The activity of human liver microsomal cytochrome P450 1A2 (CYP1A2) is readily estimated by following the O-deethylation of [O-ethyl 14C]phenacetin (PODase). The basis of the assay is the quantitative measurement of [14C]acetaldehyde, remaining in the supernatant of assay incubates, after extraction of unmetabolized [O-ethyl 14C]phenacetin with charcoal. In the presence of native human liver microsomes (K(m) = 54 +/- 27 microM; V(max) = 14 +/- 2.3 nmol/hr/mg; mean +/- SD; N = 3 different livers) and human B-lymphoblastoid cell microsomes containing cDNA-expressed CYP1A2 (K(m) = 46 microM; V(max) = 55 nmol/hr/nmol CYP), PODase activity conformed to monophasic Michaelis-Menten kinetics. Furthermore, PODase activity in a panel of microsomes prepared from a series of human livers was significantly correlated (r = 0.91; p < 0.001; N = 11) with CYP1A2-selective 7-ethoxyresorufin O-deethylase activity, and was markedly inhibited (> or = 92%) by furafylline (FURA, IC50 = 0.4 microM) and 7,8-benzoflavone (ANF, IC50 = 0.1 microM), two well known CYP1A2 inhibitors. Inhibitors selective for other forms of CYP (e.g. CYP3A, CYP2C, CYP2D6, CYP2E1) elicited a marginal effect (< or = 17% inhibition) at relatively high concentrations (> or = 10.K(i)). It is concluded that the inhibition of human liver microsomal CYP1A2 activity can be readily determined by using a charcoal-based radiometric method employing [O-ethyl 14C]phenacetin as substrate. 相似文献

Objective: Successful weight management relies on at least two health behaviors, eating and exercise. However, little is known about their interaction on a motivational and behavioral level. Based on the Hierarchical Model of Motivation the authors examined whether exercise-specific motivation can transfer to eating regulation during a lifestyle weight control program. The authors further investigated whether general, treatment-related, and exercise motivation underlie the relation between increased exercise and improved eating regulation. Design: Overweight/obese women participated in a 1-year randomized controlled trial (N = 239). The intervention focused on promoting physical activity and internal motivation for exercise and weight loss, following Self-Determination Theory. The control group received general health education. Main Outcome Measures: General and exercise specific self-determination, eating self-regulation variables, and physical activity behavior. Results: General self-determination and more autonomous exercise motivation predicted eating self-regulation over 12 months. Additionally, general and exercise self-determination fully mediated the relation between physical activity and eating self-regulation. Conclusion: Increased general self-determination and exercise motivation seem to facilitate improvements in eating self-regulation during weight control in women. The possibility of using the Syst@me Probatoire de l'Observation de la Terre (SPOT)-VEGETATION (VGT) data for global burned area mapping with a single algorithm was investigated. Using VGT images from south-eastern Africa, the Iberian Peninsula and south-eastern Siberia/north-eastern China, we analysed the variability of the spectral signature of burned areas and its relationship with land cover, and performed the selection of the best variables for burned area mapping. The results show that in grasslands and croplands, near-infrared (NIR) and short-wave infrared (SWIR) reflectance always decreases as a result of fire. In forests and woodlands, there may occur a simultaneous decrease of SWIR and NIR or an increase of SWIR and a decrease of NIR. Burning of green vegetation (high values of the Normalized Difference Vegetation Index (NDVI)) tends to result in an increase of the SWIR. The best variables for burned area mapping are different in each region. Only the NIR allows a good discrimination of burned areas in all study areas. We derived a logistic regression model for multi-temporal burned area mapping in tropical, temperate and boreal regions, which handles the spectral variability of burned areas dependent on the type of vegetation. A non-parametric estimator of the AIDS survival time (after developing AIDS) is computed for the AIDS data set from the US Air Force (USAF). Survival times are unobservable. They are censored by the screening mechanism. The Armstrong Laboratory's Epidemiologic Research Division maintains data on over 954 active duty US Air Force (USAF) individuals who tested positive for human immunodeficiency virus (HIV) antibodies. Many have been clinically evaluated seven times since 1986. The HIV-positive individual is classified in seven stages of the disease complex as time progresses. Exact times of transition from one stage to the next are unknown. It is known that transition occurred between two consecutive evaluations. The aim of this study is to analyse distributions of the times that individuals spend in each stage of the HIV disease complex. We will discuss methods used to obtain non-parametric estimators of the distribution of times that individuals spend in stage 6. Finally, it is hoped to model the median time spent in each stage of the disease. This, along with incidence and separation data, will allow the prediction of the impact of HIV disease on USAF individuals and medical care systems. 相似文献

