Bleached kraft mill requires large quantities of water and chemicals reactives and generates effluents containing active organic compounds. Specifically, hormonal changes in fish and their communities have been demonstrated, which could be attributed to endocrine-disrupting chemicals (EDCs) contained in kraft mill effluents. This chronic toxicity is attributable to extractive compounds, such as resin acids, sterols, and fatty acids. The goal of this work is to evaluate the methanogenic toxicity and anaerobic biodegradation of stigmasterol and beta-sitosterol. A continuously anaerobic filter (AF) was used for the anaerobic biodegradation of stigmasterol and beta-sitosterol. Three phases were evaluated. In phase I, an elementary chlorine-free bleached kraft mill effluent was fed to the reactor whereas in phases II and III, the effluent was supplemented with increasing stigmasterol and beta-sitosterol concentrations. The AF displays high performance in biological oxygen demand (BOD5) removal (up to 94%); however, only 50% of the chemical oxygen demand (COD) was removed. Simultaneously, the AF system shows a great ability to remove beta-sitosterols (77-100%) and stigmasterols (87-95%). No negative effect on the methanogenic activity inhibition was shown by beta-sitoesterols and stigmasterol. However, a mixture of beta-sitosterols and stigmasterols (ratio 1:1) caused a less than 10% reduction in methanogenic activity.  相似文献

We developed a simple, non-invasive method to measure the intramammary pressure (IMP) response in sows during natural milk ejection and after i.v. injections of oxytocin. The teats of sows were cannulated and the amplitude and duration of IMP changes were measured via a pressure transducer linked to a computer. During natural milk ejections, the amplitude of the response was 5.5+/-1.8 kPa (mean +/- SD) and the duration of response at half the peak amplitude was 6.8+/-1.6 s. In response to oxytocin a threshold response occurred at 60 mIU oxytocin with no further change up to 200 mIU. The amplitude of the response to an i.v. dose of 60-200 mIU oxytocin (5.5+/-2.2 kPa) and duration (6.4+/-1.7 s) were similar to that during a natural milk ejection. The responses of two galactophorous ducts of a teat measured simultaneously were similar in peak amplitude and duration. When the responses of four glands of a sow were measured simultaneously, there was a lag time in the onset of the IMP peak from the anterior to the posterior glands, and an increase in the peak amplitude from anterior to posterior. Both during natural milk ejections and in response to oxytocin injections, there were significant differences in peak amplitude between sows. We also aimed to determine whether the pressure or duration of milk flow during natural milk ejections was limiting piglet growth rates. IMP of nine sows and the growth rates of their piglets were measured. Both the peak amplitude of IMP response and the growth rate of piglets increased over the course of lactation, but the relationship was not causal. We found that the growth rate of piglets did not depend on IMP amplitude or duration, but we suggest that it may depend on the frequency of suckling and/or the ability of piglets to withdraw milk while it is available.  相似文献

This paper describes a probabilistic model, based on the Johnson-Ettinger algorithm, developed to characterize the current and historic exposure to tricholorethylene (TCE) and tetrachlorethylene (PCE) in indoor air from plumes of groundwater contamination emanating from the former Kelly Air Force Base in San Antonio, Texas. We estimate indoor air concentration, house by house, in 30?101 homes and compare the estimated concentrations with measured values in a small subset of homes. We also compare two versions of the Johnson-Ettinger model: one used by the Environmental Protection Agency (EPA) and another based on an alternative parametrization. The modeled mean predicted PCE concentration historically exceeded PCE screening levels (0.41 ug/m(3)) in 5.5% of houses, and the 95th percentile of the predicted concentration exceeded screening levels in 85.3% of houses. For TCE, the mean concentration exceeded the screening level (0.25 ug/m(3)) in 49% of homes, and the 95th percentile of the predicted concentration exceeded the screening level in 99% of homes. The EPA model predicts slightly lower indoor concentrations than the alternative parametrization. Comparison with measured samples suggests both models, with the inputs selected, underestimate indoor concentrations and that the 95th percentiles of the predicted concentrations are closer to measured concentrations than predicted mean values.  相似文献

