Accumulation of 16S rRNA and production of guanosine polyphosphates (pppGpp and ppGpp) were studied during amino acid starvation in three wild-type strains of Helicobacter pylori. All strains exhibit a relaxed phenotype with respect to accumulation of 16S rRNA. This constitutes the first example of a wild-type eubacterium showing a relaxed phenotype. The guanosine polyphosphate levels do not rise as a result of amino acid starvation, as expected for relaxed organisms. However, in both growing and starved cells, basal levels of the two polyphosphates appeared to be present, demonstrating that the enzymatic machinery for guanosine polyphosphate production is present in this organism. These findings are discussed within the framework of the hypothesis that stringent control is a physiological control mechanism more important for the fitness of prokaryotes growing in the general environment than for those that inhabit protected niches. 相似文献

An adaptive nonlinear control system for an inverter-fed induction motor is presented. The approach taken is fundamentally different from previous work on induction motor control in that the aim is to control shaft speed directly. Further, the inverter control variables, frequency and-voltage, are related via the steady-state model to the shaft speed and motor current. By making the controller adaptive, neither load nor machine parameters need be specified, although in practice the, latter are usually available from the machine manufacturer, and, of course, full use can be made of this knowledge. 相似文献

A challenge in engineering design is to choose suitable objectives and constraints from many quantities of interest, while ensuring an optimization is both meaningful and computationally tractable. We propose an optimization formulation that can take account of more quantities of interest than existing formulations, without reducing the tractability of the problem. This formulation searches for designs that are optimal with respect to a binary relation within the set of designs that are optimal with respect to another binary relation. We then propose a method of finding such designs in a single optimization by defining an overall ranking function to use in optimizers, reducing the cost required to solve this formulation. In a design under uncertainty problem, our method obtains the most robust design that is not stochastically dominated faster than a multiobjective optimization. In a car suspension design problem, our method obtains superior designs according to a k-optimality condition than previously suggested multiobjective approaches to this problem. In an airfoil design problem, our method obtains designs closer to the true lift/drag Pareto front using the same computational budget as a multiobjective optimization. 相似文献

Nucleic acid sequence-based amplification (NASBA) was applied to the detection of Salmonella enterica cells in liquid whole egg. In optimization under uncertainty for engineering design, the behavior of the system outputs due to uncertain inputs needs to be quantified at each optimization iteration, but this can be computationally expensive. Multifidelity techniques can significantly reduce the computational cost of Monte Carlo sampling methods for quantifying the effect of uncertain inputs, but existing multifidelity techniques in this context apply only to Monte Carlo estimators that can be expressed as a sample average, such as estimators of statistical moments. Information reuse is a particular multifidelity method that treats previous optimization iterations as lower fidelity models. This work generalizes information reuse to be applicable to quantities whose estimators are not sample averages. The extension makes use of bootstrapping to estimate the error of estimators and the covariance between estimators at different fidelities. Specifically, the horsetail matching metric and quantile function are considered as quantities whose estimators are not sample averages. In an optimization under uncertainty for an acoustic horn design problem, generalized information reuse demonstrated computational savings of over 60% compared with regular Monte Carlo sampling. 相似文献

A diode-laser-pumped neodymium-doped fiber laser is presented. For a launched pump power of 85 mW, the fiber laser had a cw output power of 43 mW, which is approximately an order of magnitude greater output power than any previously reported diode-pumped neodymium fiber laser operating on the(4)F(3/2)-(4)I(9/2) transition, which covers the 900-950-nm region. The fiber laser had a threshold power of 10 mW and a slope efficiency of 58% with respect to launched pump power. Tuning with a diffraction grating was obtained from 896 to 937 nm with narrow-band output powers as high as 32 mW. Emission was also obtained at 939.5 nm with use of a fiber Bragg grating as the output reflector. 相似文献

