Low-grade chronic inflammation plays a pivotal role in the pathogenesis of insulin resistance (IR), and skeletal muscle has a central role in this condition. NLRP3 inflammasome activation pathways promote low-grade chronic inflammation in several tissues. However, a direct link between IR and NLRP3 inflammasome activation in skeletal muscle has not been reported. Here, we evaluated the NLRP3 inflammasome components and their role in GLUT4 translocation impairment in skeletal muscle during IR. Male C57BL/6J mice were fed with a normal control diet (NCD) or high-fat diet (HFD) for 8 weeks. The protein levels of NLRP3, ASC, caspase-1, gasdermin-D (GSDMD), and interleukin (IL)-1β were measured in both homogenized and isolated fibers from the flexor digitorum brevis (FDB) or soleus muscle. GLUT4 translocation was determined through GLUT4myc-eGFP electroporation of the FBD muscle. Our results, obtained using immunofluorescence, showed that adult skeletal muscle expresses the inflammasome components. In the FDB and soleus muscles, homogenates from HFD-fed mice, we found increased protein levels of NLRP3 and ASC, higher activation of caspase-1, and elevated IL-1β in its mature form, compared to NCD-fed mice. Moreover, GSDMD, a protein that mediates IL-1β secretion, was found to be increased in HFD-fed-mice muscles. Interestingly, MCC950, a specific pharmacological NLRP3 inflammasome inhibitor, promoted GLUT4 translocation in fibers isolated from the FDB muscle of NCD- and HFD-fed mice. In conclusion, we found increased NLRP3 inflammasome components in adult skeletal muscle of obese insulin-resistant animals, which might contribute to the low-grade chronic metabolic inflammation of skeletal muscle and IR development. 相似文献
The dried bean beetle, Acanthoscelides obtectus, is an economically important, worldwide pest of legume crops including dry beans, Phaseolus vulgaris. Assessment of A. obtectus infestation levels in pre-harvest field crops and post-harvest granaries is difficult to achieve because there is no effective monitoring tool for early detection so that interventions can be deployed as needed. Because A. obtectus is a generic pollen and nectar feeder, we adopted an electrophysiological (EAG) screening approach, using the antennae of female A. obtectus to identify physiologically active, volatile phytochemicals, which could then be investigated for their attractiveness to A. obtectus in laboratory behavioral assays and preliminary field tests. Of the 27 compounds tested in EAG screening, 5 compounds, i.e., methyl anthranilate, methyl eugenol, benzyl alcohol, (RS)-lavandulol, and 2-phenylethanol, elicited stronger EAG responses than the standard (1-phenylethanol). In 4-arm olfactometer bioassays, female A. obtectus preferred the olfactometer arm containing the odor of either methyl anthranilate or benzyl alcohol compared to the solvent control. In preliminary field tests using these 2 compounds as a binary mixture, at least 5 times as many beetles were caught on baited traps compared to non-baited traps. The field data also suggested that benzyl alcohol was primarily responsible for the field activity of the blend. We hypothesize that the attraction of A. obtectus to the combined benzyl alcohol/methyl anthranilate and the single benzyl alcohol baits is connected to the species` nectar- and pollen-feeding behaviour and not to its intraspecific communication. To our knowledge, this is the first evidence that A. obtectus behavior in the field can be modified by the deployment of plant-derived semiochemicals.
H-AITS-1 zeolite with Si/Ti = 50 and Si/Al = 50 was employed in preparing catalyst samples by ion-exchange and impregnation with a copper nitrate solution to obtain 0.24–1.15 wt.% and 1.5, 2 and 2.5 wt.% Cu loading, respectively. The catalytic properties for the NO decomposition were compared with that of Cu-ZSM-5 (Si/Al = 25 with 2 wt.% Cu loading) and similarity was found between the AITS-1 based samples and Cu-ZSM-5. Due to the higher acidity, the activity at 500°C per total copper atoms (an apparent turnover frequency, TOF) was significantly higher over Cu based AITS-1 samples being 2–3 × 10−3 s−1 as compared to 1 × 10−3 s−1 measured on Cu-ZSM-5. For the ion-exchanged Cu-AITS-1 there was an increase in TOF with increasing copper content, whereas on the impregnated samples a decrease in TOF was found. On all catalysts there was a maximum in the NO conversion at 500–550°C. The amount of NO per copper atom measured by temperature programmed desorption (TPD) was about the same as that on Cu-ZSM-5 and the features of the TPD were also similar. At the first contact of the catalyst at 500°C with the 2 vol% NO/Ar gas a transient N2O formation and a considerable delay in the O2 formation was observed. This could, however, be reproduced only on fresh catalyst, while all further transients showed different but reproducible features using the same sample. 相似文献
This paper investigates the development of loosening at flange joints owing to creep and relaxation of gaskets at medium high temperature. The model presented here is able to describe the interaction of gasket and flange in addition to the viscoelastic behaviour of a non-metallic gasket. The time-dependent material characteristics of the investigated gaskets were determined by standard tests whilst the reliability and preciseness of the derived equations are proved by test results. In this study, the performance characteristics of a flowing electrolyte-direct methanol fuel cell (FE-DMFC) and a direct methanol fuel cell (DMFC) are evaluated by computer simulations; and results are compared to experimental data found in the literature. Simulations are carried out to assess the effects of the operating parameters on the output parameters; namely, methanol concentration distribution, cell voltage, power density, and electrical efficiency of the cell. The operating parameters studied include the electrolyte flow rate, flowing electrolyte channel thickness, and methanol concentration at the feed stream. In addition, the effect of the circulation of the flowing electrolyte channel outlet stream on the performance is discussed. The results show that the maximum power densities that could be achieved do not significantly differ between these two fuel cells; however the electrical efficiency could be increased by 57% when FE-DMFC is used instead of DMFC. 相似文献

to mimic these processes. Mixtures of free amino acids, creatinine and glucose, simulating the composition of beef or chicken,
heated at 200 °C, form eight heterocyclic amines. Besides the commonly found 2-amino-3,8-dimethylimidazo[4,5-f]quinoxaline, 2-amino-3-methylimidazo[4,5-f]quinoline, 2-amino-3,4-dimethylimidazo[4,5-f]quinoline, 2-amino-3,4,8-trimethylimidazo[4,5-f]quinoxaline and 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine, 2-amino-1,6-dimethylimidazo[4,5-b]pyridine, 2-amino-1,5,6-trimethylimidazo[4,5-b]pyridine and 2-amino-1,6-dimethylfuro[3,2-e]imidazo[4,5-b]pyridine were also found. The calculated risk of consumption of heterocyclic amines is determined by the dietary dose, the
extrapolation of carcinogenic potencies from rodents to humans, and the extrapolation of high rodent doses to low human exposures.
