Ten calves less than 14 h of age (average 8.6 h) were anaesthetized, and the intestine was ligated into segments 10 cm in length at 3-cm intervals beginning 1.8 m anterior of the ileocecal junction and proceeding proximally. Seven treatments were assigned in random order to segments in three successive sections of the small intestine. Segments received 1 ml of viable bacteria of intestinal origin, autoclaved bacteria of intestinal origin, or sterile microbiological broth at zero time; then after 4 h they were injected with iodine-125 labeled gamma-globulin. After an additional 1.5 h, the experiment was ended and uptake assayed. Two treatments measured anaerobic microbial growth after 4 h incubation with 1 ml of either sterile broth or live bacteria culture. Residual [iodine-125] gamma-globulin was measured in segments receiving 1 ml of sterile broth or live bacteria culture with 5.5-h incubation followed by 15-s exposure to labeled gamma-globulin. Uptake was lowest in segments receiving live bacteria as compared to segments receiving sterile inocula. Number of bacteria per gram of tissue was correlated negatively with uptake. Low corticosteroids in serum were associated with low uptake of gamma-globulin.  相似文献

With 32 lactating Holstein cows we evaluated physiological effects of gossypol and cyclopropenoid fatty acids in diets containing 18.5% whole cottonseed (dry matter) based on corn, corn silage, and soybean meal. All cows consumed a control diet for the first 2 wk of lactation and then were assigned to either control or whole cottonseed diet for the remainder of their lactation. Milk production, milk fat and protein percentages, and daily dry matter intake were measured. Dry matter intakes were less for cows consuming the whole cottonseed diet, but net energy intake was similar for all diets. Milk fat from cows consuming whole cottonseed contained detectable concentrations of cyclopropene fatty acids. Total lipid in plasma, total serum cholesterol, serum gossypol, and apparent liver gossypol concentrations were greater in cows fed whole cottonseed. Gossypol and cyclopropenoid fatty acids appeared to be absorbed from the gut of cows fed whole cottonseed. Small amounts of gossypol in serum and liver tissue and small amounts of cyclopropene fatty acids in adipose tissue lipids and milk fat indicate a need to elucidate the significance of these physiologically active compounds in the human diet and their biological effects on lactating dairy cows.  相似文献

In this work we present a method for the deposition of periodically ordered, c-axis aligned ZnO nanorod arrays. By using chemical bath deposited films in conjunction with silica templating through nanosphere monolayers, masks suitable for high temperature deposition are created. A vapour phase transport technique is then used to deposit ordered arrays, quickly and inexpensively in a manner ideal for low cost, scalable and reproducible growth on a diverse range of substrates.  相似文献

Using combined semiempirical quantum mechanics and molecular mechanics (QM/MM) and ab initio self-consistent reaction field (SCRF) calculations, we determined that a low-barrier hydrogen bond (LBHB) is formed when the mechanism-based substrate 8-methylpterin binds to dihydrofolate reductase (DHFR). The substrate initially was assumed bound either in the ion-pair form corresponding to N3-protonated substrate hydrogen (H) bonded to the unprotonated (carboxylate) of the conserved Glu30 residue in the active site, or in the neutral-pair form corresponding to unprotonated substrate H bonded to the neutral (carboxylic acid) from of Glu30. The free energy of interaction of these H-bonded systems with the protein/solvent surroundings was computed using a coordinate-coupled free energy perturbation (FEP) method implemented within the molecular dynamics (MD) simulation scheme and using a semiempirical (PM3) QM/MM force field. The free energy obtained from the QM/MM force-field simulations corresponds most closely with the corresponding free energy component obtained from HF/6-31G* SCRF calculations using a value of 2 for the dielectric constant (epsilon) for the solvated protein. Calculations were performed at levels ranging from HF/6-31G to MP2/6-31G* to B3LYP/6-31 + G**, with varying dielectric constants. The energy-minimized path for motion of the proton in the H bond along a one-dimensional reaction coordinate was calculated at HF/6-31G, HF/6-31G* (epsilon = 1) and B3LYP/6-31G* (epsilon = 2) levels. These calculations identified a second neutral-pair complex, involving the 2-amino group of substrate, which also interacts with Glu30, which is lower in energy than the ion-pair form. A harmonic vibrational analysis shows that the first vibrational state appears to lie near or above the TS connecting potential energy minima corresponding to the two neutral-pair configurations, thus indicating an LBHB. Consequently, the H-bonded system will have a significant probability of being found in the ion-pair form, in agreement with experimental spectral studies indicating an enzyme-bound cation and suggesting that the LBHB would activate substrate towards hydride-ion transfer from NADPH.  相似文献

