Low-grade chronic inflammation plays a pivotal role in the pathogenesis of insulin resistance (IR), and skeletal muscle has a central role in this condition. NLRP3 inflammasome activation pathways promote low-grade chronic inflammation in several tissues. However, a direct link between IR and NLRP3 inflammasome activation in skeletal muscle has not been reported. Here, we evaluated the NLRP3 inflammasome components and their role in GLUT4 translocation impairment in skeletal muscle during IR. Male C57BL/6J mice were fed with a normal control diet (NCD) or high-fat diet (HFD) for 8 weeks. The protein levels of NLRP3, ASC, caspase-1, gasdermin-D (GSDMD), and interleukin (IL)-1β were measured in both homogenized and isolated fibers from the flexor digitorum brevis (FDB) or soleus muscle. GLUT4 translocation was determined through GLUT4myc-eGFP electroporation of the FBD muscle. Our results, obtained using immunofluorescence, showed that adult skeletal muscle expresses the inflammasome components. In the FDB and soleus muscles, homogenates from HFD-fed mice, we found increased protein levels of NLRP3 and ASC, higher activation of caspase-1, and elevated IL-1β in its mature form, compared to NCD-fed mice. Moreover, GSDMD, a protein that mediates IL-1β secretion, was found to be increased in HFD-fed-mice muscles. Interestingly, MCC950, a specific pharmacological NLRP3 inflammasome inhibitor, promoted GLUT4 translocation in fibers isolated from the FDB muscle of NCD- and HFD-fed mice. In conclusion, we found increased NLRP3 inflammasome components in adult skeletal muscle of obese insulin-resistant animals, which might contribute to the low-grade chronic metabolic inflammation of skeletal muscle and IR development. 相似文献
We present a method to obtain reduced order models to calculate steady states in thermal systems of industrial interest. The method could be regarded as an evolution of the standard proper orthogonal decomposition (POD) method, in which the reduced model is obtained through standard Galerkin projection (whose application exhibits well known difficulties) on previously obtained POD modes. Instead of relying on a Galerkin projection, we use a genetic algorithm (GA) to minimize a conveniently defined residual for the (continuity, momentum, and energy) equations and boundary conditions. The method and its practical application are illustrated on a test problem that describes heat transfer in the recirculation region downstream of a backwards facing step. 相似文献
A novel DNA damage detection technique based on the characteristic fluorescence lifetimes exhibited by Pico-Green-single-stranded DNA and -double-stranded DNA complexes is employed to establish the damage produced on DNA isolated from sheep white blood cells following gamma radiation. This technique, which incorporates key concepts such as alkaline unwinding buffers and higher unwinding rates of damaged DNA compared to undamaged DNA, allows for the differentiation of DNA damage resulting from doses of gamma radiation in the 0-100-Gy range, with the potential of analyzing samples consisting of as little as 10(4) cells. Experiments were carried out using commercial DNA sources as well as DNA isolated from sheep white blood cells, suggesting its potential for use with isolated DNA from virtually any eukaryotic cell. 相似文献
Ensembles that combine the decisions of classifiers generated by using perturbed versions of the training set where the classes of the training examples are randomly switched can produce a significant error reduction, provided that large numbers of units and high class switching rates are used. The classifiers generated by this procedure have statistically uncorrelated errors in the training set. Hence, the ensembles they form exhibit a similar dependence of the training error on ensemble size, independently of the classification problem. In particular, for binary classification problems, the classification performance of the ensemble on the training data can be analysed in terms of a Bernoulli process. Experiments on several UCI datasets demonstrate the improvements in classification accuracy that can be obtained using these class-switching ensembles. 相似文献
This work addresses the resource sharing problem in broadband communication networks that can guarantee some quality of service (QoS), and develops some results about data source and traffic modelling, especially in aspects of model testing and parameter estimation. The multiplexing of variable bit rate (VBR) sources poses a mathematical and statistical problem: the estimation of the resource requirements of a source or set of sources. The estimation method shall be simple enough to be practically implemented in the connection acceptance control (CAC) function.
