Lactate dehydrogenase from Bacillus stearothermophilus is specific for NAD+. There have been several attempts to alter the cofactor specificity of this enzyme, but these have yielded enzymes with relatively low activities that still largely prefer NAD+. A modified consensus approach was used to create a library of phylogenetically preferred amino acids situated near the cofactor binding site, and variants were screened for their ability to utilize NMN+. A triple mutant (Mut31) was discovered that proved to be more catalytically efficient than wild-type. Mut31 was also better at utilizing NAD+ than the wild-type enzyme and was weakly active with NADP+ and NMN+. An analysis of single amino acid substitutions suggested that all three mutations worked in a concerted fashion to yield robust cofactor utilization. When two previously identified amino acid substitutions were introduced into the Mut31 background, the resultant quintuply substituted enzyme not only utilized NADP+ far better than the wild-type enzyme, it actually inverted its preference for NAD+ and NADP+.  相似文献

Commercial magnesium anodes were evaluated using ASTM G 97-89 for a standard test along with a technique of electrochemical impedance. Several heat treatments were performed, and the anodic efficiency was determined, showing an incremental increase of 10 to 12 % of the efficiency demonstrated by the nontreated commercial anodes (as-cast condition). The increase in efficiency was related to the microstructural characteristics. The appearance of second phase particles influenced the form of the corrosion process.  相似文献

Variability management is a recurrent issue in systems engineering. It arises for example in enterprise systems, where modules are configured and composed to meet the requirements of individual customers based on modifications to a reference model. It also manifests itself in the context of software product families, where variants of a system are built from a common code base. This paper proposes an approach to capture system variability based on questionnaire models that include order dependencies and domain constraints. The paper presents analysis techniques to detect circular dependencies and contradictory constraints in questionnaire models, as well as techniques to incrementally prevent invalid configurations by restricting the space of allowed answers to a question based on previous answers. The approach has been implemented as a toolset and has been used in practice to capture configurable process models for film post-production.

Increased operator productivity is a desired outcome of user-CAD interaction scenarios. Two objectives of this research were to (1) define a measure of operator productivity and (2) empirically investigate the potential effects of CAD interface design on operator productivity, where productivity is defined as the percentage of a drawing session correctly completed per unit time. Here, AutoCAD provides the CAD environment of interest. Productivity with respect to two AutoCAD interface designs (menu, template) and three task types (draw, dimension, display) was investigated. Analysis of user productivity data revealed significantly higher productivity under the menu interface condition than under the template interface condition. A significant effect of task type was also discovered, where user productivity under display tasks was higher than productivity under the draw and dimension tasks. Implications of these results are presented.  相似文献

This paper reports the experimental findings on the tensile behavior of strain-hardening cement-based composites (SHCC). The composites were subjected to the combined effects of elevated temperatures and internal moisture condition. Uniaxial tensile tests on dumbbell-shaped SHCC specimens with in situ temperature control were performed at 22, 60 and 100 °C. In addition, the effect of the internal humidity of SHCC (95, 50, 20 and 0%) coupled to the elevated temperatures was investigated. It was shown that the tensile strength decreases and the strain capacity increases with an increase in temperature. The influence of the internal moisture conditions was more significant in high temperatures. The strain capacity reduced significantly with a decrease in the humidity level. The crack pattern of the SHCC specimens was determined. Furthermore, single fiber pullout tests were performed under the considered high temperatures condition. Finally, the results are discussed based on the thermogravimetry analysis of the PVA fiber, alterations on its microstructure and surface coating.  相似文献

A configurable process model captures a family of related process models in a single artifact. Such models are intended to be configured to fit the requirements of specific organizations or projects, leading to individualized process models that are subsequently used for domain analysis or solution design. This article proposes a formal foundation for individualizing configurable process models incrementally, while preserving correctness, both with respect to syntax and behavioral semantics. Specifically, assuming the configurable process model is behaviorally sound, the individualized process models are guaranteed to be sound. The theory is first developed in the context of Petri nets and then extended to a process modeling notation widely used in practice, namely Event-driven Process Chains.  相似文献

High performance processor designs have evolved toward architectures that integrate multiple processing cores on the same chip. As the number of cores inside a Chip MultiProcessor (CMP) increases, the interconnection network will have significant impact on both overall performance and energy consumption as previous studies have shown. Moreover, wires used in such interconnect can be designed with varying latency, bandwidth and power characteristics. In this work, we show how messages can be efficiently managed in tiled CMP, from the point of view of both performance and energy, by combining both address compression with a heterogeneous interconnect. In particular, our proposal is based on applying an address compression scheme that dynamically compresses the addresses within coherence messages allowing for a significant area slack. The arising area is exploited for wire latency improvement by using a heterogeneous interconnection network comprised of a small set of very-low-latency wires for critical short-messages in addition to baseline wires. Detailed simulations of a 16-core CMP show that our proposal obtains average improvements of 10% in execution time and 38% in the energy-delay2 product of the interconnect. Additionally, the sensitivity analysis shows that our proposal performs well when either OoO cores or caches with higher latencies are considered.  相似文献

Business process models are becoming available in large numbers due to their widespread use in many industrial applications such as enterprise and quality engineering projects. On the one hand, this raises a challenge as to their proper management: how can it be ensured that the proper process model is always available to the interested stakeholder? On the other hand, the richness of a large set of process models also offers opportunities, for example with respect to the re-use of existing model parts for new models. This paper describes the functionality and architecture of an advanced process model repository, named APROMORE. This tool brings together a rich set of features for the analysis, management and usage of large sets of process models, drawing from state-of-the art research in the field of process modeling. A prototype of the platform is presented in this paper, demonstrating its feasibility, as well as an outlook on the further development of APROMORE.  相似文献

A configurable process model provides a consolidated view of a family of business processes. It promotes the reuse of proven practices by providing analysts with a generic modeling artifact from which to derive individual process models. Unfortunately, the scope of existing notations for configurable process modeling is restricted, thus hindering their applicability. Specifically, these notations focus on capturing tasks and control-flow dependencies, neglecting equally important ingredients of business processes such as data and resources. This research fills this gap by proposing a configurable process modeling notation incorporating features for capturing resources, data and physical objects involved in the performance of tasks. The proposal has been implemented in a toolset that assists analysts during the configuration phase and guarantees the correctness of the resulting process models. The approach has been validated by means of a case study from the film industry.  相似文献

Mainstream business process modelling techniques often promote a design paradigm wherein the activities that may be performed within a case, together with their usual execution order, form the backbone on top of which other aspects are anchored. This Fordist paradigm, while effective in standardised and production-oriented domains, breaks when confronted with processes in which case-by-case variations and exceptions are the norm. We contend that the effective design of flexible processes calls for a substantially different modelling paradigm. Motivated by requirements from the human services domain, we explore the hypothesis that a framework consisting of a small set of coordination concepts, combined with established object-oriented modelling principles, provides a suitable foundation for designing highly flexible processes. Several human service delivery processes have been designed using this framework, and the resulting models have been used to realise a system to support these processes in a pilot environment.  相似文献
