Recent research into the effects of mixtures of estrogenic chemicals has revealed the capacity for similarly acting chemicals to act in combination, according to the principles of concentration addition. This means that, collectively, they may pose a significant environmental risk, even when each component is present at a low and individually ineffective concentration. The aim of this study was to investigate the ecological significance of mixture effects at low-effect concentrations by assessing the combined effect of estrogenic chemicals on the reproductive performance of fish. Pairs of fathead minnows were exposed to five estrogenic chemicals. Endpoints analyzed included fecundity, the expression of male secondary sexual characteristics, somatic indices, and vitellogenin induction. In the first phase of the study, a concentration-response analysis was performed to investigate the relative sensitivity of these endpoints. In the second phase, mixture effects at low-effect concentrations were explored by exposing fish to each of the mixture components, individually and in combination. Data from these experiments provide evidence of mixture effects on fitness and fecundity, demonstrating the capacity for chemicals to act together to affect reproductive performance, even when each component is present belowthe threshold of detectable effects. This has important implications for hazard assessment and contributes to our understanding of mixture effects at increasing levels of biological complexity. 相似文献

The yeast Kluyveromyces marxianus produces a range of volatile molecules with applications as fragrances or flavours. The purpose of this study was to establish how nutritional conditions influence the production of these metabolites. Four strains were grown on synthetic media, using a variety of carbon and nitrogen sources and volatile metabolites analysed using gas chromatography–mass spectrometry (GC–MS). The nitrogen source had pronounced effects on metabolite production: levels of the fusel alcohols 2‐phenylethanol and isoamyl alcohol were highest when yeast extract was the nitrogen source, and ammonium had a strong repressing effect on production of 2‐phenylethyl acetate. In contrast, the nitrogen source did not affect production of isoamyl acetate or ethyl acetate, indicating that more than one alcohol acetyl transferase activity is present in K. marxianus. Production of all acetate esters was low when cells were growing on lactose (as opposed to glucose or fructose), with a lower intracellular pool of acetyl CoA being one explanation for this observation. Bioinformatic and phylogenetic analysis of the known yeast alcohol acetyl transferases ATF1 and ATF2 suggests that the ancestral protein Atf2p may not be involved in synthesis of volatile acetate esters in K. marxianus, and raises interesting questions as to what other genes encode this activity in non‐Saccharomyces yeasts. Identification of all the genes involved in ester synthesis will be important for development of the K. marxianus platform for flavour and fragrance production. Formation of bromochlorodibenzo-p-dioxins and furans from the high-temperature pyrolysis of a 2-chlorophenol/2-bromophenol mixture 总被引:1,自引:0,他引:1
The homogeneous, gas-phase pyrolytic thermal degradation of a 50:50 mixture of 2-bromophenol and 2-chlorophenol was studied in a 1 cm i.d., fused silica flow reactor at a total concentration of 88 ppm, reaction time of 2.0 s, and temperatures from 300 to 1000 degrees C. Observed products included (in decreasing yield) naphthalene, dibenzo-p-dioxin (DD), phenol, dibenzofuran (DF), bromobenzene, chloronaphthalene, 4-bromo-6-chlorodibenzofuran (4-B,6-CDF), bromonaphthalene, benzene, 4,6-dichlorodibenzofuran (4,6-DCDF), chlorobenzene, 4-monobromodibenzofuran (4-MBDF), 4-monochlorodibenzofuran (4-MCDF), 1-mono-bromodibenzo-p-dioxin (1-MBDD), 2-chloro,4-bromophenol, 2,4-dibromophenol, and 2-bromo-4-chlorophenol. Unlike the case for the pyrolysis of pure 2-chlorophenol, 4,6-DCDF was observed, but the analogous 4,6-DBDF remained undetected similar to the individual results with 2-MBP. This indicates that the presence of bromine increases the concentration of chlorine atoms available for the formation of 4,6-DCDF. Due to bromine atoms acting as better leaving groups than chlorine atoms, the yield of DD was increased over that observed for the pyrolysis of 2-chlorophenol. The addition of bromine to a chlorinated hydrocarbon system results in an increase in the total yield of PCDD/Fs as well as PBDD/Fs and mixed PBCDD/Fs due to the ease of bromine elimination reactions as well as an increase of the chlorine atom concentration. 相似文献

Effects of atropine on blood plasma amino acid profile and on the yields and concentration of milk components were investigated in 12 Friesian cows in early lactation. Cows were housed indoors and fed with cut pasture ad libitum. Each cow received four treatments over 12 d during a replicated 4 x 4 Latin square experiment. Treatments were: control (saline); low dose (L; 30 microg atropine/kg body weight (BW)); medium dose (M; 40 microg atropine/kg BW); and 2 x L dose, 2 h apart (2 x L). On each of four treatment days, cows were milked at about 7.00, after which treatments were administered by subcutaneous injection. Cows were milked again at 2 h, 6 h and 10 h after injection. Milk samples were collected at each milking. Immediately after the 2 h milking, blood samples were drawn from each cow and the second injection was given for the 2 x L treatment. Atropine reduced hourly milk yield, and concentrations and hourly yields of total protein, casein, whey protein, alpha-casein, beta-casein, kappa-casein, beta-lactoglobulin and alpha-lactalbumin, but by differing amounts. Milk concentrations of bovine serum albumin and immunoglobulin G were increased by atropine, and overall yields of these proteins were mostly unchanged. Atropine lowered concentrations of most, but not all, amino acids in blood plasma, with essential amino acids reduced more than non-essential amino acids. Concentrations of alpha-amino N in whole blood, and glucose and insulin in blood plasma, fell after atropine injection. There was no difference between the L and M doses of atropine, but the 2 x L dose had greater effects on milk composition than the single doses. For yields of milk and milk components, the effect of the 2 x L dose was also more persistent. The results highlight the differential synthesis of individual milk proteins, and suggest that atropine might be useful for evaluating the mechanisms regulating milk protein composition. 相似文献

