The use of Surrogate Based Optimization (SBO) is widely spread in engineering design to find optimal performance characteristics of expensive simulations (forward analysis: from input to optimal output). However, often the practitioner knows a priori the desired performance and is interested in finding the associated input parameters (reverse analysis: from desired output to input). A popular method to solve such reverse (inverse) problems is to minimize the error between the simulated performance and the desired goal. However, there might be multiple quasi-optimal solutions to the problem. In this paper, the authors propose a novel method to efficiently solve inverse problems and to sample Quasi-Optimal Regions (QORs) in the input (design) space more densely. The development of this technique, based on the probability of improvement criterion and kriging models, is driven by a real-life problem from bio-mechanics, i.e., determining the elasticity of the (rabbit) tympanic membrane, a membrane that converts acoustic sound wave into vibrations of the middle ear ossicular bones.  相似文献

This work presents a new adsorption technique where the adsorbent (powdered activated carbon-PAC) is in the form of suspended flocs formed with water-soluble polymer flocculants. Thus, the adsorption of a typical dye, methylene blue (MB), was studied onto polyacrylamide flocs of PAC (PACF) in a fluidized bed reactor. The technique is based on the fact that the adsorption capacity of PAC does not decrease after flocculation because the adsorbed polymer occupies only a few surface sites, in the form of trains, loops, and tails. Moreover, the adsorption was found to proceed through a rapid mass transfer of MB to the adsorbing PAC flocs, in the same extent as onto PAC. Because of the rapid settling characteristics of the aggregates formed, the two phase separations, loaded PAC and solution, become easier. Thus, the technique offers the advantages of conducting simultaneously both adsorption and solid/liquid separation all in one single stage. Results obtained showed that high MB removal values can be attained in a fluidized bed reactor (>90%) and that PACF presents a much higher adsorption capacity (breakthrough points) than granulated activated carbon (GAC) in the same adsorbing bed. It is believed that this technique highly broadens the potential of the use of powdered activated carbon or other similar ultrafine adsorbents.  相似文献

Investigation of pristine g-C3N4 and copper ions doped g-C3N4 as dispersed phases in silicone-oil based electrorheological (ER) fluids is reported for the first time. Pristine g-C3N4 was prepared via standard thermal polycondensation of urea. Introduction of Cu ions into g-C3N4 polymeric network was performed by treatment of pristine powder by a solution containing tetraammoniumdiaquacopper(II) complex ions followed by annealing. The structure and properties of products were revealed with the aid of XRD, FTIR, UV–Vis and XPS, and complemented by SEM investigation of morphology, pycnometry, BET analysis, and conductivity measurements. The response of prepared ER fluids to an external electric field was examined by rheometry and the effects visualized with optical microscopy. While ER fluids based on g-C3N4 exhibited negative ER effect (decrease in viscosity when the field is switched-on), ER fluids based on a g-C3N4/Cu-doped analogue exhibited positive ER effect. Dielectric spectroscopy using Havriliak-Negami model revealed that introduction of the dopant almost doubled the dielectric relaxation strength while the relaxation time was halved. The ability of g-C3N4/Cu as a candidate for further studies necessary to increase the ER effect was demonstrated. Thereto, described modification of g-C3N4 by copper ions shall be applicable also for other metals forming ammonia complexes.  相似文献

The formation of artificial metal base pairs is an alluring and versatile method for the functionalization of nucleic acids. Access to DNA functionalized with metal base pairs is granted mainly by solid-phase synthesis. An alternative, yet underexplored method, envisions the installation of metal base pairs through the polymerization of modified nucleoside triphosphates. Herein, we have explored the possibility of using thiolated and pKa-perturbed nucleotides for the enzymatic construction of artificial metal base pairs. The thiolated nucleotides S2C, S6G, and S4T as well as the fluorinated analogue 5FU are readily incorporated opposite a templating S4T nucleotide through the guidance of metal cations. Multiple incorporation of the modified nucleotides along with polymerase bypass of the unnatural base pairs are also possible under certain conditions. The thiolated nucleotides S4T, S4T, S2C, and S6G were also shown to be compatible with the synthesis of modified, high molecular weight single-stranded (ss)DNA products through TdT-mediated tailing reactions. Thus, sulfur-substitution and pKa perturbation represent alternative strategies for the design of modified nucleotides compatible with the enzymatic construction of metal base pairs.  相似文献

