Airborne communities (mainly bacteria) were sampled and characterized (concentration levels and diversity) at 1 outdoor and 6 indoor sites within a Swiss dairy production facility. Air samples were collected on 2 sampling dates in different seasons, one in February and one in July 2012 using impaction bioaerosol samplers. After cultivation, isolates were identified by mass spectrometry (matrix-assisted laser desorption/ionization-time-of-flight) and molecular (sequencing of 16S rRNA and rpoB genes) methods. In general, total airborne particle loads and total bacterial counts were higher in winter than in summer, but remained constant within each indoor sampling site at both sampling times (February and July). Bacterial numbers were generally very low (<100 cfu/m3 of air) during the different steps of milk powder production. Elevated bacterial concentrations (with mean values of 391 ± 142 and 179 ± 33 cfu/m3 of air during winter and summer sampling, respectively; n = 15) occurred mainly in the “logistics area,” where products in closed tins are packed in secondary packaging material and prepared for shipping. However, total bacterial counts at the outdoor site varied, with a 5- to 6-fold higher concentration observed in winter compared with summer. Twenty-five gram-positive and gram-negative genera were identified as part of the airborne microflora, with Bacillus and Staphylococcus being the most frequent genera identified. Overall, the culturable microflora community showed a composition typical and representative for the specific location. Bacterial counts were highly correlated with total airborne particles in the size range 1 to 5 µm, indicating that a simple surveillance system based upon counting of airborne particles could be implemented. The data generated in this study could be used to evaluate the effectiveness of the dairy plant’s sanitation program and to identify potential sources of airborne contamination, resulting in increased food safety. 相似文献

New directives of the European Union require operators of waste-to-energy (WTE) plants to report the amount of electricity that is produced from renewable sources in the waste feed. Until now, the standard method to determine the portion of renewable electricity is sorting the wastes into defined fractions of fossil organic and biogenic waste components and determining the lower heating value of these fractions. Out of it the amount of electricity production from renewables is calculated. This practice is labor and cost intensive. Therefore, it is usually carried out once a year which provides only a snapshot analysis of limited significance. This paper proposes a method to calculate the portion of electricity produced from renewable materials in waste continuously by solving a set of equations. All data required are either available from literature or from operating data routinely measured in WTE plants. The advantages are statistically derived uncertainty of the result, temporal resolution of the result down to daily mean values, low implementation efforts, and virtually no operational costs. Dehalococcoides strains reductively dechlorinate a wide variety of halogenated compounds including chlorinated benzenes, biphenyls, naphthalenes, dioxins, and ethenes. Recent genome sequencing of the two Dehalococcoides strains CBDB1 and 195 revealed the presence of 32 and 18 reductive dehalogenase homologous genes, respectively, and therefore suggested an even higher dechlorinating potential than previously anticipated. Here, we demonstrate reductive dehalogenation of chlorophenol congeners by Dehalococcoides strains CBDB1 and 195. Strain CBDB1 completely converted 2,3-dichlorophenol, all six trichlorophenols, all three tetrachlorophenols, and pentachlorophenol to lower chlorinated phenols. Observed dechlorination rates in batch cultures with cell numbers of 10(7) mL(-1) amounted up to 35 microM day(-1). Chlorophenols were preferentially dechlorinated in the ortho position, but also doubly flanked and singly flanked meta- or para-chlorine substituents were removed. We used a newly designed computer-assisted direct cell counting protocol and quantitative PCR to demonstrate that strain CBDB1 uses chlorophenols as electron acceptors for respiratory growth. The growth yield of strain CBDB1 with 2,3-dichlorophenol was 7.6 x 10(13) cells per mol of Cl- released, and the growth rate was 0.41 day(-1). For strain 195, fast ortho dechlorination of 2,3-dichlorophenol, 2,3,4-trichlorophenol, and 2,3,6-trichlorophenol was detected, with only the ortho chlorine removed. Because chlorinated phenolic compounds are widely distributed as natural components in anaerobic environments, our results reveal one mode in which the Dehalococcoides species could have survived through earth history. 相似文献

