Gold nanoparticles supported on titania catalyse, even at a ppm loading level, the quantitative reduction of nitroarenes into anilines and nitroalkanes into alkylhydroxylamines by the ammonia⋅ borane complex. No dehalohalogenation was seen in the case of chloro‐ or bromonitroarenes, while ester, cyano, or carboxylic acid functionalities also remain intact. Biallelic pathogenic variants in the SEC23B gene cause congenital dyserythropoietic anemia type II (CDA II), a rare hereditary disorder hallmarked by ineffective erythropoiesis, hemolysis, erythroblast morphological abnormalities, and hypo-glycosylation of some red blood cell membrane proteins. Abnormalities in SEC23B, which encodes the homonymous cytoplasmic COPII (coat protein complex II) component, disturb the endoplasmic reticulum to Golgi trafficking and affect different glycosylation pathways. The most harmful complication of CDA II is the severe iron overload. Within our case series (28 CDA II patients), approximately 36% of them exhibit severe iron overload despite mild degree of anemia and slightly increased levels of ERFE (the only erythroid regulator of hepcidin suppression). Thus, we hypothesized a direct role of SEC23B loss-of-function in the pathomechanism of hepatic iron overload. We established a hepatic cell line, HuH7, stably silenced for SEC23B. In silenced cells, we observed significant alterations of the iron status, due to both the alteration in BMP/SMADs pathway effectors and a reduced capability to sense BMP6 stimulus. We demonstrated that the loss-of-function of SEC23B is responsible of the impairment in glycosylation of the membrane proteins involved in the activation of the BMP/SMADs pathway with subsequent hepcidin suppression. Most of these data were confirmed in another hepatic cell line, HepG2, stably silenced for SEC23B. Our findings suggested that the pathogenic mechanism of iron overload in CDA II is associated to both ineffective erythropoiesis and to a specific involvement of SEC23B pathogenic variants at hepatic level. Finally, we demonstrated the ability of SEC23B paralog, i.e., SEC23A, to rescue the hepcidin suppression, highlighting the functional overlap between the two SEC23 paralogs in human hepatic cells. 相似文献

Cotton and wool fabrics were dyed with nine natural dyes obtained by aqueous extraction of the original plants/insect in an attempt to reconstruct traditional textile dyeing recipes, to optimise the methodology at all stages, i.e. extraction, mordanting and dyeing, and to standardise it. Cochineal, madder, alkanna, henna, brazilwood, red sandalwood, safflower, indigo and logwood were used for the dyeings, which were carried out directly and after mordanting of the textile material. A variety of mordants, namely, aluminium potassium sulphate, potassium dichromate, copper sulphate, zinc chloride, iron(iii ) chloride, iron(ii ) sulphate and tin chloride, is anticipated to meet both early and recent requirements and options. The dyeings were evaluated through colour measurements and standard wash, light and rub fastness tests. Generally, the mordanting process known for many centuries and connected with the textile dyeing resulted in an improvement in dye absorption and fastness properties mainly for the cotton samples, as is concluded from the tests and measurement assessments. 相似文献

Ribosomal RNA (rRNA) biogenesis takes place in the nucleolus, the most prominent condensate of the eukaryotic nucleus. The proper assembly and integrity of the nucleolus reflects the accurate synthesis and processing of rRNAs which in turn, as major components of ribosomes, ensure the uninterrupted flow of the genetic information during translation. Therefore, the abundant production of rRNAs in a precisely functional nucleolus is of outmost importance for the cell viability and requires the concerted action of essential enzymes, associated factors and epigenetic marks. The coordination and regulation of such an elaborate process depends on not only protein factors, but also on numerous regulatory non-coding RNAs (ncRNAs). Herein, we focus on RNA-mediated mechanisms that control the synthesis, processing and modification of rRNAs in mammals. We highlight the significance of regulatory ncRNAs in rRNA biogenesis and the maintenance of the nucleolar morphology, as well as their role in human diseases and as novel druggable molecular targets. 相似文献

Transmission electron microscopy‐based grain orientation mapping method was employed to investigate the microtexture of the alumina scale formed in commercial thermal barrier coatings (TBCs) with two standard types of Pt‐enriched bond coats. Reliable orientation/phase maps with a spatial resolution down to 2 nm were acquired on the alumina grains. It was observed that the alumina scale on the Pt‐aluminide β‐phase bond coat has a stronger c‐axis texture normal to the bond coat surface, in comparison with that on the Pt‐diffused γ/γ′‐phase bond coat. The microtexture of the alumina scale could affect its effective coefficient of thermal expansion, which is a contributor to the severity of the bond coat rumpling mechanism of TBCs failure. 相似文献

Mature ‘Chondrolia Chalkidikis’ olives were processed in an industrial olive oil mill equipped with a three‐phase decanter. Water was added to the decanter at a 1:2 water‐to‐paste ratio. Olive mill waste water (ΟΜWW) was used to replace the added water at a rate of 50 or 100%. Following the final separation, the obtained oil was used for chemical analysis and sensory evaluation. All oils had similar acidity, peroxide and Κ values. OMWW‐treated olive oils presented higher total phenolic content and higher antioxidant activity based on DPPH and oven tests, but lower chlorophyll and carotenoids content. However, there was no significant difference between the 50 and 100% replacement. The phenolic profile of the treated olive oils analyzed by quantitative 1Η NMR revealed more than twofold oleocanthal and oleacein as well as oleuropein and ligstroside aglycone contents than in the control. Sensory evaluation of treated oils also showed an enhancement of fruity, bitter and pungent attributes compared to the control. 相似文献

Two carbon samples derived from poly(4-styrenesulfonic acid-co-maleic acid) based polymer by carbonization between 700 and 800 °C were oxidized to two different levels of surface acidity. The surfaces of resulting adsorbents were characterized by potentiometric titration, adsorption of nitrogen, FTIR, SEM/EDAX and thermal analysis. The materials were used as adsorbents of 4,6-dimethyldibenzothiophene (4,6-DMDBT) from hexadecane with initial concentration of sulfur between 10-150 ppmw. Although it was found that pores with diameter less than 10 Å govern the amount of 4,6-DMDBT adsorbed, that amount is enhanced when acidic groups are present in the larger pores owing to the contributions of specific interactions. Surface chemistry plays an important role in reactive adsorption and deposition of the products of surface reactions in the pore system. 相似文献
