In this study the production of extracellular polysaccharides by the non-pathogenic soil bacteria Arthrobacter viscosus has been investigated. Different variables affecting extracellular polysaccharide production such as the carbon source (glucose or xylose), the agitation speed and the pH have been analysed.
In a first stage, experiments in shaken conical flasks (250 ml), containing 50 ml of culture medium, were carried out. Using xylose (25 g/l) as the carbon source at an initial pH 8 improved the extracellular polysaccharides levels obtained.
In a second stage, the experiments were scaling in bioreactors. Cultivation was carried out in discontinuous mode and with/without pH control. Polysaccharide production reached a maximum of 10 g of crude product per litre of growth medium after 14 days and the relationship between product formation and cell growth of A. viscosus is 2.7 g polysaccharide per gram biomass. This production was obtained at the optimal conditions determined with pH control at pH 7, xylose as carbon source (25 g/l) and an agitation rate of 800 rpm. 相似文献
A review is carried out on how queueing network models with blocking have been applied so far into the performance evaluation and prediction of Software Architectures (SA). Queueing network models with finite capacity queues and blocking have recently been introduced and applied as more realistic models of systems with finite capacity resources and population constraints. Queueing network models have been often adopted as models for the evaluation of software performance. Starting from our own experience, we observe the need of a more accurate definition of the performance models of SA to capture some features of the communication systems. We consider queueing networks with finite capacity and blocking after service (BAS) to represent some synchronization constraints that cannot be easily modeled with queueing network models with infinite capacity queues. We investigate the use of queueing networks with blocking as performance models of SA with concurrent components and synchronous communication. Queueing theoretic analysis is used to solve the queueing network model and study the synchronous communication and performance of concurrent software components. Our experience is supported by other approaches that also propose the use of queueing networks with blocking. Directions for future research work in the field are included. 相似文献
A numerical and an experimental analysis of velocity and temperature fields inside a storage tank submitted to natural convection is presented. The analysis was performed in two stages. In the first stage, the temperature profile along the vertical axis of the storage tank was obtained experimentally and numerically, for cooling time ranging from 45 to 60 h. The numerical analysis was carried out using a transient bi-dimensional model in cylindrical co-ordinates. In the second stage, after the numerical code validation, 40 cases of cooling with four aspect ratios, five insulation thicknesses, and two different volumes were simulated. In all simulations, thermal losses for the environment in all tank walls (side, top and bottom) were considered. Two correlations for the Nusselt number, encompassing all the forty cases, were obtained with these results. 相似文献
Fatty acid esters, high in linoleic acid, were prepared and stored for long-term engine tests. Storage tests with these esters were undertaken to obtain more information on optimal storage requirements and general stability characteristics. Samples were kept at three temperature levels (20 C, 30 C and fluctuating around 50 C) for a 90-day period and were removed at regular intervals for chemical and physical analysis. The influence of air, temperature, light, TBHQ and contact with mild steel was evaluated by comparing the free fatty acid, peroxide, anisidine, ultraviolet absorption, viscosity and induction periods. A statistical model was used to evaluate the data and to reduce the large number of data points to comparable curves. Storage of esters in contact with air, especially at a temperature above 30 C, resulted in significant increases in peroxide, ultraviolet absorption, free fatty acid, viscosity and anisidine values. Exclusion of air retarded oxidation at all temperature levels. A direct relationship between viscosity increases and oxidation parameters was evident. Exposure to light caused a small increase in the oxidation parameters of esters stored at the highest temperature level. Addition of TBHQ prevented oxidation of samples stored under moderate conditions. Under unfavorable storage conditions the anti-oxidant was no longer effective. Mild steel had very little effect on the oxidation parameters. Only the anisidine values of samples stored at the highest temperature level were slightly increased. Methyl esters performed slightly better than ethyl esters during the storage test. The following practical guidelines for storage of fatty acid ester fuels are: (i) airtight containers should be used; (ii) the storage temperature should be <30 C; (iii) mild steel (rust free) containers may be used, and (iv) TBHQ has a beneficial effect on oxidation stability. 相似文献
An optimised low-frequency solution algorithm is proposed that reduces the electromagnetic wave equation to a small number of scalar potential problems. The latter can be solved using black-box algebraic multigrid solvers. Within the framework of the finite integration technique, the reduction of the wave equation is accomplished by tree/cotree decompositions of primary and dual grids, which allow for highly efficient solutions of Ampere's and Faraday's equations. Here, we describe the development of a reacting flow multi-species/combustion methodology, implemented as an extension to the differential reduced ejector analysis (DREA) computer program [Mathematical and computer modeling, vol. 31, 2000, p. 21; Appl. Math. Model. 25 (2001) 427; Comput. Math. Appl. 43(10–11); NASA Contractor Report, 1998]. Use of the single fluid IVP solver framework that was developed for the original DREA model has been directly coupled into the combustion formulation. With these modifications, the analysis has an elementary single step reaction Fuel+Oxidizer→Product combustion capability. Though approximate in nature, the simplicity and efficiency of the DREA formulation make it suitable for its original niche, namely design and preliminary design environments where more complex and expensive models may be inappropriate. 相似文献
This paper presents a hyperstability theorem for a class of hybrid dynamic systems composed of coupled differential and difference equations subject to (possibly) time-varying nonlinearities satisfying a Popov-type inequality. The nonlinear controller generates the plant input at all times from its sampled values by defining an extended discrete system. The hyperstability results are obtained from this discrete system of special type whose state consists of the sampled continuous substate and the digital substate of the given hybrid system. Acetohydroxy acid synthase (AHAS) and threonine deaminase (TD) activities were found in Streptococcus bovis and shown to be involved in the biosynthesis of the branched chain amino acids isoleucine, leucine and valine. Apparent lack of repression of AHAS synthesis by the end-products and reduced sensitivity of S. bovis growth to analogues of the branched chain amino acids suggested that secretion of isoleucine, leucine and valine in the growth medium may be a consequence of the regulatory features of AHAS. A glycyl-leucine-resistant mutant with reduced TD activity secreted a reduced amount of isoleucine and an increased amount of valine, which might be a result of the reduced rate of synthesis of the isoleucine precursor alpha-ketobutyrate and of a consequent preferential carbon flow through the valine branch of the pathway. 相似文献
The evolution of Drosophila subobscura mitochondrial DNA has been studied in experimental populations, founded with flies from a natural population from Esporles (Majorca, Balearic Islands, Spain). This population, like other European ones, is characterized by the presence of two very common (>96%) mitochondrial haplotypes (called I and II) and rare and endemic haplotypes that appear at very low frequencies. There is no statistical evidence of positive Darwinian selection acting on the mitochondrial DNA variants according to Tajima's neutrality test. Two experimental populations, with one replicate each, were established with flies having a heterogeneous nuclear genetic background, which was representative of the composition of the natural population. Both populations were started with the two most frequent mitochondrial haplotypes, but at different initial frequencies. After 13 to 16 generations, haplotype II reached fixation in three cages and its frequency was 0.89 by generation 25 in the fourth cage. Random drift can be rejected as the force responsible for the observed changes in haplotype frequencies. There is not only statistical evidence of a linear trend favoring a mtDNA (haploid) fitness effect, but also of a significant nonlinear deviation that could be due to a nuclear component. 相似文献