The paper discusses the concept of using morphological examination of wear debris to assess the condition of gears in a gear box. The debris examined was produced by three sets of gears that were tested in two special-purpose gear rigs. The experimental techniques employed were transmission and scanning electron microscopy, optical microscopy, and ferrography. It is suggested that an understanding of gear tooth wear may be soundly based on the role of plastic deformation in the wear process. It is concluded that the links between wear and tooth fracture have yet to be established.  相似文献

The objective of the present study was to compare serum IgG concentration, weight gain, and health characteristics in Irish spring-born dairy calves fed colostrum stored using a range of conditions. Immediately after birth, 75 dairy heifer calves were assigned to 1 of 5 experimental colostrum treatments: (1) fresh pasteurized colostrum, fed immediately after pasteurization; (2) fresh colostrum, fed immediately after collection but not pasteurized; (3) colostrum stored unpasteurized at 4°C in a temperature-controlled unit for 2 d before being fed to calves; (4) colostrum stored unpasteurized at 13°C in a temperature-controlled unit for 2 d before being fed to calves; and (5) colostrum stored unpasteurized at 22°C in a temperature-controlled unit for 2 d before being fed to calves. All colostrum had IgG concentrations >50 g/L and was fed to calves promptly after birth. Blood samples were obtained from calves via the jugular vein at 0 h (before colostrum feeding) and at 24 h of age to determine the rate of passive transfer of IgG; individual calf live-weights were recorded to monitor weight gain (kg/d) from birth to weaning. Colostrum stored in warmer conditions (i.e., 22°C) had >42 times more bacteria present and a pH that was 0.85 units lower and resulted in a serum IgG concentration that was almost 2 times lower compared with colostrum that was pasteurized, untreated, or stored at 4°C for 2 d. Colostrum stored at 4°C for 2 d had more bacteria present than pasteurized and fresh colostrum but did not result in reduced calf serum IgG concentrations. Average daily weight gain from birth to weaning did not differ among treatments. Even if colostrum has sufficient IgG (>50 g/L) but cannot be fed to calves when freshly collected, storage at ≤4°C for 2 d is advisable to ensure adequate passive transfer when it is consumed by the calf.  相似文献

The authors compared longitudinal treatment outcomes for depressed substance-dependent veterans (N = 206) assigned to integrated cognitive–behavioral therapy plus standard pharmacotherapy (ICBT + P) or 12-step facilitation therapy plus standard pharmacotherapy (TSF + P). Drug and alcohol involvement and depressive symptomology were measured at intake and at 3-month intervals during treatment and up to 1 year posttreatment. Participants in both treatment conditions showed decreased depression and substance use from intake. ICBT + P participants maintained improvements in substance involvement over time, whereas TSF + P participants had more rapid increases in use in the months following treatment. Decreases in depressive symptoms were more pronounced for TSF + P than ICBT + P in the 6 months posttreatment. Within both treatment groups, higher attendance was associated with improved substance use and depression outcomes over time. Initial levels of depressive symptomology had a complex predictive relationship with long-term depression outcomes. Early treatment response predicted long-term substance use outcomes for a portion of the sample. Although both treatments were associated with improvements in substance use and depression, ICBT + P may lead to more stable substance use reductions compared with TSF + P. A novel metal-supported solid oxide fuel cell has been developed that is capable of operating at temperatures of 500–600 °C. The rationale behind the materials used to construct this fuel cell type is given, and results are presented from cell testing on hydrogen and reformed natural gas, including durability trials of some 2500 h duration. This new fuel cell variant is shown to be tolerant of carbon monoxide, durable, robust to thermal and redox cycling, and capable of delivering technologically relevant power densities. The effects of remote stimulation on the refractory characteristics of myelinated nerve fibers were investigated using computer simulations of nerve action potentials, in response to spatially separated conditioning and test stimuli. The behavior of the test action potential was strongly influenced by its direction of propagation relative to that of the conditioning action potential. Under certain conditions, the variation of relative refractory period with conduction velocity (CV) changed from inverse, for propagation in opposing directions, to direct, for propagation in the same direction. A similar directionally dependent result occurred in the study of relative refractory period as a function of stimulus intensity. At certain interstimulus intervals, the test stimulus elicited action potentials which would conduct in the direction opposite to the conditioning action potential, but would not conduct in the wake of that conditioning action potential. These results are explained in terms of the spatial spread of stimulus current resulting from distant placement of the stimulating electrode in a volume conductor. Clinical repercussions of these results for correction of refractory period in collision neurography are discussed.  相似文献