Grating-coupled surface plasmon resonance imaging (GCSPRI) utilizes an optical diffraction grating embossed on a gold-coated sensor chip to couple collimated incident light into surface plasmons. The angle at which this coupling occurs is sensitive to the capture of analyte at the chip surface. This approach permits the use of disposable biosensor chips that can be mass-produced at low cost and spotted in microarray format to greatly increase multiplexing capabilities. The current GCSPRI instrument has the capacity to simultaneously measure binding at over 1000 unique, discrete regions of interest (ROIs) by utilizing a compact microarray of antibodies or other specific capture molecules immobilized on the sensor chip. In this report, we describe the use of GCSPRI to directly detect multiple analytes over a large dynamic range, including soluble protein toxins, bacterial cells, and viruses, in near real-time. GCSPRI was used to detect a variety of agents that would be useful for diagnostic and environmental sensing purposes, including macromolecular antigens, a nontoxic form of Pseudomonas aeruginosa exotoxin A (ntPE), Bacillus globigii, Mycoplasma hyopneumoniae, Listeria monocytogenes, Escherichia coli, and M13 bacteriophage. These studies indicate that GCSPRI can be used to simultaneously assess the presence of toxins and pathogens, as well as quantify specific antibodies to environmental agents, in a rapid, label-free, and highly multiplexed assay requiring nanoliter amounts of capture reagents.  相似文献

Environmental impacts due to engineered nanomaterials arise both from releases of the nanomaterials themselves as well as from their synthesis. In this work, we employ the USEtox model to quantify and compare aquatic ecotoxicity impacts over the life cycle of carbon nanotubes (CNTs). USEtox is an integrated multimedia fate, transport, and toxicity model covering large classes of organic and inorganic substances. This work evaluates the impacts of non-CNT emissions from three methods of synthesis (arc ablation, CVD, and HiPco), and compares these to the modeled ecotoxicity of CNTs released to the environment. Parameters for evaluating CNT ecotoxicity are bounded by a highly conservative "worst case" scenario and a "realistic" scenario that draws from existing literature on CNT fate, transport, and ecotoxicity. The results indicate that the ecotoxicity impacts of nanomaterial production processes are roughly equivalent to the ecotoxicity of CNT releases under the unrealistic worst case scenario, while exceeding the results of the realistic scenario by 3 orders of magnitude. Ecotoxicity from production processes is dominated by emissions of metals from electricity generation. Uncertainty exists for both production and release stages, and is modeled using a combination of Monte Carlo simulation and scenario analysis. The results of this analysis underscore the contributions of existing work on CNT fate and transport, as well as the importance of life cycle considerations in allocating time and resources toward research on mitigating the impacts of novel materials.  相似文献

The Internet provides a medium to administer and evaluate evidence-based interventions for highly prevalent public health problems worldwide. The authors report a series of four Internet smoking cessation studies conducted in English and Spanish. These studies examined both outcome (self-reported 7-day abstinence) and mechanisms related to outcome (the impact of major depressive episodes [MDEs] on the likelihood of quitting). Over 4,000 smokers from 74 countries entered the studies. Studies 1 and 2 evaluated a standard smoking cessation guide (the "Guía"). Studies 3 and 4 were randomized trials comparing the Guía+ITEMs (individually timed educational messages) to the Guía+ITEMs+a mood management course. ITEMs were E-mails inviting participants back to the site at specific times. Online follow-up assessments resulted in completion rates of 44%-54% at 1 month and 26%-30% at 6 months in studies 1 and 2. Incentives and follow-up phone calls increased these rates to 70%, 66%, 65%, and 62% at 1, 3, 6, and 12 months in study 4. At 6 months, self-reported 7-day abstinence rates using missing = smoking data were 6% in studies 1 and 2, 10%-14% in study 3, and 20%-26% in study 4. The Guía+ITEMs condition tended to have higher quit rates, which reached significance at the 12-month follow-up in study 3 and at the 3-month follow-up in study 4. Smokers with past (but not current) MDEs tended to be the most likely to abstain and those with current MDEs the least likely. This trend reached significance in studies 1 and 4.  相似文献

