Enzymatic transamidation of gliadins by microbial transglutaminase (mTG) inhibits interferon-γ (IFN-γ) secretion by intestinal T cell lines in patients with celiac disease (CD). To gain insight into the cellular mechanisms underlying the down-regulatory effects of transamidation, we tested a single recombinant α-gliadin (r-gliadin) harbouring two immunodominant peptides, p13 (aa. 120–139) and p23 (aa. 220–239), in HLA-DQ8 transgenic mice, a model of gluten sensitivity. Mice were intranasally immunised with r-gliadin or r-gliadin transamidated by mTG (K-r-gliadin) along with cholera toxin, and the response of mesenteric lymph node cells was analysed by cytokine multiplex assay. An in vitro challenge with r-gliadin was characterised by secretion of specific cytokines featuring both innate immunity and the Th1/Th2/Th17 pattern of the adaptive response. Notably, transamidation specifically down-regulated the Th1 response. Structural studies performed on K-r-gliadin confirmed that specific glutamine residues in p13 and p23, previously found to be deamidated by tissue transglutaminase, were also transamidated by mTG. In silico analysis, simulating p13 and p23 peptide binding to HLA-DQ8 showed that these glutamines, in the form of glutamate, could interact by means of salt bridges with peculiar amino acids of the alpha chain of HLA-DQ8, suggesting that their transamidation may influence the HLA-restricted recognition of these peptides. Thus, the structural findings provided a rationale to explain the down-regulation of the r-gliadin-specific Th1 response following transamidation.  相似文献

This study examined effects of reward and response costs on the ability of 19 attention-deficit hyperactivity disorder (ADHD) and 17 control children to inhibit responding. Children were tested under 4 reinforcement conditions on a go/no-go learning task developed by J. P. Newman, C. S. Widom, and S. Nathan (1985). Two conditions involved both reward and response costs, 1 response costs only, and 1 reward only. ADHD children made more commission errors than controls across the 4 conditions. Analyses of learning curves indicated that group differences became larger on later trials. Thus, impaired inhibition was more generalized in ADHD children than in the psychopaths and extraverts studied by Newman and colleagues, and it became most evident when the children were required to improve learning across trials. The photooxidation of poly[bis(4-isopropylphenoxy)phosphazene] under accelerated conditions has been followed by FTIR and UV visible spectroscopic techniques. The main photooxidation products are acetophenone and phenol groups. In addition, acetone vapors have been detected by GC MS combined techniques concomitant with the IR spectral changes in the CH stretching region, suggesting a significant decrease in the isopropyl moieties. The presence of polymeric sequences having phenol groups under our conditions gives origin to further oxidation reactions due to electron transfer or radical recombination or to hydrogen abstraction reactions promoted by unhindered phenoxyl radicals. In addition, the absence of UV visible light, i.e., under thermooxidation reaction at 60° C has demonstrated that phenol groups are the main responsible of secondary oxidation products. We propose a robust variational model for the restoration of images corrupted by blur and the general class of additive white noises. The key idea behind our proposal relies on a novel hard constraint imposed on the residual of the restoration, namely we characterize a residual whiteness set to which the restored image must belong. As the feasible set is unbounded, solution existence results for the proposed variational model are given. Moreover, based on theoretical derivations as well as on Monte Carlo simulations, we provide well-founded guidelines for setting the whiteness constraint limits. The solution of the non-trivial optimization problem, due to the non-smooth non-convex proposed model, is efficiently obtained by an alternating directions method of multipliers, which in particular reduces the solution to a sequence of convex optimization subproblems. Numerical results show the potentiality of the proposed model for restoring blurred images corrupted by several kinds of additive white noises.  相似文献

The present study was designed to test the hypothesis that atypical, but not typical, antipsychotics produce a functional in vivo blockade of 5-HT2A receptors. The magnitude of functional in vivo 5-HT2A receptor blockade elicited by representative compounds from each of the six major structural classes of typical antipsychotics, and the representative atypical antipsychotics clozapine and risperidone, was indicated by their respective abilities to block the stimulus effects of the phenylalkylamine hallucinogen (-)DOM in the rat. Chlorpromazine, thioridazine, fluphenazine, thiothixene and haloperidol did not produce a significant antagonism of the (-)DOM stimulus. The benzoxapine, loxapine (60%), and the atypical dibenzodiazepine, clozapine (62%), partially blocked and risperidone fully blocked (100%) the (-)DOM stimulus. None of these agents elicited significant levels of (-)DOM-appropriate responding when administered alone. These results indicate that the typical antipsychotics, with the exception of lozapine, fail to produce effective in vivo antagonism of 5-HT2A receptors at doses compatible with the preservation of operant behavior. In contrast, the atypical antipsychotics clozapine and risperidone elicit effective in vivo antagonism of 5-HT2A receptors without severe behavioral disruption. Thus, these data are supportive of the hypothesis that the mechanism of action of atypical, but not typical, antipsychotics involves the antagonism of 5-HT2A receptors in vivo.  相似文献

Although tomato flavor has not been a major goal for breeders, nowadays it becomes important as it is a subject of consumer complaint. A better knowledge of tomato consumer preferences, at the European level, should provide the basis for improvement of fruit quality and for market segmentation. In the framework of a large European project, 806 consumers from 3 countries, The Netherlands, France, and Italy, were presented with a set of 16 varieties representing the diversity of fresh tomato offer in order to evaluate their preferences. In parallel, sensory profiles were constructed by expert panels in each country. Preference maps were then constructed in each country revealing the structure of consumer preferences and allowing identification of the most important characteristics. Then a global analysis revealed that preferences were quite homogeneous across countries. This study identified the overall flavor and firmness as the most important traits for improving tomato fruit quality. It showed that consumer preferences from different European countries, with different cultures and food practices, are segmented following similar patterns when projected onto a common referential plan. Moreover, the results clearly showed that diversification of taste and texture is required to satisfy all consumers' expectations as some consumers preferred firm tomatoes, while others preferred melting ones and were more or less demanding in terms of sweetness and flavor intensity. Detailed comparisons also showed the importance of the fruit appearance in consumer preference.  相似文献
