytic degradation of Kevlar 49 fibers when dissolved in H2SO4 is characterized from (i) Fourier transform infrared (FTIR), CNH analyses and optical microscopy studies of insoluble crystals formed in the PPTA-H2SO4 dope and (ii) the rates of H2SO4-catalyzed hydrolysis of model compound aromatic amides. The living carbocationic polymerization (LC Pzn) of p-methylstyrene (pMeSt) was achieved by the use of the 2-chloro-2,4,4-trimethylpentane (TMPCl)/TiCl4 initiating system in the presence of triethylamine (Et3N) electron donor in CH3Cl/nC6H14 mixed solvent at-80°C. Next, the synthesis, characterization, and some physical-mechanical characteristics of a series of poly (p-methylstyrene-b-isobutylene-b-p-methylstyrene) (PpMeSt-PIB-PpMeSt) triblocks is described. The latter synthesis involved two steps in one reactor: 1) The generation of diliving polyisobutylene dications (PIB) by the dicumyl chloride (DiCumCl)/TiCl4 system under similar conditions followed by 2) The addition of pMeSt and growth of living PpMeSt cations. Hydrogeologic parameters evaluated for rocks investigated in deep well testing projects for nuclear waste repository feasibility are transmissivity, hydraulic conductivity and storativity. Such studies have been carried out in rock formations of different geologic types, origins and ages for nuclear waste management agencies in North America and Europe. From transient pressure testing, an assessment of static pressure in selected fracture zones was measured from which a profile with depth was developed using modelling techniques. As well, temperatures of formation fluids and hence thermal gradients have also been provided. Hydrogeologic parameters are commonly used in risk analysis for repositories with respect to the possible escape of contaminants and worst case scenarios. The parameters are related to the rate of possible radionuclide migration into the biosphere. The techniques of the investigation and analysis methods of this work have been fully described in the literature. This paper is the first to date, however, to compare results from testing in differen geologic environments. High transmissivity zones have been observed in the upper few hundred metres of granite formations. Below that depth, the rock is more competent and discrete zones of higher permeability occur less frequently. Our studies show, however, that in these rocks, zones of high hydraulic conductivity do not always correspond to zones of high fracture frequency. Most formation pressure profiles have a vertical gradient of about 10 kPa/m. In discrete zones, deviations from this trend can occur due to vertical inhomogeneity in the rock. Thermal gradients vary considerably from one investigation site to another. For example, in the Canadian Shield, gradients are of the order of 1.0 degree C per 100 m, whereas in similar rock types in Europe, the gradient is up to 4.0 degrees C per 100 m.  相似文献

The rheological behavior of sulfonated polyisobutylene based elastomeric ionomers has been studied. The effects of molecular architecture, type of cation, and addition of excess neutralization agent were investigated. The effect of temperature was studied to a limited extent. In a specific case, the influence of an ionic plasticizer, zinc stearate was also examined. It was found that in these telechelic ionomers where the ionic groups are located exclusively at the chain ends, significant Ionic interactions may persist even at 180°C. The zinc-neutralized ionomers had the lowest viscosity as compared to the corresponding potassium- or calcium-neutralized ionomers. The covalent character of zinc is believed responsible for this behavior. Other factors being constant, the triarm based ionomers are more viscous than the monofunctional ionomers. A mixture of monofunctional ionomers with the triarm, species is a model for dangling chain ends, and results in a slight lowering of the viscosity under the conditions studied. Zinc stearate acts as an ionic plasticizer. Upon the addition of 15 percent by weight of zinc-stearate to the ionomer, the low shear rate viscosity drops by several orders of magnitude and renders the ionomer thermally processable at moderate temperatures.  相似文献