Diatomite is a natural fossil material of sedimentary origin, constituted by fragments of diatom siliceous skeletons. In this preliminary work, the properties of diatomite nanoparticles as potential system for the delivery of drugs in cancer cells were exploited. A purification procedure, based on thermal treatments in strong acid solutions, was used to remove inorganic and organic impurities from diatomite and to make them a safe material for medical applications. The micrometric diatomite powder was reduced in nanoparticles by mechanical crushing, sonication, and filtering. Morphological analysis performed by dynamic light scattering and transmission electron microscopy reveals a particles size included between 100 and 300 nm. Diatomite nanoparticles were functionalized by 3-aminopropyltriethoxysilane and labeled by tetramethylrhodamine isothiocyanate. Different concentrations of chemically modified nanoparticles were incubated with cancer cells and confocal microscopy was performed. Imaging analysis showed an efficient cellular uptake and homogeneous distribution of nanoparticles in cytoplasm and nucleus, thus suggesting their potentiality as nanocarriers for drug delivery.

The present work deals with reverse engineering on four rubber formulations. Information about the material composition of the compounding formulations was obtained by using techniques such as acetone extract, thermogravimetric analysis, energy dispersive X‐ray fluorescence studies, and Fourier transform infrared (FTIR) spectroscopy in attenuated total reflection mode. The reverse engineered formulations were compared on a qualitative and quantitative basis with the initial formulations. The aim was to ascertain the validity of the methodology, which may be used as reverse engineering of rubber compounds to quantitatively analyze unknown ground tire rubber in future. The state of the art was based on a fractional mass transfer from acetone extract to the rubbers detected in TGA. Through this new approach, the formulations calculated were very consistent with the formulations employed, thus establishing the validity of the methodology used. POLYM. ENG. Autophagy is a self-degradative process that involves turnover and recycling of cytoplasmic components in healthy and diseased tissue. Autophagy has been shown to be protective at the early stages of programmed cell death but it can also promote apoptosis under certain conditions. Earlier we demonstrated that oxygen contributes to the pathogenesis of neonatal brain damage, which can be ameliorated by intervention with recombinant human erythropoietin (rhEpo). Extrinsic- and intrinsic apoptotic pathways are involved in oxygen induced neurotoxicity but the role of autophagy in this model is unclear. We analyzed the expression of autophagy activity markers in the immature rodent brain after exposure to elevated oxygen concentrations. We observed a hyperoxia-exposure dependent regulation of autophagy-related gene (Atg) proteins Atg3, 5, 12, Beclin-1, microtubule-associated protein 1 light chain 3 (LC3), LC3A-II, and LC3B-II which are all key autophagy activity proteins. Interestingly, a single injection with rhEpo at the onset of hyperoxia counteracted these oxygen-mediated effects. Our results indicate that rhEpo generates its protective effect by modifying the key autophagy activity proteins.  相似文献

The presented work describes mechanical properties of materials prepared by the pyrolysis of polysiloxane resins. The polymeric precursors have different chemical composition. Materials under investigation are predetermined for high temperature resistant applications usually in composite form, i.e. accompanied by some reinforcement. Instrumented hardness tests were employed for material characterisation. The Vickers hardness, Martens hardness and indentation elastic modulus were the key parameters, determined either from the standard optical technique or load–indentation depth curves. The influence of mechanical properties on the monomers ratio was established. The exposition to the temperature between 1200 °C and 1500 °C in air was applied to observe damage caused by these severe conditions.  相似文献
