Quantum oscillations in the transverse magnetoresistance of aluminum single crystals were measured at 4.2 K in fields near the [001] direction of up to 50 kOe. The rotation diagrams around two axes, the current axis (-rotation) and normal to it (-rotation), were investigated by directly measuring the amplitude of oscillations in steps of about 0.1. As the field was tilted from [001] by about 2.5 in the plane, the separation of double maxima in the rotation diagram changed from 0.8 to 5.0 for the [010] current samples, and from 2.8° to 5.6° for the [110]current samples, and the amplitude of oscillation decreased. A fast oscillation, which had not been observed in conventional dc magnetoresistance measurements, was simultaneously detected near the field direction where the -orbit oscillations have a maximum amplitude.  相似文献

Object Most functional magnetic resonance imaging (fMRI) studies record the blood oxygen leveldependent (BOLD) signal using gradient-echo echo-planar imaging (GE EPI). EPI can suffer from substantial BOLD sensitivity loss caused by magnetic field inhomogeneities. Here, BOLD sensitivity losses due to susceptibility- induced gradients in the readout (RO) direction are characterized and a compensation approach is developed. Materials and Methods Based on a theory describing the dropout mechanism, an EPI sequence was optimized for maximal BOLD sensitivity in the orbitofrontal cortex (OFC) using a specific combination of an increased spatial resolution in the RO direction and a reduced echo time. Using measured BOLD sensitivity maps and a breath hold experiment, the model and compensation approach were tested. Results Using typical fMRI EPI parameters, susceptibility-induced gradients in the RO direction caused dropouts in the OFC and the inferior temporal lobe. Optimizing the echo time and spatial resolution effectively reduced the dropout as predicted by the theory. Conclusion The model-based compensation approach effectively reduces BOLD sensitivity losses due to susceptibility-induced gradients in the RO direction. It retains the high temporal resolution of single-shot EPI and can be readily combined with methods for the compensation of susceptibility-induced field gradients in the phase-encoding and through-plane direction.  相似文献

Land-use regression modeling was used to develop maps of annual average black smoke (BS) and sulfur dioxide (SO(2)) concentrations in 1962, 1971, 1981, and 1991 for Great Britain on a 1 km grid for use in epidemiological studies. Models were developed in a GIS using data on land cover, the road network, and population, summarized within circular buffers around air pollution monitoring sites, together with altitude and coordinates of monitoring sites to consider global trend surfaces. Models were developed against the log-normal (LN) concentration, yielding R(2) values of 0.68 (n = 534), 0.68 (n = 767), 0.41 (n = 771), and 0.39 (n = 155) for BS and 0.61 (n = 482), 0.65 (n = 733), 0.38 (n = 756), and 0.24 (n = 153) for SO(2) in 1962, 1971, 1981, and 1991, respectively. Model evaluation was undertaken using concentrations at an independent set of monitoring sites. For BS, values of R(2) were 0.56 (n = 133), 0.41 (n = 191), 0.38 (n = 193), and 0.34 (n = 37), and for SO(2) values of R(2) were 0.71 (n = 121), 0.57 (n = 183), 0.26 (n = 189), and 0.31 (n = 38) for 1962, 1971, 1981, and 1991, respectively. Models slightly underpredicted (fractional bias: 0~-0.1) monitored concentrations of both pollutants for all years. This is the first study to produce historic concentration maps at a national level going back to the 1960s.  相似文献

Pisum sativum is of great economic and nutritional interest due to its protein content. Nevertheless, pea products are underused as a protein source in human food because of their strong beany flavour. Therefore, the objective of this study was to select an efficient and representative method to extract volatile molecules of pea flour. In the first step, three extraction methods were chosen: solid phase micro extraction (SPME); Purge and Trap extraction and solvent assisted flavour evaporation (SAFE). The corresponding extracts were analysed by gas chromatography coupled with mass spectrometry. In the second step, the sensory representativity of the extracts was assessed either by direct gas chromatography-olfactometry for SPME and for Purge and Trap extracts, or by sniffing for the aqueous SAFE extract. It appeared that SAFE extraction was the most suitable method because of its good extraction capacities and its high sensory representativity of the global odour of pea flour.  相似文献

