Ethanol is one of the most widespread psychotropic agents in western society. While its psychoactive effects are mainly associated with GABAergic and glutamatergic systems, the positive reinforcing properties of ethanol are related to activation of mesolimbic dopaminergic pathways resulting in a release of dopamine in the nucleus accumbens. Given these neurobiological implications, the detection of ethanol in brain extracellular fluid (ECF) is of great importance. In this study, we describe the development and characterization of an implantable biosensor for the amperometric detection of brain ethanol in real time. Ten different designs were characterized in vitro in terms of Michaelis-Menten kinetics (V(MAX) and K(M)), sensitivity (linear region slope, limit of detection (LOD), and limit of quantification (LOQ)), and electroactive interference blocking. The same parameters were monitored in selected designs up to 28 days after fabrication in order to quantify their stability. Finally, the best performing biosensor design was selected for implantation in the nucleus accumbens and coupled with a previously developed telemetric device for the real-time monitoring of ethanol in freely moving, untethered rats. Ethanol was then administered systemically to animals, either alone or in combination with ranitidine (an alcohol dehydrogenase inhibitor) while the biosensor signal was continuously recorded. The implanted biosensor, integrated in the low-cost telemetry system, was demonstrated to be a reliable device for the short-time monitoring of exogenous ethanol in brain ECF and represents a new generation of analytical tools for studying ethanol toxicokinetics and the effect of drugs on brain ethanol levels.  相似文献

Shelf life extension of fresh fruit and vegetables by chitosan treatment   总被引:1,自引:0,他引:1  
Among alternatives that are currently under investigation to replace the use of synthetic fungicides to control postharvest diseases in fresh produce and to extend their shelf life, chitosan application has shown promising disease control, at both preharvest and postharvest stages. Chitosan shows a dual mode of action, on the pathogen and on the plant, as it reduces the growth of decay-causing fungi and foodborne pathogens and induces resistance responses in the host tissues. Chitosan coating forms a semipermeable film on the surface of fruit and vegetables, thereby delaying the rate of respiration, decreasing weight loss, maintaining the overall quality, and prolonging the shelf life. Moreover, the coating can provide a substrate for incorporation of other functional food additives, such as minerals, vitamins, or other drugs or nutraceutical compounds that can be used to enhance the beneficial properties of fresh commodities, or in some cases the antimicrobial activity of chitosan. Chitosan coating has been approved as GRAS substance by USFDA, and its application is safe for the consumer and the environment. This review summarizes the most relevant and recent knowledge in the application of chitosan in postharvest disease control and maintenance of overall fruit and vegetable quality during postharvest storage.  相似文献

Metal shear panels represent a highly performing anti seismic system for medium–high rise buildings located in strong earthquake prone zones. A new configuration of dissipative shear panel is presented in the current paper. The base shear plate is made of a low-strength material, namely the heath treated EN-AW-1050A aluminium alloy, which has been already used by the authors for other types of dissipative shear panels. In this proposed new system, the main buckling modes are inhibited by the application of external devices placed in parallel to the base plate and not directly loaded by the external forces. Two shear panel samples are tested as prototypes, which are based on two different plate buckling inhibition strategies. In the former the out-of-plane displacements of only some parts of the base plate are restrained, whereas in the latter the restraining action of the external device is extended to the whole base plate. Experimental tests are presented in order to prove the performance of the proposed devices in terms of dissipative capacity and stiffness. In addition, the obtained results are compared with the ones related to other more conventional shear panel typologies, characterized by the same geometry and material, but in which buckling phenomena were mitigated by means of welded stiffeners, according to a more traditional practice.  相似文献

The CONSEN (CONServation of ENergy) code is a fast running code to simulate thermal-hydraulic transients, specifically developed for fusion reactors. In order to demonstrate CONSEN capabilities, the paper deals with the accident analysis of the magnet induced confinement bypass for ITER design 1996. During a plasma pulse, a poloidal field magnet experiences an over-voltage condition or an electrical insulation fault that results in two intense electrical arcs. It is assumed that this event produces two one square meters ruptures, resulting in a pathway that connects the interior of the vacuum vessel to the cryostat air space room. The rupture results also in a break of a single cooling channel within the wall of the vacuum vessel and a breach of the magnet cooling line, causing the blow down of a steam/water mixture in the vacuum vessel and in the cryostat and the release of 4 K helium into the cryostat. In the meantime, all the magnet coils are discharged through the magnet protection system actuation. This postulated event creates the simultaneous failure of two radioactive confinement barrier and it envelopes all type of smaller LOCAs into the cryostat. Ice formation on the cryogenic walls is also involved. The accident has been simulated with the CONSEN code up to 32 h. The accident evolution and the phenomena involved are discussed in the paper and the results are compared with available results obtained using the MELCOR code.  相似文献