Two different electronic nose systems (metal oxide and conducting polymer based) were used to identify Salmonella typhimurium contaminated beef strip loin samples (stored at two temperatures). The sensors present in the two systems were ranked based
on their Fisher criteria of ranking to evaluate their importance in discriminant analysis. The most informative sensors were
then used to develop linear discriminant analysis and quadratic discriminant analysis-based classification models. Further,
sensor signals collected from both the sensor systems were combined to improve the classification accuracies. The developed
models classified meat samples based on the Salmonella population into “No Salmonella” (microbial counts < 0.7 log10 cfu/g) and “Salmonella inoculated” (microbial counts ≥ 0.7 log10 cfu/g). The performances of the developed models were validated using leave-1-out cross-validation. Classification accuracies
of 80% and above were observed for the samples stored at 10 °C using the sensor fusion approach. However, the classification
accuracies were relatively low for the meat samples stored at 4 °C when compared to the samples stored at 10 °C. The results
indicate that the electronic nose systems could be effectively used as a first stage screening device to identify the meat

Colored dissolved organic material (CDOM) is an important sunlight absorbing substance affecting the optical properties of natural waters. However, little is known about its structural and optical properties mainly due to its complex matrix and the limitation of the techniques available. A comparison of two southwestern Florida rivers [the Caloosahatchee River (CR) and the Shark River (SR)] was done in terms of molecular mass (MM) and diffusion coefficients (D). The novel technique Frit inlet/frit outlet-flow field-flow fractionation (FIFO-FIFFF) with absorbance and fluorescence detectors was used to determine these properties. The SR receives organic material from the Everglades. By contrast, the CR arises from Lake Okeechobee in central Florida, receiving anthropogenic inputs, farming runoff, and natural organics. Both rivers discharge to the Gulf of Mexico. Fluorescence identified, for both rivers, two different MM distributions in low salinity water samples: the first was centered at approximately 1.7 kDa (CR) and approximately 2 kDa (SR); the second centered at approximately 13 kDa for both rivers, which disappeared gradually in the river plumes to below detection limit in coastal waters. Absorbance detected only one MM distribution centered at approximately 2 kDa (CR) and 2.2-2.4 kDa (SR). Fluorescence in general peaked at a lower MM than absorbance, suggesting a different size distribution for fluorophores vs chromophores. A photochemical study showed that, after sunlight, irradiated freshwater samples have similar characteristics to more marine waters, including a shift in MM distribution of chromophores. The differences observed between the rivers in the optical characteristics, MM distributions, and D values suggest that the CDOM sources, physical, and photochemical degradation processes are different for these two rivers. 相似文献

Online tutorials are useful for reinforcing information literacy concepts addressed in library instruction. A wide range of tutorial types cater to different learning styles and feature varying levels of interactivity. In this study, students and faculty reported experiences using two types of tutorials teaching basic search strategies—an interactive tutorial, and a video tutorial. Although both groups favored the video, faculty were split about which tutorial they would rather assign, a discrepancy suggesting questions about how tutorial developers should balance user preferences with learning needs. Laboratory geophysics tests including bender elements and acoustic emission measure the speed of propagation of stress or sound waves in granular materials to derive elastic stiffness parameters. This contribution builds on earlier studies to assess whether the received signal characteristics can provide additional information about either the material’s behaviour or the nature of the material itself. Specifically it considers the maximum frequency that the material can transmit; it also assesses whether there is a simple link between the spectrum of the received signal and the natural frequencies of the sample. Discrete element method (DEM) simulations of planar compression wave propagation were performed to generate the data for the study. Restricting consideration to uniform (monodisperse) spheres, the material fabric was varied by considering face-centred cubic lattice packings as well as random configurations with different packing densities. Supplemental analyses, in addition to the DEM simulations, were used to develop a more comprehensive understanding of the system dynamics. The assembly stiffness and mass matrices were extracted from the DEM model and these data were used in an eigenmode analysis that provided significant insight into the observed overall dynamic response. The close agreement of the wave velocities estimated using eigenmode analysis with the DEM results confirms that DEM wave propagation simulations can reliably be used to extract material stiffness data. The data show that increasing either stress or density allows higher frequencies to propagate through the media, but the low-pass wavelength is a function of packing density rather than stress level. Prior research which had hypothesised that there is a simple link between the spectrum of the received signal and the natural sample frequencies was not substantiated. 相似文献