Therapeutic glucocorticoids (GCs) are powerful anti-inflammatory tools in the management of chronic inflammatory diseases such as rheumatoid arthritis (RA). However, their actions on bone in this context are complex. The enzyme 11β-hydroxysteroid dehydrogenase type 1 (11β-HSD1) is a mediator of the anti-inflammatory actions of therapeutic glucocorticoids (GCs) in vivo. In this study we delineate the role of 11β-HSD1 in the effects of GC on bone during inflammatory polyarthritis. Its function was assessed in bone biopsies from patients with RA and osteoarthritis, and in primary osteoblasts and osteoclasts. Bone metabolism was assessed in the TNF-tg model of polyarthritis treated with oral GC (corticosterone), in animals with global (TNF-tg11βKO), mesenchymal (including osteoblast) (TNF-tg11βflx/tw2cre) and myeloid (including osteoclast) (TNF-tg11βflx/LysMcre) deletion. Bone parameters were assessed by micro-CT, static histomorphometry and serum metabolism markers. We observed a marked increase in 11β-HSD1 activity in bone in RA relative to osteoarthritis bone, whilst the pro-inflammatory cytokine TNFα upregulated 11β-HSD1 within osteoblasts and osteoclasts. In osteoclasts, 11β-HSD1 mediated the suppression of bone resorption by GCs. Whilst corticosterone prevented the inflammatory loss of trabecular bone in TNF-tg animals, counterparts with global deletion of 11β-HSD1 were resistant to these protective actions, characterised by increased osteoclastic bone resorption. Targeted deletion of 11β-HSD1 within osteoclasts and myeloid derived cells partially reproduced the GC resistant phenotype. These data reveal the critical role of 11β-HSD1 within bone and osteoclasts in mediating the suppression of inflammatory bone loss in response to therapeutic GCs in chronic inflammatory disease.  相似文献

The marine cone snail produces one of the fastest prey strikes in the animal kingdom. It injects highly efficacious venom, often causing prey paralysis and death within seconds. Each snail has hundreds of conotoxins, which serve as a source for discovering and utilizing novel analgesic peptide therapeutics. In this study, we discovered, isolated, and synthesized a novel α3/5-conotoxins derived from the milked venom of Conus obscurus (α-conotoxin OI) and identified the presence of α-conotoxin SI-like sequence previously found in the venom of Conus striatus. Five synthetic analogs of the native α-conotoxin OI were generated. These analogs incorporated single residue or double residue mutations. Three synthetic post-translational modifications (PTMs) were synthetically incorporated into these analogs: N-terminal truncation, proline hydroxylation, and tryptophan bromination. The native α-conotoxin OI demonstrated nanomolar potency in Poecilia reticulata and Homo sapiens muscle-type nicotinic acetylcholine receptor (nAChR) isoforms. Moreover, the synthetic α-[P9K] conotoxin OI displayed enhanced potency in both bioassays, ranging from a 2.85 (LD50) to 18.4 (IC50) fold increase in comparative bioactivity. The successful incorporation of PTMs, with retention of both potency and nAChR isoform selectivity, ultimately pushes new boundaries of peptide bioengineering and the generation of novel α-conotoxin-like sequences.  相似文献

Intermittent rivers are dynamic ecosystems that experience a predictable or unpredictable loss of surface water and are characterised by changing lotic, lentic (ponding) and dry habitats. Plant communities colonising dry channels during the desiccation stage can be diverse, abundant and differ in their tolerances to water availability and habitat conditions. This study examines the colonisation of terrestrial vegetation in two intermittent rivers in the United Kingdom, and whether terrestrial plant taxonomic richness and functional diversity increase during the dry phase. Six reaches were surveyed for terrestrial plants during the dry phase over a standard 100 m length every month from April to October 2021. We found the channel and bank taxonomic richness increased with drying duration. Functional traits of vegetation height, clonality, clonality richness and Ellenberg's value of light moisture also increased with stream desiccation. Bed sediment conditions (the proportion of sand and gravel) and the 12-month antecedent percentage of zero flow days were the key drivers of plant community composition. We believe plant propagules from the riparian zone and channel vegetation on topographic high points in the channel aided plant colonisation of the riverbed once flow ceased. Past research may have underestimated the biodiversity value of intermittent rivers by failing to include the ecological importance of plants during the dry phase. Information on plant diversity of the dry phase is important to determine the overall biodiversity of intermittent rivers for their long-term conservation and management.  相似文献