Ultrasonic imaging of biofilms in water is difficult due to the very low contrast in acoustic impedance between the biofilm and water. In this paper, biofilms exposed to moist air are scanned through the substrate in order to obtain echoes from the biofilm/air interface. A 50 MHz scanning system was used to scan 1 mm x 1 mm areas of biofilms in a 10 microm grid pattern. Two fast Fourier transform (FFT) based methods for enhancement of the film thickness measurement resolution are compared. Using these techniques, the surface topography of biofilms with thickness less than the acoustic wavelength can be imaged.  相似文献

The stability of polyelectrolyte multilayer assemblies was investigated with emphasis on the effects of solution ionic strength, pH, and polymer molecular weight on the film thickness and surface topography. The multilayers consisting of two polysaccharides, the polyanion sodium hyaluronate (HA) and the polycation chitosan (CH) were studied using surface plasmon resonance (SPR) spectroscopy, impedance quartz crystal microbalance (QCM), and atomic force microscopy (AFM). SPR/QCM experiments show that coatings consisting of four HA/CH bilayers assembled at pH 4.5 in the presence of 0.15 M NaCl are stable in NaCl solutions of concentration less than 0.8 M. These multilayers are stable when placed in contact with aqueous solutions ranging in pH from 3.5 to 9. The molecular weight of the polysaccharides has only a marginal effect on the stability of the films in the range explored here (HA: Mn = 360,000 or 31,000 g/mol; CH: Mn = 160,000 or 30,000 g/mol). AFM imaging reveals that different mechanisms may account for the multilayers stability versus salt and pH treatments. While increasing the ionic strength induces reorganization of the surface topography from isolated spherical islets to elongated worm-like features, changes in pH have no appreciable effects on the coating topography prior to complete disintegration.  相似文献

Aquatic plants take up and sequester organic contaminants such as chlorophenols through incorporation in cell wall materials and storage in vacuoles. The ultimate fate of plant-sequestered chlorophenols, however, remains unclear. This research investigated 2,4-dichlorophenol (2,4-DCP) sequestration by the aquatic plant Lemna minor and evaluated contaminant release and bioavailability after plant death and cellular disruption. 14C-labeled 2,4-DCP was used to establish that contaminant removed from the aqueous phase was retained internal to L. minor. An assay with Desulfitobacterium sp. strain Viet1 was used to assess the readily bioavailable fraction of plant-sequestered 2,4-DCP and plant metabolites of 2,4-DCP. In plant-free systems, strain Viet1 dechlorinated 2,4-DCP to stoichiometric amounts of 4-chlorophenol (4-CP) as a stable and quantifiable end product. Anaerobic microcosms containing inactivated L. minor, which had accumulated 3.8 micromol of 2,4-DCP equivalents/g of plant material (fresh weight) during a preceding aerobic exposure, were inoculated with strain Viet1. After 118 d of incubation with strain Viet1, 43.5% (+/-1.4%) of the contaminant was recovered as 4-CP, indicating a large portion of plant-sequestered 2,4-DCP was bioavailable for dechlorination by strain Viet1. In contrast, 4-CP formation was not observed in autoclaved microcosms, and only 26.1% (+/-1.0%) of plant-sequestered 2,4-DCP was recovered in the aqueous phase. These findings demonstrate contaminant cycling between plants and microorganisms, and emphasize that understanding the mechanisms and pathways of contaminant sequestration by plants is critical for predicting long-term contaminant fate.  相似文献
