Cows' and donkey milks (raw and thermally processed) and respective whey were analysed for quantification of major proteins. Two different chromatographic approaches, size exclusion (SE-HPLC) and reversed-phase high performance liquid chromatography (RP-HPLC) both coupled to UV detection were used. Usefulness of these methods for routine control of the effect of thermal processing was evaluated. The external standard method was used to calibrate the SE-HPLC and RP-HPLC systems. Concerning quantification of β-lactoglobulin (β-lg), α-lactalbumin (α-la), lysozyme (lys), and total casein (cn), no significant differences between results obtained by SE-HPLC and by RP-HPLC (t-test, P>0·05) were observed for raw milks and whey. Heating of cows' milk promoted aggregation of denatured proteins as observed by SE-HPLC, whereas α-la and β-lg from donkey milk were stable to thermal processing at 100 °C (5 min). Lys was quantified in donkey raw milk and whey however, in thermally processed donkey milk lys was denatured and could not be quantified by HPLC.  相似文献

The influence of high-pressure homogenisation (HPH) at 100 MPa on Lacticaseibacillus paracasei 90 (L90) was studied in vitro and in situ (fermented milk and cheese). Lysis and injury signs were observed in cells treated with HPH, which were linked to the release of intracellular enzymes and changes in the metabolic activity of L90. HPH treatment led to a higher lactic acid content and lower pH and pyruvic acid levels in fermented milk. The microbiological and peptide profiles of cheeses were modified by using cells or cell-free extracts of L90. Finally, this study suggests that L90 could release bioactive peptides.  相似文献

We report on low-temperature transport measurements on single and double quantum dots defined using local gates to electrostatically deplete InAs nanowires grown by chemical beam epitaxy. The diffusion approximation of the radiative transfer equation is a model used widely to describe photon migration in highly diffusing media and is an important matter in biological tissue optics. An analysis of the time-dependent diffusion equation together with its solutions for the slab geometry and for a semi-infinite diffusing medium are reported. These solutions, presented for both the time-dependent and the continuous wave source, account for the refractive index mismatch between the turbid medium and the surrounding medium. The results have been compared with those obtained when different boundary conditions were assumed. The comparison has shown that the effect of the refractive index mismatch cannot be disregarded. This effect is particularly important for the transmittance. The discussion of results also provides an analysis of the role of the absorption coefficient in the expression of the diffusion coefficient.  相似文献

In our companion paper we presented a model to describe photon migration through a diffusing slab. The model, developed for a homogeneous slab, is based on the diffusion approximation and is able to take into account reflection at the boundaries resulting from the refractive index mismatch. In this paper the predictions of the model are compared with solutions of the radiative transfer equation obtained by Monte Carlo simulations in order to determine the applicability limits of the approximated theory in different physical conditions. A fitting procedure, carried out with the optical properties as fitting parameters, is used to check the application of the model to the inverse problem. The results show that significant errors can be made if the effect of the refractive index mismatch is not properly taken into account. Errors are more important when measurements of transmittance are used. The effects of using a receiver with a limited angular field of view and the angular distribution of the radiation that emerges from the slab have also been investigated.  相似文献

Furthering G. W. Allport's (1954) contentions for optimal contact, the authors introduce a new construct: the perceived importance of contact. They propose that perceived importance is the best proximal predictor of contact's reduction of prejudice. If individuals have opportunities for contact at work or in the neighborhood, their chances to have intergroup acquaintances and friends increase. Intergroup contact among acquaintances and friends can be perceived as more or less important, which in turn determines intergroup evaluations. A 1st study shows that the new measure of perceived importance is indeed distinct from established quantity and quality indicators. The results are cross-validated in a 2nd study that also sheds light on the meaning of importance. In 3rd and 4th studies, structural equation analyses and a meta-analysis support the hypotheses. The simultaneous consumption of different classes of phytochemical antioxidants in the diet can result in more beneficial effects than when consumed alone. In the present study, the in vitro and in vivo antioxidative effects of espresso coffee brew (EC) (rich in chlorogenic acids) with added crude hazelnut skin phenolic extract (HSPE) from hazelnut skin waste (rich in flavonoids) were studied. Both post-brewing and pre-brewing phenolic-enriched espresso coffees (PE-ECs) were analysed for total phenols and screened for their in vitro antiradical ability. Moreover, the in vivo biological effect on the antioxidant potential of plasma in rats was evaluated. The PE-ECs showed increased both in vitro and in vivo antiradical activity proportional to the added HSPE. The in vivo experiments suggested that HSPE was much more antioxidant active than the phenolic fraction naturally contained in EC. Moreover, evidence of possible synergic effects of EC and HSPE phenolics was observed in vivo.  相似文献