Primary cell cultures of mouse ventricular myocardium were infected with Trypanosoma cruzi, to study the consequences of T. cruzi-muscle cell interaction on the rate of spontaneous contractions, on the responses to norepinephrine, and on action potential parameters. Single cells or small cell groups of infected cultures were subjected to pharmacological and electrophysiological experiments. In concentrations ranging from 1 nM to 100 microM, norepinephrine exerted positive or negative chronotropic effects mediated by alpha-adrenergic receptors. A significant number of infected cells (25%) did not respond to the agonist. Two days after infection the cultures exhibited a higher frequency of spontaneous contractions (20%), paralleled by an increase in firing rate and a decrease in the action potential duration without significant changes in maximum diastolic potential and action potential amplitude. A decrease in alpha-adrenergic receptor-mediated positive chronotropic response to norepinephrine was also observed in 2-day infected cells. Cells made to phagocyte ferritin particles showed an increase in the rate of spontaneous contractions, but no changes in the positive chronotropic responses to norepinephrine. In conclusion, these observations show that during acute infection with T. cruzi, there are alterations in automaticity and in the chronotropic responses to norepinephrine, whose mechanisms are related to the process of parasite endocytosis by the cardiac cells. 相似文献

The lithium extraction from a lepidolite concentrate using roasting, followed by water leaching, was studied. Several alternative additives were initially tested. The use of sodium and calcium sulfates as additives was evaluated in more detail. The influence of some process variables, namely the roasting time, roasting temperature and the additive/concentrate mass ratio, was studied applying a design of experiments. The lithium extraction was modelled and the fitted and validated model was used to optimize the process response. The increase in the additive/concentrate mass ratio, roasting time and temperature seems to result in solid state reactions and transformations that lead to phase, morphological and particle size distribution modifications, which were assessed by XRPD, SEM, and particle size analyses. In this process, lithium sodium sulfate formation constitutes a crucial step enabling the Li water leaching. High lithium extractions were estimated for several combinations of factors. At 850°C, lithium extractions over 90% are obtained when the roasting time is above 1.90 hour and the additive/concentrate mass ratios are over 0.77. An increase in the temperature to 875°C also leads to lithium extractions over 90% for a roasting time of 1 hour and an additive/concentrate mass ratio of 0.60. 相似文献

A Portuguese kindred with autosomal dominant isolated primary hyperparathyroidism (HPT) that was associated with parathyroid adenomas and carcinomas was investigated with the aim of determining the chromosomal location of this gene, designated HPTPort. Leukocyte DNA from 9 affected and 16 unaffected members and 7 parathyroid tumors from 4 patients was used in comparative genomic hybridization (CGH), tumor loss of heterozygosity (LOH), and family linkage studies. The CGH studies revealed abnormalities of chromosomes 1 and 13, and the results of LOH studies were consistent with the involvements of tumor suppressor genes from these regions. Family segregation studies mapped HPTPort to chromosome 1q22-q31 by establishing linkage with eight loci (D1S254, D1S222, D1S202, D1S238, D1S428, D1S2877, D1S422, and D1S412) (peak two-point LOD scores = 3. 46-5.14 at 0% recombination), and defined the location of HPT Port to a 21 cM region flanked centromerically by D1S215 and telomerically by D1S306. Thus, HPTPort has been mapped to chromosome 1q22-q31, and a characterization of this gene will help to elucidate further the mechanisms that are involved in the development of parathyroid tumors. 相似文献

In this work, event related potentials (ERPs) induced by visual stimuli categorized with different value of affective valence are studied. EEG signals are recorded during visualization of selected pictures belonging to International Affective Picture System (IAPS). A Morlet wavelet filter is used to transform the EEG input space to a topography-time–frequency feature space. Support vector machine-recursive feature elimination (SVM-RFE) is applied for detecting scalp spectral dynamics of interest (SSDOIs) in this feature space, allowing to identify the most relevant time intervals, frequency bands and EEG channels. This feature selection method has proven to outperform the classical t-test in the discrimination of brain cortex regions involved in affective valence processing. Furthermore, the presented combination of feature extraction and selection techniques can be applied as an alternative in other different clinical applications. 相似文献
