A practical procedure for hypophysectomy of the mouse by paraphyaryngeal approach has been developed by modifying the technics of Motizuki [5] and Lostroh and Jordan [4]. Postoperative care in C3H/He strain of female mice was also studied. The main points improved are as follows: (a) The dental drill bar made of a needle with 2mm outer diameter was devised for making a hole in the cranium (Fig. 2). The bar does not grind the cranium, but cuts it in the circle, resulting in speeding up of the operation and in little bleeding, since the hole is very small. (b) Special attention was paid in breathing of mice during the operation. A longitudinal midline incision of about 1cm was made in muscle encircled the trachea and a pinhole was made by sewing needle into the trachea. A small piece of cotton wool was put on the pinhole and covered by the muscle. It admirably keeps the tracheal hole open for breathing during operation (Fig. 1). (c) After the operation, the mouse was placed in a chamber filled with an atmosphere of 95% O2-5%CO2 gas until recovered, otherwise, all mice died in a few days after operation with inflamation of the lung. The temperature of the recovery chamber is recommended to be kept at about 28 degrees C for ensuring the resuscitation. The intraperitoneal injections of hydrocortone-acetate (0.25mg/0.1ml) just before and 2 days after operation further ameriolated the survival rate.  相似文献

Dexamethasone‐loaded poly(lactide‐co‐glycolide) (PLGA) devices are commonly used as model systems for controlled release. In this study, PLGA nanoparticles containing dexamethasone acetate were prepared by a nanoprecipitation technique in the absence of organochlorine solvents and were characterized by their mean size, ζ potential, scanning electron microscopy, and differential scanning calorimetry to develop a controlled release system. The analytical method for the quantification of dexamethasone acetate by high‐performance liquid chromatography was validated. The results show that it was possible to prepare particles at a nanometric size because the average diameter of the drug‐loaded PLGA particles was 540 ± 4 nm with a polydispersity index of 0.07 ± 0.01 and a ζ potential of ?2.5 ± 0.3 mV. These values remained stable for at least 7 months. The drug encapsulation efficiency was 48%. In vitro tests showed that about 25% of the drug was released in 48 h. © 2014 Wiley Periodicals, Inc. J. This study is carried out on the effect of processing conditions including preparation methods, ultrasonication, film thickness, and thermal annealing on the thermal, mechanical, and piezoelectric properties of copolymer of poly(vinylidene difluoride‐trifluoroethylene) (P(VDF‐TrFE)). Free‐standing films and films on substrates are prepared by solvent casting and spin‐coating, respectively. The results obtained by differential scanning calorimetry show that the Curie transition of the copolymer films was influenced by the preparation methods, with very little difference in the melting transition. The difference was attributed to a less uniform distribution of TrFE comonomer units within the crystalline and amorphous regions of the spin‐coating films. However, no effect of the preparation methods on the piezoelectric properties of the films was observed. It is also found that the short ultrasonication, film thickness higher than critical value of crystallization of P(VDF‐TrFE) (about 100 nm), and drying before annealing did not have a significant effect on the properties of the films. Surprisingly, the ultrasonication had a clear impact on the relaxations at high temperature of the polymer chains. In addition, this study indicates that a short annealing by 10 min at 140°C was enough to obtain well‐crystallized films, which is interesting from the industrial point of view. POLYM. ENG. Fresh poultry meat and poultry products are highly perishable foods and high potential sources of human infection due to the presence of several foodborne pathogens. Focusing on the microbial control of poultry products, the food industry generally implements numerous preventive measures based on the Hazard Analysis and Critical Control Points (HACCP) food safety management system certification together with technological steps, such as refrigeration coupled to modified atmosphere packaging that are able to control identified potential microbial hazards during food processing. However, in recent years, to meet the demand of consumers for minimally processed, high-quality, and additive-free foods, technologies are emerging associated with nonthermal microbial inactivation, such as high hydrostatic pressure, irradiation, and natural alternatives, such as biopreservation or the incorporation of natural preservatives in packaging materials. These technologies are discussed throughout this article, emphasizing their pros and cons regarding the control of poultry microbiota and their effects on poultry sensory properties. The discussion for each of the preservation techniques mentioned will be provided with as much detail as the data and studies provided in the literature for poultry meat and products allow. These new approaches, on their own, have proved to be effective against a wide range of microorganisms in poultry meat. However, since some of these emergent technologies still do not have full consumer's acceptability and, taking into consideration the hurdle technology concept for poultry processing, it is suggested that they will be used as combined treatments or, more frequently, in combination with modified atmosphere packaging.  相似文献

Despite the strong growth of wind power in the world, it is considered a high risk investment due to variability and unpredictability of wind speed. If the energy trade occurs in the free market, there is another risk factor that is the short-term price. This study proposes a methodology of investment risk analysis for wind power plants using innovative stochastic models to generate synthetic time series of wind speed and short-term price. The simulation is implemented using the Monte Carlo method associated with a Cholesky decomposition. The cashflow's gross revenue is obtained through an energy trading model for the Free Contracting Environment (FCE) in Brazil. The investment analysis is based on probability distribution of Net Present Value (NPV) of the power plant's cash flow, as well as the Value at Risk (VaR) and the Conditional Value at Risk (CVaR). A case study was performed for Natal city of Rio Grande do Norte (RN) state in Brazil, and the results indicated the economic viability of the power plant. A sensitivity analysis is developed considering important parameters of the modeling, and its results provide information to the decision-making process.  相似文献
