The total synthesis of the naturally occurring (Z)-2-methoxy-5-hexadecenoic acid and (Z)-2-methoxy-6-hexadecenoic acid was accomplished using as a key step Mukaiyama's trimethylsilyl cyanide addition to 4- and 5-pentadecenal, respectively. These syntheses further confirm the structures of the natural marine fatty acids and corroborate their cis double-bond stereochemistry. The title compounds were antimicrobial against the Gram-positive bacteria Staphylococcus aureus (MIC 0.35 micromol/mL) and Streptococcus faecalis (MIC 0.35 micromol/mL).  相似文献

Glycan microarrays have emerged as novel tools to study carbohydrate–protein interactions. Here we describe the preparation of a covalent microarray with lipochitin oligosaccharides and its use in studying proteins containing LysM domains. The glycan microarray was assembled from glycoconjugates that were synthesized by using recently developed bifunctional chemoselective aminooxy reagents without the need for transient carbohydrate protecting groups. We describe for the first time the preparation of a covalent microarray with lipochitin oligosaccharides and its use for studying proteins containing LysM domains. Lipochitin oligosaccharides (also referred to as Nod factors) were isolated from bacterial strains or chemoenzymatically synthesized. The glycan microarray also included peptidoglycan‐related compounds, as well as chitin oligosaccharides of different lengths. In total, 30 ligands were treated with the aminooxy linker molecule. The identity of the glycoconjugates was verified by mass spectrometry, and they were then immobilized on the array. The presence of the glycoconjugates on the array surface was confirmed by use of lectins and human sera (IgG binding). The functionality of our array was tested with a bacterial LysM domain‐containing protein, autolysin p60, which is known to act on the bacterial cell wall peptidoglycan. P60 showed specific binding to Nod factors and to chitin oligosaccharides. Increasing affinity was observed with increasing chitin oligomer length.  相似文献

In this study, we investigated the possibility of creating easy to handle films based on poly(vinyl alcohol) (PVA) and β‐lactoglobulin (βlg) blends using the casting method. Four different variables were studied to obtain these films: different proportions of PVA and βlg, different pHs (10, 7, 5.5, and 2.5), several molecular masses of PVA (130,000, 13,000–23,000, and 2000 g/mol), and the denaturation of βlg. The first objective was to obtain films, and the second was to characterize them by differential scanning calorimetry, thermogravimetric analysis, and mechanical testing. Significant variations in behavior were found, and the variables studied modified the blends in different ways. In particular, better results were achieved when the βlg proportion was less than 30 wt %, and when a pH of 2.5 was used in conjunction with a high molecular mass (130,000 g/mol) of PVA. © 2014 Wiley Periodicals, Inc. In this paper, we report on a general approach for the detection of a specific tumoural biomarker directly in serum. Such detection is made possible using a protein-binding peptide selected through an improved phage display technique and then conjugated to engineered microparticles (MPs). Protein biomarkers represent an unlimited source of information for non-invasive diagnostic and prognostic tests; MP-based assays are becoming largely used in manipulation of soluble biomarkers, but their direct use in serum is hampered by the complex biomolecular environment. Our technique overcomes the current limitations as it produces a selective MP—engineered with an antifouling layer—that ‘captures’ the relevant protein staying impervious to the background. Our system succeeds in fishing-out the human tumour necrosis factor alpha directly in serum with a high selectivity degree. Our method could have great impact in soluble protein manipulation and detection for a wide variety of diagnostic applications.  相似文献

The very long-chain fatty acids, (5E,9Z)-6-bromo-5,9-tetracosadienoic, (5E,9Z)-6-bromo-23-methyl-5,9-tetracosadienoic, (5E,9Z)-6-bromo-5,9-pentacosadienoic and (5E,9Z)-6-bromo-24-methyl-5,9-pentacosadienoic acids, were identified in the phospholipids (mainly phosphatidylethanolamine) of the spongeAgelas sp. Structure elucidation was accomplished by means of mass spectrometry and chemical transformations, including deuteration with Wilkinson's catalyst. All of the sterols from the sponge had the Δ5,7 nucleus, with 24-methylcholesta-5,7,22-trien-3β-ol (ergosterol) and 24-ethylcholesta-5,7,22-trien-3β-ol being the most abundant.  相似文献