The technique of diffusive gradients in thin-films (DGT) accumulates metals on a Chelex resin after their diffusive transport through a hydrogel. It lowers metal concentrations in soil solution adjacent to the device and induces resupply of metal associated with the solid phase. DGT devices were deployed in an alluvial gley soil for 21 different time periods between 4 h and 19.5 d. The accumulated masses of Cu, Cd, Ni, and Zn were used to calculate the distribution coefficient for labile metal, Kdl, and adsorption and desorption rate constants. Calculations were performed using a dynamic numerical model of DGT-induced fluxes in soils (DIFS). It assumes first-order exchange between solid phase and solution and diffusional transport in both the soil solution and the hydrogel. The DIFS model fitted changes in accumulated mass with time very well. Values of Kdl calculated from DIFS of 100 (Cd), 250 (Cu), 150 (Ni), and 150 (Zn) were larger than values of distribution coefficients estimated by exchange with Ca(NO3)2 but similar to those estimated by isotopic exchange (Cd and Zn only). These results suggest that the solid-phase pool of metal affected by the removal of labile metal by DGT, which operates on a time scale of minutes, is similar to the solid-phase pool of metal that can isotopically exchange with solution on a time scale of 2 d. Response times of minutes were consistent with interaction rates with surfaces, and desorption rate constants agreed with other reported values. An appraisal of the DIFS model demonstrated the importance of the labile pool size in the solid phase for controlling supply to a sink, such as DGT or a plant. As values of Kdl and kinetic parameters are obtained using DGT with minimal soil disturbance and by a similar mechanism to that involved in plant uptake, they may be pertinent to bioavailability studies. 相似文献

Alle Stereoisomeren werden, ausgehend von optisch reinen-Nonalacton-Enantiomeren, synthetisiert und mit Diisobutylaluminiumhydrid selektiv zu den entsprechenden Lactolen reduziert. Die Lactole werden mit den gewünschten Alkoholen in schwach saurer Lösung zucis/trans-Diastereomeren umgesetzt und durch Flüssigchromatographie in optisch reine Stereoisomere getrennt. Die sensorische Charakteristik aller Stereoisomeren der sieben Verbindungen wird aufgezeigt. Die Trennung dercis/trans-Diastereomeren an DB-210 wird vorgestellt und die chirospezifische Differenzierung der acht Verbindungen diskutiert.
Stereoisomeric flavour compounds L. Optically pure 2-alkoxy-5-alkyl-tetrahydrofurans — structures and properties
Summary The synthesis of eight different 2-alkoxy-5-alkyltetrahydrofurans and their stereoisomers is presented, seven of which are known to be flavour compounds of water melons. All stereoisomers were synthesized from optically pure-lactone enantiomers, which were selectively reduced by diisobutylaluminium hydride to the corresponding lactols. The lactols were treated with the desired alcohol component in weakly acidic media to yield thecis/trans diastereoisomers, which were separated into optically pure stereoisomers by liquid chromatography. The sensory characteristics of all four stereoisomers of the seven compounds are given. The Stereodifferentiation of thesecis/trans diastereoisomers on DB-210 is demonstrated and the direct chirospecific differentiation of all eight compounds discussed.相似文献

Several hazelnut allergens with different clinical relevance and crossreactive properties have been identified and characterized so far. The aim of this study was to develop protocols for producing relatively large amounts of three recombinant hazelnut allergens Cor a 1.04, Cor a 2, and Cor a 8 in a folded and immunologically active form. The availability of well-characterized, pure recombinant allergens will improve diagnostic in vitro tests for food allergy, by allowing a highly sensitive component resolved diagnosis. Depending on the individual hazelnut allergen, protocols for heterologous production - either as fusion or nonfusion protein - were developed to obtain homogenous protein batches. The resulting proteins were purified by a two-step FPLC method and their IgE antibody reactivity was verified. Identity was verified by N-terminal sequencing and MALDI-TOF-MS analysis. Their secondary and tertiary structure was controlled by circular dichroism (CD)-spectroscopy and NMR analysis. Decisions on the strategies for expression and purification of allergens on a large scale were made on a case by case basis: Preparation of rCor a 1.04 and rCor a 2 as fusion proteins in E. coli from inclusion bodies resulted in approximately 10 mg pure protein per liter whereas rCor a 8 expression in yeast as nonfusion protein yielded 30 mg/L. 相似文献

