The medial Golgi enzymes, N-acetylglucosaminyltransferase I (NAGT I) and mannosidase II (Mann II), and the trans Golgi enzyme, beta-1,4-galactosyltransferase (GalT) were each retained in the endoplasmic reticulum (ER) by grafting on the cytoplasmic tail of the p33 invariant chain. Transient and stable expression of p33/NAGT I in HeLa cells caused relocation of endogenous Mann II to the ER and transient expression of p33/Mann II had a similar effect on endogenous NAGT I. Neither of these endogenous medial enzymes were affected by transient expression of p33/GalT. These data provide strong evidence for kin recognition between medial Golgi enzymes and suggest a role for them in the organization of the Golgi stack.  相似文献

We have analyzed the V-gene usage in gamma delta T cells of the human gut and joint by using a new mAb (B18) specific for V gamma 8 of human TCR-gamma delta+ T cells. The B18+ population constituted a minor subset of the gamma delta T cells in peripheral blood (PB) of healthy persons (6 +/- 5%) and only 1 of 35 gamma delta T cell clones analyzed was positive. In contrast, the B18+ subset was a dominant gamma delta T cell population among intraepithelial lymphocytes (IEL) derived from the human intestine (74 +/- 29, p < 0.002), and two of three IEL clones from patients with coeliac disease were B18+. Interestingly, a higher proportion of B18+ gamma delta T cells was found in the synovial fluid of patients with rheumatoid arthritis (RA) (21 +/- 18%, 0.02 < p < 0.05) compared with normal PB. Furthermore, the B18+ subset was more frequent among IL-2-expanded gamma delta T cells (42 +/- 20%) derived from synovial tissue than among IL-2-expanded cells derived from synovial fluid (p < 0.002) and PB from RA patients (p < 0.02) as well as normal PB (p < 0.002). The V-gene usage of 13 gamma delta T cell clones from the synovial fluid of arthritic patients was analyzed. All B18+ clones (n = 7) expressed mRNA for V gamma 8 together with mRNA for V delta 1 (n = 5) or mRNA for V delta 3 (n = 2). None of the B18- clones expressed V gamma 8 (n = 6). We conclude that the gamma delta T cell that expresses V gamma 8, together with mainly V delta 1, is a major gamma delta T cell subset among the IEL of the gut and a highly frequent subset in the synovial tissue of patients with RA. This subset may correspond to the mouse V gamma 7+ IEL, which has a high degree of amino acid sequence homology with the human V gamma 8 protein.  相似文献

The distribution of barium in the mouse has been determined qualitatively by whole-body autoradiography after i.v. administration of 133BaCl2 solution. The quantitative distribution of 104Ba and 140La has been analyzed after i.p. injections of the two nuclides in equilibrium by measuring the activity in excised organs in a two-channel scintillation counter. Approximate doses to the eyes and different parts of the skeleton have been calculated.  相似文献

Antiserum to fentanyl was obtained in rabbits repeatedly injected with carboxyfentanyl conjugated to bovine serum albumin. Using the antiserum, a highly sensitive radioimmunoassay has been developed, based on the dextran-coated charcoal method. It proved possible to assay the drug directly in plasma, in amounts as small as 30 picogram in 0.5 ml. The antibody was highly specific for fentanyl and no cross-reaction was observed with its major metabolites. This sensitive and specific radioimmunoassay method was employed to determine fentanyl in plasma from six volunteers after an intravenous bolus of 0.2 mg, and in plasma from dogs treated both intravenously and subcutaneously with 0.02 mg/kg. The plasma level of fentanyl could be followed for up to 6 h after a therapeutic dose in dogs and man.  相似文献

Failure in sheet metal structures of ductile material is usually caused by one of, or a combination of, ductile fracture, shear fracture or localised instability. In this paper the failure of the high strength steel Docol 600DP and the ultra high strength steel Docol 1200M is explored. The constitutive model used in this study includes plastic anisotropy and mixed isotropic-kinematic hardening. For modelling of the ductile and shear fracture the models presented by Cockroft–Latham and Bressan–Williams have been used. The instability phenomenon is described by the constitutive law and the finite element (FE) models. For calibration of the failure models and validation of the results, an extensive experimental series has been conducted including shear tests, plane strain tests and Nakajima tests. The geometries of the Nakajima tests have been chosen so that the first quadrant of the forming limit diagram (FLD) were covered. The results are presented both in an FLD and using prediction of force–displacement response of the Nakajima test employing element erosion during the FE simulations. The classical approach for failure prediction is to compare the principal plastic strains obtained from FE simulations with experimental determined forming limit curves (FLCs). It is well known that the experimental FLC requires proportional strains to be useful. In this work failure criteria, both of the instability and fracture, are proposed which can be used also for non-proportional strain paths.  相似文献

The acoustic field radiation from an impulsive source located in a discretely layered and laterally bounded fluid is investigated with the aid of the generalized-ray theory. The main ingredient of the presented approach is the Cagniard–de Hoop technique in combination with the method of images that provide exact space–time expressions for the activated acoustic wavefield components.
