The role of inhibition of return (IOR) in serial visual search was reinvestigated using R. Klein's (1988) paradigm of a search task followed by a probe-detection task. Probes were presented at either the location of a potentially inhibited search distractor or an empty location. No evidence of IOR was obtained when the search objects were removed after the search-task response. But when the search objects remained on, a pattern of effects similar to Klein's results emerged. However, when just the search-critical object parts were removed or when participants received immediate error feedback to prevent rechecking of the search objects, IOR effects were observed only when probes appeared equally likely at search array and empty locations. These results support the operation of object-based IOR in serial visual search, with IOR demonstrable only when rechecking is prevented (facilitating task switching) and monitoring for probes is not biased toward search objects. The Saccharomyces cerevisiae Dnm1 protein is structurally related to dynamin, a GTPase required for membrane scission during endocytosis. Here we show that Dnm1p is essential for the maintenance of mitochondrial morphology. Disruption of the DNM1 gene causes the wild-type network of tubular mitochondrial membranes to collapse to one side of the cell but does not affect the morphology or distribution of other cytoplasmic organelles. Dnm1 proteins containing point mutations in the predicted GTP-binding domain or completely lacking the GTP-binding domain fail to rescue mitochondrial morphology defects in a dnm1 mutant and induce dominant mitochondrial morphology defects in wild-type cells. Indirect immunofluorescence reveals that Dnm1p is distributed in punctate structures at the cell cortex that colocalize with the mitochondrial compartment. These Dnm1p-containing structures remain associated with the spherical mitochondria found in an mdm10 mutant strain. In addition, a portion of Dnm1p cofractionates with mitochondrial membranes during differential sedimentation and sucrose gradient fractionation of wild-type cells. Our results demonstrate that Dnm1p is required for the cortical distribution of the mitochondrial network in yeast, a novel function for a dynamin-related protein. 相似文献

This study seeks to assess the potential use of the CIEDE2000 color difference equation in wood science. Therefore, a set of oak wood samples were prepared and assessed by experts. The visual perception of the observers was correlated with the two different color difference equations CIELAB from DIN 6174 and the CIEDE2000 equation. For a practical example two different types of wood were bleached and exposed to UV-light. The results showed that the CIEDE2000 equation outperformed the CIELAB equation. The CIELAB equation showed generally an overestimation of the color change (ΔE) for wood applications, compared to the nontrivial CIEDE2000 equation from 2001. 相似文献

To reduce the release of pharmaceuticals and endocrine disruptors into the aquatic environment or to remove them from wastewater intended for direct or indirect reuse, the application of advanced wastewater treatment may be required. In the present study, municipal wastewater effluents were treated with ozone (O3) in a pilot-scale plant consisting of two bubble columns. The investigated effluents, which varied in suspended solids concentrations, comprised an effluent of conventional activated sludge treatment (CAS), the same effluent dosed with 15 mg of TSS L(-1) of activated sludge (CAS + SS), and the effluent of a membrane bioreactor pilot plant (MBR). Selected classes of pharmaceuticals were spiked in the wastewater at realistic levels ranging from 0.5 to 5 microg L(-1). Samples taken at the inlet and the outlet of the pilot plant were analyzed with liquid chromatography (LC)-electrospray tandem mass spectrometry (MS). Macrolide and sulfonamide antibiotics, estrogens, and the acidic pharmaceuticals diclofenac, naproxen, and indomethacin were oxidized by more than 90-99% for O3 doses > or = 2 mg L(-1) in all effluents. X-ray contrast media and a few acidic pharmaceuticals were only partly oxidized, but no significant differences were observed among the three effluents. These results show that many pharmaceuticals present in wastewater can be efficiently oxidized with O3 and that suspended solids have only a minor influence on the oxidation efficiency of nonsorbing micropollutants. 相似文献

