Obesity-associated low-grade inflammation favors weight gain, whereas systemic infection frequently leads to anorexia. Thus, inflammatory signals can either induce positive or negative energy balance. In this study, we used whole-cell patch-clamp to investigate the acute effects of three important proinflammatory cytokines, tumor necrosis factor α (TNF-α), interleukin-6, and interleukin-1β (IL-1β) on the membrane excitability of agouti-related peptide (AgRP)- or proopiomelanocortin (POMC)-producing neurons. We found that both TNF-α and IL-1β acutely inhibited the activity of 35–42% of AgRP-producing neurons, whereas very few POMC neurons were depolarized by TNF-α. Interleukin-6 induced no acute changes in the activity of AgRP or POMC neurons. Our findings indicate that the effect of TNF-α and IL-1β, especially on the activity of AgRP-producing neurons, may contribute to inflammation-induced anorexia observed during acute inflammatory conditions.  相似文献

Six olive oils extracted from the cultivars Arbequina, Arbosana, Coratina, Frantoio, Koroneiki, and Picual from 2017 and 2018 harvests, cultivated in Pinheiro Machado, Rio Grande do Sul, Brazil, are evaluated for standard oil composition parameters and bioactive constituents (pigments, tocopherols, and phenolic compounds). Multivariate principal component analysis (PCA) and univariate ANOVA and Fisher's LSD test are used to verify the effect of cultivar and harvest year on oil composition. Olive oil composition met extra virgin olive oil (EVOO) standard parameters and is influenced by both cultivar and harvest year. EVOO produced in 2018 has greater chlorophyll, caffeic acid, ligstroside aglycone, hydroxyoleuropein aglycone, syringic acid, and hydroxytyrosol acetate contents than the EVOOs from 2017. Linoleic acid, ferulic acid, ligstroside aglycone, and hydroxytyrosol acetate are the variables whose contents most contributed to the differentiation of oils by cultivar in both harvest years. Chemical characterization analyses allow for the differentiation of oil composition based on harvest year and cultivar. Metabolic quality data obtained here support the establishment of a local EVOO profile and the compounds that most contributed to treatment differentiation may serve as markers that can be utilized in determining origin, cultivar, and harvest year. Practical Applications: Olive production in Brazil is recent and is based on European cultivars which have not been bred for the local environmental conditions. Therefore, the measurement of olive oil metabolic quality will determine cultivar adaptability to local edaphoclimatic conditions as well as assist in the establishment of a standard of identity for the product and promote the development of its market. Olive oil produced in Southern Brazil shows high quality, and is especially rich in phenolic compounds. Although harvest year influences oil composition, oil from both harvests meet EVOO standards and cultivar specific metabolic markers are observed. This study provides the foundation for olive producers in Southern Brazil to seek authentication of the geographical origin of olive oil.  相似文献

The use of toxic petroleum-based heavy oils is common in industrial processes. The cleaning of machines, equipment, and other surface covered in these oils is achieved with expensive products that are often also toxic and harmful to both the health of workers and the environment. The present paper proposes the development of a sustainable biodetergent made from plant-based materials. Tests were performed to determine the properties of the biodetergent in terms of its surfactant and emulsifying capacities, stability, toxicity, and the removal of heavy oil from glass plates and metallic surfaces. The formulation was composed of a natural solvent (cottonseed oil), a plant-based surfactant agent (saponin), and two natural stabilizers (carboxymethylcellulose and glycerine). The formulation was stable, nontoxic, and highly efficient, removing 100% of the heavy oil from glass and metallic surfaces. This solution developed in this study could be used in diverse industries with the need to clean machines and parts encrusted with oil and grease as well as the cleaning of floors covered with petroleum-based residues. A preliminary analysis of the economic feasibility of using the detergent was carried out at a Brazilian power plant. Besides the reduction in environmental impact due to the use of a nontoxic, biodegradable product as well as the reduction in health risks to workers associated with toxic cleaning products, this new product can have a considerable impact on the market as an environmentally friendly solution.  相似文献

Salmonella strains (n = 75) isolated from foods involved in foodborne outbreaks occurred in Rio Grande do Sul State, Brazil, during 1999 and 2000 were studied. Strains were serotyped and submitted to PCR analysis to verify the prevalence of Salmonella plasmid virulence (spvR) regulatory gene. Among the 75 isolates, 73 (97%) were classified as Salmonella enterica serovar Enteritidis. All of the Salmonella strains isolated in 1999 were classified as serotype Enteritidis, whereas in 2000 two isolates were serotyped as Salmonella Derby and Salmonella Typhimurium. Regarding the prevalence of spvR gene, 62 strains (82.7%) were PCR positive, and a positive correlation (P < 0.05) between the strains of Salmonella Enteritidis and the presence of spvR gene was demonstrated, which suggests that this gene is a characteristic of the Salmonella Enteritidis analyzed.  相似文献