Consumers are increasingly aware of the importance of regular consumption of fresh fruit in their diet. Since fresh fruit are highly sensitive to postharvest decay, several investigations focused on the study natural compounds alternative to synthetic fungicides, to extend their shelf life. A long list of studies reported the effectiveness of the natural biopolymer chitosan in control of postharvest diseases of fresh fruit. However, these findings remain controversial, with many mixed claims in the literature. In this work, we used random-effects meta-analysis to investigate the effects of 1% chitosan on (a) postharvest decay incidence; (b) mycelium growth of fungal pathogens Botrytis cinerea, Penicillium spp., Colletotrichum spp. and Alternaria spp.; and (c) phenylalanine ammonia-lyase, chitinase and β-1,3-glucanase activities. Chitosan significantly reduced postharvest disease incidence (mean difference [MD], −30.22; p < 0.00001) and in vitro mycelium growth (MD, −54.32; p  < 0.00001). For host defense responses, there were significantly increased activities of β-1,3-glucanase (MD, 115.06; p = 0.003) and chitinase (MD, 75.95; p < 0.0002). This systematic review contributes to confirm the multiple mechanisms of mechanisms of action of chitosan, which has unique properties in the natural compound panorama. Chitosan thus represents a model plant protection biopolymer for sustainable control of postharvest decay of fresh fruit.  相似文献

In this work the fragmental approach was used to prepare several molecularly imprinted ethylene dimethacrylate-co-methacrylic acid polymers with molecular recognition towards the mycotoxin ochratoxin A, with the aim of searching for simpler mimic templates than the well-known N-(4-chloro-1-hydroxy-2-naphthoylamido)-(l)-phenylalanine. The screening for binding of two different kinds of ochratoxin-related molecules was performed by HPLC analysis. Ochratoxin A and the mimic templates were eluted in acetonitrile–acetic acid (0.1% v/v) and the imprinting factor was measured for all the ligands on all the columns packed with the imprinted polymers. The experimental results show that changes to the amino acidic sub-structure or the presence/absence of a chlorine atom in position 4 on the naphthalene ring system does not affect the molecular recognition of ochratoxin A by the resulting imprinted polymer. On the contrary, the presence of the bulky naphthalene ring system in the mimic template seems to be necessary to preserve the molecular recognition of ochratoxin. This binding behavior was found to be compatible with in silico simulations of the complexation between some of the mimic templates and molecules of methacrylic acid. The use of the mimic template N-(1-hydroxy-2-naphthoylamido)-(L)-phenylalanine seems to represent a synthetically simple approach to the preparation of imprinted polymers with molecular recognition properties towards ochratoxin A.  相似文献

We report the detection of Listeria spp. and Listeria monocytogenes in 34 dairy plants. In total, 547 of food, product contact surface and floor drain samples were collected along the product lines. Nineteen cheese factories (55.8%) were contaminated by Listeria spp. Of these 20.6% were L. monocytogenes positive. Listeria spp. was found in 6.8% of food samples, 11.3% of product contact surfaces and 40.6% of floor drains. L. monocytogenes was found in 2.4% of food samples, 4.9% of product contact surfaces and 18.8% of floor drains. Twentyfive L. monocytogenes isolates were serotyped using commercial specific antisera and genotyped using Amplified Fragment Length Polymorphism (AFLP) analysis. AFLP genotyping discriminated the four species of Listeria isolated and different genotypes for each species, moreover it could identify persistent genotypes in some dairy facilities. Listeria spp. and L. monocytogenes are widely spread in the dairy sector and probably contaminate foods during the production process. Facility-based monitoring can identify possible routes of transmission and thus allow establishment of more effective strategies to prevent food contamination.  相似文献

This paper provides a set of specific examples to show the effectiveness of the trigeneration CO2emission reduction (TCO2ER) indicator proposed in the companion paper (Part I: Models and indicators) to assess the greenhouse gas (GHG) emission reduction from cogeneration and trigeneration systems. Specific break-even analyses are developed by introducing further indicators, with the aim of assessing the conditions for which different types of combined systems and conventional separate production systems are equivalent in terms of GHG emissions. The various emission indicators are evaluated and discussed for a number of relevant application cases concerning cogeneration and trigeneration solutions with different types of equipment. Scenario analyses are carried out to assess the possible emission reduction benefits from extended diffusion of cogeneration and trigeneration in regions characterized by different energy generation frameworks. The results strongly depend on the available technologies for combined production, on the composition of the energy generation mix, and on the trend towards upgrading the various generation systems. The numerical outcomes indicate that cogeneration and trigeneration solutions could bring significant benefits in countries with prevailing electricity production from fossil fuels, quantified by the use of the proposed indicators.  相似文献

The precise connections between the Barkhausen effect and non-equilibrium critical phenomena and self-organized criticality (SOC) have been strongly debated in the past few years. We review the recent theoretical studies of the Barkhausen effect and discuss their relevance to quantitatively understand the experiments. We compare the results of disordered spin models, micromagnetic equations, and models for rigid and flexible domain walls with experiments on amorphous and polycrystalline ferromagnetic alloys. In particular, we discuss the theoretical predictions for the critical exponents of the power law decay and for the cutoff of the avalanche distributions. We conclude that models based on the dynamics of a flexible domain wall are the most successful in explaining existing experimental data.  相似文献