Zusammenfassung Für vergleichende Untersuchungen werden aus den entfetteten Mehlen von Weizen, Roggen, Gerste, Hafer, Reis, Hirse und Mais durch successive Extraktion mit Wasser, Salzlösung und Ethanol/Wasser Albumin-, Globulin-, Prolamin- und Glutelin-fraktionen gewonnen. Hinsichtlich der Verteilung des Gesamtproteins auf these Fraktionen treten zwischen den Getreidearten große Unterschiede auf. Die Aminosäurezusammensetzung der Proteinfraktionen korreliert nur bei den Prolaminen mit der Phylogenie. Bei Getreidearten, die Cöliakie auslösen können (Weizen, Roggen, Gerste), ist die Aminosaurezusämmensetzung der Prolamine sehr ähnlich und stark abweichend von der anderer Getreidearten (Mais, Hirse, Reis). Die in bezug auf Kleberbildung bestehenden Unterschiede zwischen Weizen und den übrigen Getreidearten spiegeln sich in der unterschiedlichen Verteilung und Aminosäurezusammensetzung der Prolamine und Gluteline wider.

Comparative investigations of partial amino acid sequences of prolamines and glutelins from cerealsI. Fractionation of proteins according to Osborne
Summary Albumins, globulins and prolamines of wheat, rye, barley, oats, rice, sorghum and maize were obtained by extraction of defatted meals successively with water, salt solution and aqueous ethanol. The residual proteins were taken as glutelins. The cereals contain very different amounts of the four protein fractions mentioned. The amino acid composition of the prolamines only is correlated with the phylogenetic relationships. With the cereals causing coeliac disease (wheat, rye, barley) the amino acid composition of the prolamines is very similar and strongly different from that of the other cereals. The differences between wheat and other cereals in respect to gluten formation is reflected very well by the amounts and amino acid compositions of prolamines and glutelins.

Zusammenfassung Die Konservierungsstoffe Sorbinsäure, Benzoesäure, PHB-Ester und Propionsäure können nebeneinander in einem Analysengang nach einer isotachophoretischen Trennung auf Grund ihrer unterschiedlichen Beweglichkeiten quantitativ bestimmt werden. Störende Lebensmittelinhaltsstoffe (Lipide) werden vor der eigentlichen Bestimmung durch Äther-Extraktion unter Verwendung von Extrelut-Säulen entfernt. Der hier mitgeteilte Analysengang zeichnet sich nach unserer Erfahrung durch einen geringen zeitlichen Aufwand bei gleichzeitig hohen Wiederfindungsraten aus. Zumindest bei den von uns untersuchten Lebensmittelgruppen konnte eine Störung der quantitativen Bestimmung durch Begleitsubstanzen nicht beobachtet werden. Der erfaßte Bereich ist groß und bestreicht praktisch das ganze Konzentrationsgebiet, in dem Konservierungsstoffe üblicherweise angewendet werden.
Isotachophoretic determination of preservatives
Summary The preservatives sorbic acid, benzoic acid, methyl-, ethyl-, propylester of p-hydroxybenzoic acid and propionic acid can be determined quantitatively in one step by isotachophoretic separation based on the different electrophoretic mobilities of these compounds. The determination involves a direct extraction step in the so called Extrelut-columns, where the preservatives are separated from the food sample, mainly from the lipid phase. The whole analysis procedure is short and recoveries for the preservatives in the samples tested are very good. There is no interference by other substances and the range of concentrations for which a determination is possible is large and encompasses virtually all concentrations used in commercial practices.相似文献
