Concentration and retention of a rod-shaped virus during tangential flow ultrafiltration (UF) was assessed to evaluate the potential of membrane-based downstream methods with advantages such as easy scale-up for industrial processes. A recombinant baculovirus of the non-spherical Autographa californica M nucleopolyhedrovirus (AcMNPV), vHSGFP, expressing egfp was filtered using polyethersulfone membranes ranging from 30 to 1000 kDa molecular weight cut-off (MWCO). A 20-fold virus concentration was achieved when a membrane cut-off range of 100 to 1000 kDa was tested. Fouling was observed and cake formation and pore plugging were postulated as concurrent causes with different impact depending on the MWCO. A reduction of virus concentration in the range of 2 to 5 log units in the permeate was observed illustrating the potential of membrane-based virus filtration as a useful unit operation in downstream processing. 相似文献

A procedure for the recovery of aromatic extracts from raw grape-spirit (alcohol strength 73 vol.%) by means of a countercurrent supercritical fluid extraction (CC-SFE) on a pilot plant scale is presented. The effect of the solvent-to-feed ratio at 7, 8, 10, and 20 kgCO2/kgFeed on the extraction of flavor compounds, operating at constant pressure of 100 bar and extraction temperature of 50 °C have been studied. Increasing the solvent-to-feed ratio from 7 to 20 kgCO2/kgFeed, the total volatile compounds yield decreases from 64 to 5% and that of ethanol from 85 to 49%. The highest concentration of volatile compounds has been obtained at 7 and 8 kgCO2/kgFeed solvent-to-feed ratio. Mixtures containing grape-spirit (alcohol strength 40 vol.%) added with 30% (v/v) of different extracts have been tested and judged by a panel. Sensory evaluation indicated as preferred the mixtures added with the extracts obtained at lowest solvent-to-feed ratio. 相似文献

Bakery products such as biscuits, cookies, and pastries represent a good medium for iron fortification in food products, since they are consumed by a large proportion of the population at risk of developing iron deficiency anemia, mainly children. The drawback, however, is that iron fortification can promote oxidation. To assess the extent of this, palm oil added with heme iron and different antioxidants was used as a model for evaluating the oxidative stability of some bakery products, such as baked goods containing chocolate. The palm oil samples were heated at 220°C for 10 min to mimic the conditions found during a typical baking processing. The selected antioxidants were a free radical scavenger (tocopherol extract (TE), 0 and 500 mg/kg), an oxygen scavenger (ascorbyl palmitate (AP), 0 and 500 mg/kg), and a chelating agent (citric acid (CA), 0 and 300 mg/kg). These antioxidants were combined using a factorial design and were compared to a control sample, which was not supplemented with antioxidants. Primary (peroxide value and lipid hydroperoxide content) and secondary oxidation parameters (p‐anisidine value, p‐AnV) were monitored over a period of 200 days in storage at room temperature. The combination of AP and CA was the most effective treatment in delaying the onset of oxidation. TE was not effective in preventing oxidation. The p‐AnV did not increase during the storage period, indicating that this oxidation marker was not suitable for monitoring oxidation in this model. 相似文献

The objective of this study was to evaluate the production of hydrogen in a two-stage CSTR system – both reactors having the same volume – and compare its performance with a conventional one-stage process. The lab-scale two-stage and one-stage systems were operated at five pHs and five hydraulic retention time (HRTs). The maximum volumetric hydrogen productivity and yield obtained with the two-stage system were 5.8 mmol L−1 h−1 and 2.7 mol H2 mol glucose−1, respectively, at an HRT of 12 h and pH 5.5. Overall, the two-stage system showed, at steady state, a better performance that the one-stage system for all the evaluated pHs. However, a comparison between the one-stage system, operating at 6 h of HRT, and the first reactor of the two-stage system at the same HRT did not show any significant difference, highlighting the positive impact of having a two-stage process. The determination of the ratio between the experimental measured H2 in the gas phase and the theoretical H2 generated in the liquid phase (discrepancy factor) indicated that an important part of the hydrogen produced in the first reactor was transferred into the second reactor instead of being desorbed in the headspace. Therefore, the improving of hydrogen production in the two-stage system is rather attributed to the increased transfer of hydrogen from liquid to gas than an actual total hydrogen production increase. 相似文献

In this paper we present algorithms for solving some combinatorial problems on one-dimensional processor arrays in which data flows in only one direction through the array. The problems we consider are: ranking the elements in a chain of sizen, rooting a spanning tree withn vertices, and computing biconnected components of a connected graph withn vertices. We show that each of these problems can be solved using arrays of sizen in which the data enters at the first cell and flows through the array in only one direction until it leaves the last cell as output. We also show how the biconnectivity algorithm for the array yields a new sequential algorithm for computing biconnected components which uses onlyO(n) locations of random access memory. 相似文献

Novel antiviral nanotherapeutics, which may inactivate the virus and block it from entering host cells, represent an important challenge to face viral global health emergencies around the world. Using a combination of bioorthogonal copper-catalyzed 1,3-dipolar alkyne/azide cycloaddition (CuAAC) and photoinitiated thiol–ene coupling, monofunctional and bifunctional peptidodendrimer conjugates were obtained. The conjugates are biocompatible and demonstrate no toxicity to cells at biologically relevant concentrations. Furthermore, the orthogonal addition of multiple copies of two different antiviral peptides on the surface of a single dendrimer allowed the resulting bioconjugates to inhibit Herpes simplex virus type 1 at both the early and the late stages of the infection process. Multivariate calibration models based on data from mid‐infrared spectroscopy of biodiesel/diesel blends were obtained. The blends were prepared from diesel oil and esters of soybean oil, waste cooking oil, and hydrogenated vegetable oil in proportions ranging from 0 to 100 % biodiesel. The results showed that the multivariate regression models with interval partial least squares (iPLS), backward interval partial least squares (biPLS), and synergy interval partial least squares (siPLS) were able to determine the fractions of the infrared spectrum that contain the relevant information for estimating the values of physicochemical properties, flash point, specific gravity, and cetane number, which are used in quality control of the blends. In the best models, the values of determination coefficients were greater than 0.9500, proving their efficiency as an alternative to traditional analytical methods. 相似文献