In this paper, the VBR video sources are taken as a typical case of variable rate, with real-time constraints. This association of requirements makes the case especially interesting. A Markov model is assumed for the VBR sources. The validity of such models is under research; they seem to be appropriate at least in certain time scales. The model is tested against real video traces. In order to estimate the resource allocation or “channel occupation” of each source, the concept of equivalent bandwidth proposed by Kelly [Notes on effective bandwidth, in: F.P. Kelly, S. Zachary, I.B. Ziedins (Eds.), Stochastic Networks: Theory and Applications, Oxford University Press, Oxford, 1996, pp. 141] is used; it is based on a consistent mathematical theory, and has proven to be robust and useful for technical applications.
A calculation of the equivalent bandwidth of a Markov source, given its parameters, can be found in the literature [IEEE ACM Trans. Networking 1 (4) (1993) 424]. But in fact, one can only estimate model and parameters. In this work, an estimation of the equivalent bandwidth is given, which can be obtained from real data. The convergence and the consistency of the estimation are studied, and practical bounds are found. Illustrative calculations are performed from real video traces that were obtained using a software MPEG coder, developed by the authors. The mathematical and statistical results are valid for whatever phenomenon that can be modelled as a Markov process. 相似文献
We use the point spread function and the modulation transfer function (MTF) as two figures-of-merit to evaluate the performance of the multiaperture interferometric configurations for the detection of a faint planet in the vicinity of its bright star. We design nonredundant interferometric layouts that provide satisfactory coverage of the spatial frequencies of interest. We propose a design incorporating a rotating, rotationally shearing interferometer in a gravity-free environment and compare its performance with the Earth-based, fixed, linear configurations. The side peak of its MTF may be centered on the coordinate associated with a likely planet spatial frequency, resulting in planet signal enhancement and isolation. 相似文献
We study the characteristics and radiation mechanism of antenna superstrates based on closely located periodical grids of loaded wires. An explicit analytical method based on the local field approach is used to study the reflection and transmission properties of such superstrates. It is shown that as a result of proper impedance loading there exists a rather wide frequency band over which currents induced to the grids cancel each other, leading to a wide transmission maximum. In this regime radiation is produced by the magnetic dipole moments created by circulating out-of-phase currents flowing in the grids. An impedance matrix representation is derived for the superstrates, and the analytical results are validated using full-wave simulations. As a practical application example we study numerically the radiation characteristics of dipole antennas illuminating finite-size superstrates. 相似文献
Automatic ultrasound (US) image segmentation is a difficult task due to the quantity of noise present in the images and the lack of information in several zones produced by the acquisition conditions. In this paper, we propose a method that combines shape priors and image information to achieve this task. In particular, we introduce knowledge about the rib-eye shape using a set of images manually segmented by experts. A method is proposed for the automatic segmentation of new samples in which a closed curve is fitted taking into account both the US image information and the geodesic distance between the evolving curve and the estimated mean rib-eye shape in a shape space. This method can be used to solve similar problems that arise when dealing with US images in other fields. Oil (healthier lipid combination of olive, linseed and fish oils)-in-water emulsions stabilized with different protein systems (prepared with sodium caseinate (SC), soy protein isolate (SPI), and microbial transglutaminase (MTG)) were used as pork backfat replacers in low-fat frankfurters. Microstructure, lipid oxidation, nitrite content, microbiological changes and biogenic amine formation of frankfurters were analyzed and found to be affected by the type of oil-in-water emulsion and by chilling storage (2° C, 41 days). Although the lipid oxidation levels attained were low, replacement of animal fat by healthier oil combinations in frankfurter formulation did promote a slight increase in lipid oxidation. Residual nitrite was affected (P < 0.05) by formulation and storage. Only 51-61% of the added nitrite was detectable in the product after processing and 17-46% at the end of storage. The microbial population was low in all formulations during chilling storage. Spermine was the most abundant amine (19-20 mg/kg), but similar in level to all samples. 相似文献