Results suggest that DNA binding is linear with dose, but that the human DNA forms more adducts per unit dose than that of
the rat. Altogether, the risk appears to be equivalent to that for many carcinogens that are regulated.

The thermal response of a multi-tank thermal storage was studied under variable charge conditions. Tests were conducted on an experimental apparatus that simulated the thermal charging of the storage system by a solar collector over predetermined (prescribed) daylong periods. The storage was assembled from three standard 270 L hot-water storage tanks each charged through coupled, side-arm, natural convection heat exchangers which were connected in either a series- or parallel-flow configuration. Both energy storage rates and tank temperature profiles were experimentally measured during charge periods representative of two consecutive clear days or combinations of a clear and overcast day. During this time, no draw-offs were conducted. Of particular interest was the effect of rising and falling charge-loop temperatures and collector-loop flow rate on storage tank stratification levels. Results of this study show that the series-connected thermal storage reached high levels of temperature stratification in the storage tanks during periods of rising charge temperatures and also limited destratification during periods of falling charge temperature. This feature is a consequence of the series-connected configuration that allowed sequential stratification to occur in the component tanks and energy to be distributed according to temperature level. This effect was not observed in the parallel charge configuration. A further aspect of the study investigated the effect of increasing charge-loop flow rate on the temperature distribution within the series-connected storage and showed that, at high flow rates, the temperature distributions were found to be similar to those obtained during parallel charging. A disadvantage of both the high-flow series-connected and parallel-connected multi-tank storage is that falling charge-loop temperatures, which normally occur in the afternoon, tend to mix and destratify the storage tanks. 相似文献
Powdered black pepper from Egyptian markets, was irradiated with different recommended doses of gamma rays (5.0 and 10.0 kGy) and with microwaves for different periods (20, 40 and 75 s) to improve its hygienic quality. The most common bacterial isolates were of three generaBacillus, Clostridium andMicrococcus (7.5 × 106), whereas the predominant fungi (7.8 × 104) wereAspergillus species,A. glaucus, A. flavus, A. niger andA. ochraceus. Doses of gamma irradiation used (5.0 and 10 kGy) were sufficient to decrease spore-forming bacteria (SFB) and to inhibit the fungal flora and coliforms which contaminated the black pepper powder. Microwave treatments for 40 s and 75 s were of the same effectiveness whereas treatment for 20 s was less so. GLC analysis proved the presence of 31 peaks, only 19 compounds were identified as monoterpene hydrocarbons (56.21%), the major one being -phellandrene and limonene. Sesquiterpenes were also present, mainly -caryollphyllene (3.69%) as well as oxygenated compounds such as terpenol, geraniol, Me-chavicol, eugenol and anisol. Gamma irradiation at 5 kGy and 10 kGy respectively decreased the numbers of identified compounds from 21 (86.58% concentration) in untreated pepper to 16 (59.22% concentration), 15 (54.06% concentration). In comparison, microwave treatments, particularly for 40 s and 75 s, increased the concentration of the same compounds. The results obtained indicate that microwave treatment, under these conditions, is a safe and suitable technique for decontamination of black pepper which does not result in a great loss of flavour compounds, as compared with recommended doses of gamma irradiation. 相似文献
Structural entropy was developed for detecting the type of localization in charge distributions on a finite grid, especially in mesoscopic electronic systems. However, it is possible to detect and analyze superstructures, i.e., topologies consisting of more structures with different types of localization properties. In the definition of the structural entropy, the von Neumann entropy of the system is divided into two parts: first, the extension entropy, which is simply the logarithm of the occupation number; the second part is the structural entropy. On a structural entropy versus logarithm of the spatial filling factor map, the different types of localizations follow different, well-characterized curves. Spatial filling factor measures the percentage of the “filled” (i.e., high intensity) pixels of the image.An atomic force microscopy (AFM) image can be interpreted as some kind of charge distribution on a grid: after normalization, the darkness (or lightness) of the pixels fulfills all the necessary conditions. AFM image artifacts can be detected by plotting the structural entropy versus the logarithm of the spatial filling factor maps of the images. Not only the type of an added large-scale Gaussian, parabolic, exponential, or other function can be identified, but also by careful study of the curves belonging to the structures, the parameters can be detected, too. 相似文献