Desorption experiments were carried out in flow through columns following long-term sorption batch experiments (up to 1010 days at 20 degrees C; Rügner, H.; Kleineidam, S.; Grathwohl, P. Long-term sorption kinetics of phenanthrene in aquifer materials. Environ. Sci. Technol. 1999, 33, 1645-1651) to elucidate sorption/desorption hysteresis phenomena of phenanthrene in aquifer materials. Most of the sorbents employed in this study (homogeneous lithocomponents separated from aquifer sediments or fresh rock fragments) showed highly nonlinear sorption isotherms because of coal particles embedded inside the grains. Because sorption capacities were high, sorption equilibrium was not reached in most of the sorbents during the initial sorptive uptake experiments lasting up to 1010 days. Desorption was studied up to 90 days at 20 degrees C. The temperature was raised after that stepwise from originally 20 to 30, 40, 50, and finally to 70 degrees C for selected samples to estimate activation energies of desorption. A numerical intraparticle pore diffusion model was used to fit sorptive uptake data and subsequently for pure forward prediction of the release rates in the desorption column experiments. Desorption was initially fast followed by extended tailing which in other studies is fitted by using multirate first-order models. Our results demonstrate that the retarded intraparticle pore diffusion model can predict the desorption rates with a single diffusion rate constant obtained independently from the long-term batch sorption experiment. No evidence for hysteresis was found, suggesting that many hysteresis phenomena reported earlier are experimental artifacts resulting from nonequilibrium effects and "nonphysical" models. The different temperature steps allowed one to additionally calculate activation energies of desorption (45-59 kJ mol(-1)), which were in reasonably good agreement with results from earlier studies for a retarded pore diffusion process. In addition, equilibrium sorption isotherms were determined at 20 and 40 degrees C to compare sorption and desorption enthalpies. The production of bulk chemicals from biomass can make a significant contribution to solving two of the most urgent environmental problems: climate change and depletion of fossil energy. We analyzed current and future technology routes leading to 15 bulk chemicals using industrial biotechnology and calculated their CO2 emissions and fossil energy use. Savings of more than 100% in non-renewable energy use and greenhouse gas emissions are already possible with current state of the art biotechnology. Substantial further savings are possible for the future by improved fermentation and downstream processing. Worldwide CO2 savings in the range of 500-1000 million tons per year are possible using future technology. Industrial biotechnology hence offers excellent opportunities for mitigating greenhouse gas emissions and decreasing dependence on fossil energy sources and therefore has the potential to make inroads into the existing chemical industry. 相似文献

The objectives of this study were (1) to analyze the agreement of a standard laboratory ELISA for progesterone (P4) with an automated on-farm ELISA kit operated under commercial conditions in 1,297 milk samples from 50 dairy cows; (2) to study the influence of the method of detection of luteal activity on genetic parameters of fertility traits based on P4 measured with an automated on-farm ELISA once weekly from wk 3 to 9 postpartum in the milk of 1,304 cows; and (3) to study the influence of sampling frequency (once or twice weekly from wk 3 to 9) on the same traits from 296 cows. Luteal activity can be detected when there is an active corpus luteum in the ovary producing P4 and indicating the onset of reproductive cyclicity after calving. The on-farm ELISA overestimated P4 contents by a mean square error of prediction of 2.76 ng/mL and had an intermediate Spearman correlation with the laboratory kit (0.54). For the second objective, the postpartum interval to the commencement of luteal activity (C-LA), proportion of luteal activity between d 15 and 63 postpartum (P-LA), calculated as the number of samples above the threshold for high P4 values divided by the number of all samples, and delay of first ovulation (DOV1), defined as C-LA occurring later than d 45 postpartum, were derived from the P4 profiles. Both C-LA and DOV1 were determined by (a) thorough qualitative visual inspection of the profile, (b) the profile's mean as threshold for the first increase in P4 postpartum, indicating commencement of luteal activity, and (c) 3 ng/mL as threshold for the first increase in P4, a value that has been used by many other studies. Similarly, P-LA was determined by using methods (b) and (c). Estimates of heritability were 0.04 to 0.13 for C-LA, 0.12 to 0.23 for P-LA, and 0.03 to 0.07 for DOV1. Genetic correlation of P-LA with C-LA and with the profile's mean P4 was ?1.00. The profile's mean had a higher estimate of heritability (0.11–0.12) than C-LA or DOV1. It can be calculated as the arithmetic mean of all P4 values of a profile, whereas C-LA, P-LA, and DOV1 need a definition of a threshold for high P4 values. We therefore suggest the profile's mean as a promising candidate for further research. For the third objective, once-weekly sampling was mimicked by neglecting every second sample, and C-LA and DOV1 shifted toward a later onset of cyclicity. Thus, a common standard for sampling regimen and detection algorithm is essential to avoid incompatibility between studies. 相似文献
