The modeling of a hybrid passive damping system is presented for suppressing the multiple vibration modes of beams in this paper. This hybrid passive damping system consists of a constrained layer damping and a resonant shunt circuit. In a passive mechanical constrained layer damping, a viscoelastic layer, which is sandwiched between a host structure and a cover layer, is used to suppress vibration amplitudes in the high frequency range. A passive electrical damping is designed for targeting the vibration amplitude in the low frequency range. The governing equations of motion of the hybrid passive damping system are derived through the Hamilton's principle. The obtained mathematical model is validated experimentally. The theoretical and experimental techniques presented provide an invaluable tool in controlling the multiple vibration modes across a wide frequency band.  相似文献

Poly(butylene succinate) (PBS) nanocomposites with multiwalled carbon nanotubes (MWNTs) prepared by melt compounding were studied for the effect of MWNT dispersion on the modulus and crystallization kinetics. The nucleating effect of the addition of 0.1 wt % MWNT to PBS was clearly demonstrated. Differential scanning calorimetry nonisothermal crystallization studies showed a clear decrease in the half‐time of crystallization with increasing MWNT content in PBS/MWNT nanocomposites. It was observed with the Ozawa method that the Ozawa parameter values for the nanocomposites were lower than those for neat PBS, and this indicated that the crystal morphology was different. The storage modulus of the nanocomposites increased about 23% with the addition of only 0.1% MWNT in comparison with neat PBS, whereas the glass‐transition temperature was unaltered. © 2008 Wiley Periodicals, Inc. A modified process for the dry spinning of carbon nanotube (CNT) yarn is reported. The approach gives an improved structure of CNT bundles in the web drawn from the CNT forest and in the yarn produced from the twisted web leading to improved mechanical properties of the yarn. The process enables many different mechanical and physical treatments to be applied to the individual stages of the pure CNT spinning system, and may allow potential for the development of complex spinning processes such as polymer-CNT-based composite yarns. The tensile strength and yarn/web structure of yarn spun using this approach have been investigated and evaluated using standard tensile testing methods along with scanning electron microscopy. The experimental results show that the tensile properties were significantly improved. The effect of heat treatments and other yarn constructions on the tensile properties are also reported.  相似文献

ABSTRACT

Polypyrrole (PPy) was prepared on the mild steel substrate by electrochemical polymerisation in the solution containing pyrrole monomer and succinic acid. The mild steel surface could be passivated before and during electropolymerisation by molybdate. The morphology and structure of the PPy film were studied with SEM. The typical cauliflower structure of PPy was observed. Raman and IR spectroscopy showed that the obtained PPy was in an oxidised state. The thermal stability of PPy was investigated by the thermal gravimetric analysis, showing that PPy was stable at higher than 480°C. The electrochemical property of the PPy film was performed by open circuit potential, polarisation curves (I/E), and electrochemical impedance spectroscopy. The corrosion behaviour of mild steel (CT3) with PPy film in solution NaCl 3% was studied.

Novel 1,8-naphthyridine-2-carboxamide derivatives with various substituents (HSR2101-HSR2113) were synthesized and evaluated for their effects on the production of pro-inflammatory mediators and cell migration in lipopolysaccharide (LPS)-treated BV2 microglial cells. Among the tested compounds, HSR2104 exhibited the most potent inhibitory effects on the LPS-stimulated production of inflammatory mediators, including nitric oxide (NO), tumor necrosis factor-α, and interleukin-6. Therefore, this compound was chosen for further investigation. We found that HSR2104 attenuated levels of inducible NO synthase and cyclooxygenase 2 in LPS-treated BV2 cells. In addition, it markedly suppressed LPS-induced cell migration as well as the generation of intracellular reactive oxygen species (ROS). Moreover, HSR2104 abated the LPS-triggered nuclear translocation of nuclear factor-κB (NF-κB) through inhibition of inhibitor kappa Bα phosphorylation. Furthermore, it reduced the expressions of Toll-like receptor 4 (TLR4) and myeloid differentiation factor 88 (MyD88) in LPS-treated BV2 cells. Similar results were observed with TAK242, a specific inhibitor of TLR4, suggesting that TLR4 is an upstream regulator of NF-κB signaling in BV2 cells. Collectively, our findings demonstrate that HSR2104 exhibits anti-inflammatory and anti-migratory activities in LPS-treated BV2 cells via the suppression of ROS and TLR4/MyD88/NF-κB signaling pathway. Based on our observations, HSR2104 may have a beneficial impact on inflammatory responses and microglial cell migration involved in the pathogenesis of various neurodegenerative disorders.  相似文献

Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a member of the colony-stimulating factor (CSF) family, which functions to enhance the proliferation and differentiation of hematopoietic stem cells and other hematopoietic lineages such as neutrophils, dendritic cells, or macrophages. These proteins have thus generated considerable interest in clinical therapy research. A current obstacle to the prokaryotic production of human GM-CSF (hGM-CSF) is its low solubility when overexpressed and subsequent complex refolding processes. In our present study, the solubility of hGM-CSF was examined when combined with three N-terminal fusion tags in five E. coli strains at three different expression temperatures. In the five E. coli strains BL21 (DE3), ClearColi BL21 (DE3), LOBSTR, SHuffle T7 and Origami2 (DE3), the hexahistidine-tagged hGM-CSF showed the best expression but was insoluble in all cases at each examined temperature. Tagging with the maltose-binding protein (MBP) and the b′a′ domain of protein disulfide isomerase (PDIb′a′) greatly improved the soluble overexpression of hGM-CSF at 30 °C and 18 °C. The solubility was not improved using the Origami2 (DE3) and SHuffle T7 strains that have been engineered for disulfide bond formation. Two conventional chromatographic steps were used to purify hGM-CSF from the overexpressed PDIb′a′-hGM-CSF produced in ClearColi BL21 (DE3). In the experiment, 0.65 mg of hGM-CSF was isolated from a 0.5 L flask culture of these E. coli and showed a 98% purity by SDS-PAGE analysis and silver staining. The bioactivity of this purified hGM-CSF was measured at an EC50 of 16.4 ± 2 pM by a CCK8 assay in TF-1 human erythroleukemia cells.  相似文献

Human stem-cell factor (hSCF) stimulates the survival, proliferation, and differentiation of hematopoietic cells by binding to the c-Kit receptor. Various applications of hSCF require the efficient and reliable production of hSCF. hSCF exists in three forms: as two membrane-spanning proteins hSCF248 and hSCF229 and truncated soluble N-terminal protein hSCF164. hSCF164 is known to be insoluble when expressed in Escherichia coli cytoplasm, requiring a complex refolding procedure. The activity of hSCF248 has never been studied. Here, we investigated novel production methods for recombinant hSCF164 and hSCF248 without the refolding process. To increase the solubility of hSCF164, maltose-binding protein (MBP) and protein disulfide isomerase b’a’ domain (PDIb’a’) tags were attached to the N-terminus of hSCF164. These fusion proteins were overexpressed in soluble form in the Origami 2(DE3) E. coli strain. These solubilization effects were enhanced at a low temperature. His-hSCF248, the poly-His tagged form of hSCF248, was expressed in a highly soluble form without a solubilization tag protein, which was unexpected because His-hSCF248 contains a transmembrane domain. hSCF164 was purified using affinity and ion-exchange chromatography, and His-hSCF248 was purified by ion-exchange and gel filtration chromatography. The purified proteins stimulated the proliferation of TF-1 cells. Interestingly, the EC50 value of His-hSCF248 was 1 pg/mL, 100-fold lower than 9 ng/mL hSCF164. Additionally, His-hSCF248 decreased the doubling time, increased the proportion of S and G2/M stages in the cell cycle, and increased the c-Myc expression at a 1000-fold lower concentration than hSCF164. In conclusion, His-hSCF248 was expressed in a soluble form in E. coli and had stronger activity than hSCF164. The molecular chaperone, MBP, enabled the soluble overexpression of hSCF164.  相似文献

Cell localisation in the matrix microstructure of fermented food is important for microbial activity and ripening. To evaluate the importance of physicochemical factors in these phenomena, the physicochemical behaviour of four strains of lactic acid bacteria were investigated after inoculation of raw milk or milk fractions. To discriminate between physicochemical and metabolic properties, media were (i) rapidly acidified to avoid cell division or (ii) bacteria were allowed to ferment milk. To evaluate the impact of milk components, cells were dispersed in protein or fatty globule fractions. From two Lactococcus lactis subsp. lactis biovar. diacetylactis strains selected from their different surface properties investigated in previous studies, after rapid acidification one was encountered in fatty globules and protein gels whereas the other was found in serum and on the surface of globules. After fermentation, the first one formed colonies in the lipid phase or next to it in the whey and the second formed colonies in the protein gel. Strains forming long chains (Lactobacillus bulgaricus and Streptococcus thermophilus) were always found in contact with fatty globules, where they developed. Our results suggest that localisation and subsequent spatial colonisation depend on the physicochemical properties of cell surfaces, on the structure in chains as well as on the metabolic ability to develop on proteins or lipids.  相似文献
