The possibility of storage of human bone marrow CD34+ cells at 4 degrees C for bone marrow transplantation purposes was investigated. The cells were placed in Iscove medium supplemented with 20% serum (15% bovine calf serum + 5% human AB serum) for three weeks at 4 degrees C. During storage time clonogenicity of granulocytic-monocytic (CFU-GM) erythropoietic (BFU-E) and megakaryocytic (CFU-Meg) progenitors were investigated. It turns out that it is possible to keep human CD34+ cells at 4 degrees C for at least few days before transplantation. At day four of storage the number of CFU-GM and BFU-E cells still exceeded 50% of cells present at day 0. We have found however, CFU-Meg progenitors in comparison to others are much more sensitive to metabolic storage stress. This enhanced sensitivity of megakaryocytic progenitors could explain at least partially the well known ++phenomenon of retardation of thrombopoietic recovery in patients undergoing bone marrow transplantation.  相似文献

We investigated whether glycinergic transmission develops organotypically in auditory brain stem cultures. Slices of the medial nucleus of the trapezoid body and the lateral superior olive were incubated in medium with a raised extracellular K+ concentration. As in vivo, glycine receptor alpha1 subunit immunoreactivity increased and became clustered on somata and proximal dendrites. Together with organotypic expression of glycine transporter GLYT2, this indicates that molecular components of glycinergic synapses form properly. In contrast, glycinergic synaptic currents did not develop as in vivo: after 7 days in vitro they were still similar to those at the time of culture preparation. We suggest that for organotypic development of glycine receptors and transporters, Ca2+ influx due to elevated K+ is sufficient. The development of functional synaptic transmission, however, may require patterned electrical activity.  相似文献

An approach to short-term economic rescheduling is proposed. The method is based on an iterative combination of alternating consideration of technical and economic criteria. Its attractiveness lies in incorporating both technical and economic goals without the computational expense of combining them in a mathematical optimization process. A specific algorithm is proposed which is based on using a localized mismatch spreading algorithm. The effect of spinal alpha adrenoceptor blockage on the inhibition of renal sympathetic nerve activity (RSNA) and natriuresis induced by blood volume expansion was investigated in anesthetized and bilateral sinoaortic denervated rabbits. In the groups of rabbits with intrathecal injection of alpha-adrenoceptor blocker phentolamine or artificial cerebrospinal fluid the inhibition of RSNA induced by blood volume expansion were (-25.4 +/- 5.4)% and (-42.5 +/- 5.2)% respectively (P < 0.05). In the groups of rabbits with intrathecal injection of alpha 1 adtenoceptor blocker prazosin or artificial cerebrospinal fluid the inhibition of RSNA induced by blood volume expansion were (-29.3 +/- 6.1)% and (-42.5 +/- 5.2)% respectively (P < 0.05). These results suggested that both spinal alpha and alpha 1 adrenceptor blockage with attenuated the inhibition of RSNA induced by blood volume expansion. The spinal alpha 1 adrenceptor blockage with intrathecal injection of prazosin also attenuated signiticantly the natriuresis and diuresis induced by blood volume expansion (P < 0.05).  相似文献

X-ray diffraction analysis at 1.5 A resolution has confirmed the helical conformation of a de novo designed 18-residue peptide. However, the crystal structure reveals the formation of continuous molecular layers of parallel-packed amphiphilic helices as a result of much more extensive helix-helix interactions than predicted. The crystal packing arrangement, by virtue of distinct antiparallel packing interactions, segregates the polar and apolar surfaces of the helices into discrete and well-defined interfacial regions. An extensive "ridges-into-grooves" interdigitation characterizes the hydrophobic interface, whereas an extensive network of salt bridges and hydrogen bonds dominates the corresponding hydrophilic interface.  相似文献