The TCV tokamak contributes to the physics understanding of fusion plasmas, broadening the parameter range of reactor relevant regimes, by investigations based on an extensive use of the existing main experimental tools: flexible shaping and high power real time-controllable electron cyclotron heating (ECH) and current drive (ECCD) systems. A proposed implementation of direct ion heating on the TCV by the installation of a 20–35 keV neutral beam injection (NBI) with a total power of 1–3 MW would permit an extension of the accessible range of ion to electron temperatures (Ti/Te  0.1–0.8) to well beyond unity, depending on the NBI/ECH mix and the plasma density. A NBI system would provide TCV with a tool for plasma study at reactor relevant Ti/Te ratios ~1 and in investigating fast ion and MHD physics together with the effects of plasma rotation and high plasma β scenarios. The feasibility studies for a NBI heating on TCV presented in this paper were undertaken to construct a specification for the neutral beam injectors together with an experimental geometry for possible operational scenarios.  相似文献

The main purpose of this study was to determine if a method developed in the laboratory by HPLC-ESI-QToF was able to identify the characteristic compounds of the Mexican cochineal (Dactylopius coccus) in freshly dyed wool fiber and following an artificially accelerated aging process. Wool fibers were dyed with various formulations of popular interest (red-orange, Mexican red, cochineal red, and purple). These formulations are currently used by indigenous communities in the south of Mexico. Some dyed wools were aged in an UV chamber, in order to simulate the deterioration that a fiber dyed in a previous time could have. Notable results were the identification of the main component, carminic acid, and the respective biomarkers of the Mexican cochineal (dcII, dcIV, and dcVII) with a small amount of sample and in a short time, in three of the four formulations before and after aging, thus providing a method that can probably be used for conservation purposes of cultural heritage. It was concluded that when alum is used as a mordant and as a carbonate pH fixer, the fixation of cochineal on the wool is not very strong. This is one of the few works where Mexican cochineal has been studied, simulating the deterioration due to the passage of time in wool samples of different colors with the same natural dye. As a way of describing the color more objectively, the color was measured with the CIE L*A*B* scale in the four formulations of interest, in recently dyed and artificially aged wool.  相似文献

Veiled virgin olive oil (VOO) samples of nine different olive cultivars are chosen to have a wide range of physicochemical and biological properties of colloidal dispersions. The contents of proteins and phospholipids range from 40 to 190 mg kg−1 and from 70 to 200 mg kg−1, respectively. The effect of lab-scale centrifugation on cloudy appearance is studied measuring the decrease of turbidity grade values. The time to obtain unveiled oils (20 NTU) is modeled by a logistic equation, and a clear relationship between the initial water content and the above time is observed with a different trend between two groups of the VOO samples. Four VOO samples are selected to study the aggregation phenomena of microdroplets of water, pulp particles, and olive stone fragments via optical microscopy and dynamic light scattering during lab-scale gravity sedimentation. All VOOs are unstable with the cloudiness disappearing within the 230 days of investigation due to an overall diameter increase of cloudy components which is modeled by a power-law equation. The VOO samples, characterized by both small diameter values of dispersed components (150–250 nm) and high values of water content, show the fastest aggregation kinetics, but they have the longest time of cloudiness stability. Practical Applications: Water content and size distribution of VOO cloudy components can be key factors to control the colloidal stability. If removal of cloudy appearance is required, centrifugation can be applied to obtain a fast oil clarification which shows a power law relationship of water content with time. Instead, if physical stability of the colloidal dispersion is required, the aggregation phenomena should be slow down through VOO processing to obtain small diameters of the cloudy components. Tuning both the water content and dispersed phase diameter in the VOO can be the first step towards the control of phenomena related to the colloidal dispersion for every olive oil processing organization, above and beyond the simple removal of cloudy appearance by filtration.  相似文献

The residual vibrations of highly resonant flexible structures can be usually composed of sinusoidal signals. The identification of the frequencies, amplitudes and/or phases of these sinusoidal signals may be of interest for health monitoring or to update controller parameters in active vibration control systems. This work considers a previously developed algebraic estimator of the frequencies, amplitudes and phases of two sinusoidal signals in order to identify the first two natural frequencies of flexible-beam-like structures. The influence of the unconsidered vibration modes on the estimation accuracy is studied by simulating a residual vibration formed by four sinusoidal components. It has been shown that the velocity and accuracy in the estimation of the first two natural frequencies depend on the distance between frequencies and the amplitudes of the first three vibration components. Finally, the beam parameters are related to the estimation performance.  相似文献