Broiler chicks were spray treated on the day of hatch with titrated dosages (10(6), 10(7), or 10(8) anaerobic CFU) of a characterized competitive exclusion culture (CF3) and challenged orally on day 3 with 10(4) CFU of Salmonella typhimurium. On day 10, cecal contents from control and CF3-treated chicks were cultured for S. typhimurium to determine the minimal efficacious dosage of the CF3 culture. The experiment was repeated in three replicated trials. Resistance to Salmonella cecal colonization was dosage related and progressively enhanced at the 10(7)- and 10(8)-CFU dosages compared with the 10(6)-CFU dosage. The 10(7)-CFU dosage was selected as the minimal effective dosage and evaluated for efficacy during a 43-day broiler growout study. Six hundred broilers were spray treated on the day of hatch and compared with 600 controls. One-half of the control and CF3-treated birds were challenged orally on day 3 with 10(4) CFU of S. typhimurium and designated "seeders." The remaining unchallenged birds were designated "contacts." Compared with the controls, the recovery of Salmonella cells from the ceca of the CF3-treated broilers was significantly decreased (P < 0.01) in the challenged seeders on days 21 and 43 of growout. Salmonella contamination of floor pen litter was significantly reduced (P < 0.05) in pens of CF3-treated birds compared with controls. The transmission of Salmonella cells from seeder to contact birds in the same pens was decreased significantly (P < 0.01). The results indicated that treatment of broiler chicks on the day of hatch with the 10(7)-CFU dosage of CF3 culture effectively increased resistance to S. typhimurium challenge during growout to market age. 相似文献

Due to the shift from software-as-a-product to software-as-a-service, software components that were developed to run in a
single address space must increasingly be accessed remotely across the network. Distribution middleware is frequently used
to facilitate this transition. Yet a range of middleware platforms exist, and there are few existing guidelines to help the
programmer to choose an appropriate middleware platform to achieve desired goals for performance, conciseness, intuitiveness,
and reliability. To address this limitation, in this article, we describe a case study of transitioning an Open Service Gateway
Initiative service from local to remote access. In our case study, we evaluate five remote versions of this service, constructed
using different distribution middleware platforms. These platforms are implemented by widely-used commercial technologies
or have been proposed as improvements on the state of the art. In particular, we implemented a service-oriented version of
our own Remote Batch Invocation abstraction. We compare and contrast these implementations in terms of their respective performance,
conciseness, complexity, and reliability. Our results can help remote service programmers make informed decisions when choosing

The quantification of carbon fluxes between the terrestrial biosphere and the atmosphere is of scientific importance and also relevant to climate-policy making. Eddy covariance flux towers provide continuous measurements of ecosystem-level exchange of carbon dioxide spanning diurnal, synoptic, seasonal, and interannual time scales. However, these measurements only represent the fluxes at the scale of the tower footprint. Here we used remotely sensed data from the Moderate Resolution Imaging Spectroradiometer (MODIS) to upscale gross primary productivity (GPP) data from eddy covariance flux towers to the continental scale. We first combined GPP and MODIS data for 42 AmeriFlux towers encompassing a wide range of ecosystem and climate types to develop a predictive GPP model using a regression tree approach. The predictive model was trained using observed GPP over the period 2000-2004, and was validated using observed GPP over the period 2005-2006 and leave-one-out cross-validation. Our model predicted GPP fairly well at the site level. We then used the model to estimate GPP for each 1 km × 1 km cell across the U.S. for each 8-day interval over the period from February 2000 to December 2006 using MODIS data. Our GPP estimates provide a spatially and temporally continuous measure of gross primary production for the U.S. that is a highly constrained by eddy covariance flux data. Our study demonstrated that our empirical approach is effective for upscaling eddy flux GPP data to the continental scale and producing continuous GPP estimates across multiple biomes. With these estimates, we then examined the patterns, magnitude, and interannual variability of GPP. We estimated a gross carbon uptake between 6.91 and 7.33 Pg C yr− 1 for the conterminous U.S. Drought, fires, and hurricanes reduced annual GPP at regional scales and could have a significant impact on the U.S. net ecosystem carbon exchange. The sources of the interannual variability of U.S. GPP were dominated by these extreme climate events and disturbances. 相似文献