The common assumption that chloride (Cl-) is conservative in soils and can be used as a groundwater tracer is currently being questioned, and an increasing number of studies indicate that Cl- can be retained in soils. We performed lysimeter experiments with soil from a coniferous forest in southeast Sweden to determine whether pore water residence time and nitrogen and Cl- loads affected Cl- retention. Over the first 42 days there was a net retention of Cl- with retention rates averaging 3.1 mg CI- m(-2) d(-1) (68% of the added Cl- retained over 42 days). Thereafter, a net release of Cl- at similar rates was observed for the remaining experimental period (85 d). Longer soil water residence time and higher Cl- load gave higher initial retention and subsequent release rates than shorter residence time and lower Cl- load did. Nitrogen load did not affect Cl transformation rates. This study indicates that simultaneous retention and release of Cl- can occur in soils, and that rates may be considerable relative to the load. The retention of Cl- observed was probably due to chlorination of soil organic matter or ion exchange. The cause of the shift between net retention and net release is unclear, but we hypothesize that the presence of O2 or the presence of microbially available organic matter regulates Cl- retention and release rates.  相似文献

Aflatoxin B1 (AFB1) and fumonisin B1 (FB1) are mycotoxins that often co-occur in feedstuffs. The ingestion of AFB1 causes aflatoxicosis in humans and animals. Sodium bentonite (NaB), a cheap non-nutritive unselective sequestering agent incorporated in animal diets, can effectively prevent aflatoxicosis. Fumonisins are responsible for equine leukoencephalomalacia and porcine pulmonary oedema, and often have subclinical toxic effects in poultries. Fumonisin B1 and aflatoxin B1 are both strongly adsorbed in vitro on sodium bentonite. Co-adsorption studies, carried out with a weight ratio of FB1 to AFB1 that mimics the natural occurrence (200:1), showed that FB1 greatly decreases the in vitro ability of NaB to adsorb AFB1. The ability of two activated carbons to adsorb FB1 was also investigated. Both carbons showed high affinity for FB1. A complex behaviour of the FB1 adsorption isotherms with pH was observed. In vitro results suggest that under natural contamination levels of AFB1 and FB1, a mixture of activated carbon and sodium bentonite might be potentially useful for prevention of sub-acute aflatoxicosis.  相似文献

The encapsulation of enterocins synthesized by Enterococcus faecium CRL1385 through ionic gelation with calcium ions was analyzed. Different enterocins samples were lyophilised and encapsulated using low-methoxyl pectin as the coating material. Lipids present in milk butter were also added to control the release of antimicrobial peptides from the capsules. The morphology of fresh and freeze-dried capsules was examined using light microscopy and scanning electron microscopy, respectively. Antimicrobial activity of encapsulated bacteriocins was assessed against Listeria monocytogenes 01/155 using the agar diffusion technique and direct contact in microplates. The capsules with higher lipid content showed a more spherical and uniform shape. Pathogen inhibition was observed for capsules prepared with different bacteriocin solutions both on solid (halo diameter = 8.5–13.5 mm) and in an aqueous medium (ca. 2 log orders decline in L. monocytogenes viability). The outcomes suggest that bacteriocin encapsulation through ionic gelation can be a potential alternative for the application of these antimicrobial peptides as biopreservatives in food.  相似文献