Preheating potatoes at 50 to 80°C has a firming effect on the cooked potato tissue. This effect is particularly pronounced at a preheating temperature of 60 to 70°C followed by cooling. Several theories have been presented in the literature to explain this firming effect: retrogradation of starch, leaching of amylose, stabilization of the middle lamellae and cell walls by the activation of the pectin methylesterase (PME) enzyme, and by the release of calcium from gelatinized starch and the formation of calcium bridges between pectin molecules. Most probably, none of these theories alone can explain the phenomenon and more than one mechanism seems to be involved. Some of these mechanisms seem to be interdependent. As an example, calcium could be considered as a link all the way through release after starch gelatinization to cross‐linking pectin substances in the cell wall and the middle lamellae, which has been demethylated by the PME enzyme. More research and “clear cut” experiments are needed in order to elucidate the role of each mechanism, especially which of them is the main contributor to the process of firming. Most probably, the calcium‐pectin‐PME mechanism plays a secondary role, that is, it only retards the collapse of the tissue structure that would otherwise occur during the final heating without preheating, and it is not the main factor of firmness.  相似文献

A two dimensional model of bread baking was developed including, for the first time, the dependence of dough viscosity on both temperature and moisture content, the carbon dioxide dissolved from liquid water together with gas generation from yeast at the beginning of baking and the shrinkage due to dough drying. Particular attention was paid to experimental validation of both overall and local variables such as local temperature, overall mass loss, and local moisture content, overall CO2 released into the oven, and overall deformation and local expansion or shrinkage. This work presents useful composites for oil spill cleanup processes. These systems are composed of a polyester matrix loaded with coffee ground powder and maghemite. They were prepared by in situ polymerization. The aliphatic monomers proportion—castor oil and glycerin—was studied with the aim of understanding the effect of feed ratio on the product properties. The materials were studied using several techniques, including Fourier Transform Infrared Spectroscopy, Ultraviolet‐visible Spectrophotometry, and Wide Angle X‐ray Scattering, with magnetic force tests used for the characterization of materials. Density tests showed the presence of coffee grounds causes an important reduction in the density values of composites, improving their flotation. The interaction between composites and petroleum is more than twice that between composites and water. Moreover, for all magnetizable composites, the removal capability was (25.1 ± 1.2) g/g (petroleum/composite), allowing us to state that this is a promising material for use in oil spill cleanup processes. © 2015 Wiley Periodicals, Inc. This study aims to develop a dosage methodology based on tensile and compressive strength for artificially cemented fibre reinforced soils considering filament length. The controlling parameters evaluated were the fibre length (l), the cement content (C), the porosity (η) and the porosity/cement ratio (η/Civ). A number of unconfined compression and split tensile tests were carried out in the present work. The results show that fibre insertion in the cemented soil, for the whole range of cement content studied, and the increase of reinforcement length improve unconfined compressive and split tensile strengths. It was shown that the porosity/cement ratio, in which volumetric cementitious material content is adjusted by an exponent (0.28 for all the fibre-reinforced and non-reinforced cemented soil mixtures) to end in unique correlations for each mixture, is a good parameter in the evaluation of the unconfined compressive and split tensile strength of the fibre-reinforced and non-reinforced cemented soil studied. Analysis of variance (ANOVA) performed on the results of a factorial experiment considering the effect of adjusted cement content, fibre length and porosity showed that all of these factors are significant in affecting the measured changes in split tensile and unconfined compressive strength values. Finally, unique dosage relationships could be achieved linking the unconfined compressive strength (qu) and the split tensile strength (qt) of the sandy soil studied with porosity/cement ratio (η/Civ) and fibre length (l).  相似文献

Giardia spp. is a flagellate protozoan that presents two evolution forms, cysts and trophozoites. Cysts are resistant to chlorine, the most employed disinfectant agent in the treatment of water. For this reason, new techniques for the disinfection of waters that contain this parasite are necessary. This work evaluated the efficiency of the disinfection by ozone and ultrasound individually and simultaneously upon wastewater. The data obtained showed that after application, ozone, ultrasound, and combined techniques induced a significant elimination of Giardia spp. cysts. Furthermore, this effect was more accentuated when the two techniques were applied simultaneously.  相似文献
