Assigned 108 mothers (mean age 40 yrs) and daughters (mean age 13.4 yrs), mostly low in educational and socioeconomic status, to receive no treatment, a discussion-oriented traditional treatment, or a Relationship Enhancement (Parent Adolescent Relationship Development) treatment. There was no improvement without treatment. Participation in the traditional treatment—designed to improve (a) specific communication skills, (b) general communication patterns, and (c) the general quality of the relationship—resulted in improvement on 3 of the 5 measures of (c). The improvement on 1 of these measures raised them above their no-treatment counterparts at posttreatment. Ss in Relationship Enhancement improved significantly and reached higher levels than Ss in both other conditions on each of the measures assessing areas (a), (b), and (c). In addition to concluding that Relationship Enhancement is superior to a traditional discussion-oriented treatment, it can be concluded that a significant portion of its effectiveness stems from the particular methods it employs rather than from nonspecific factors such as experimenter demand, placebo, attention, and thank-you effects. We present a sample preparation method for measuring magnesium in individual whole lymphocytes by electron probe X-ray microanalysis. We use Burkitt's lymphoma cells in culture as the test sample and compare X-ray microanalysis of individual cells with atomic absorption analysis of pooled cell populations. We determine the magnesium peak-to-local continuum X-ray intensity ratio by electron probe X-ray microanalysis and calculate a mean cell magnesium concentration of 39± 19 mmol/kg dry weight from analysis of 100 cells. We determine a mean cell magnesium concentration of 34 ±4 mmol/kg dry weight by atomic absorption analysis of pooled cells in three cell cultures. The mean cell magnesium concentrations determined by the two methods are not significantly different. We find a 10% coefficient of variation for both methods of analysis and a 30% coefficient of variation in magnesium concentration among individual cells by electron probe X-ray microanalysis. We wash cells in ammonium nitrate for microanalysis or in buffered saline glucose for atomic absorption analysis. We find cells washed in either solution have the same cell viability (85%), recovery (75%), cell volume (555 μm3) and cytology. We air dry cells on thin film supports and show by magnesium X-ray mapping that magnesium is within the cells. We conclude that: (a) our microanalysis cell preparation method preserves whole intact lymphocytes; (b) there is no systematic difference in results from the two methods of analysis; (c) electron probe X-ray microanalysis can determine the variation in magnesium concentration among individual cells.  相似文献

The specificity of Watson-Crick base pairing, unique mechanical properties of DNA, and intrinsic stability of DNA double helices makes DNA an ideal material for the construction of dynamic nanodevices. Rationally designed strand displacement reactions can be used to produce dynamic reconfiguration of DNA nanostructures postassembly. Here we describe a 'fold-release-fold' strategy of multiple strand displacement and hybridization reactions to reconfigure a simple DNA origami structure into a complex, quasifractal pattern, demonstrating a complex transformation of DNA nanoarchitectures.  相似文献

Stroke is associated with the intracerebral formation of fibrin clots which may lead to irreversible brain damage. Thrombolytic therapies employ a variety of natural and/or recombinant plasminogen activators to initiate fibrinolytic degradation of cerebral thrombi. However, such therapies when installed beyond 4‐ to 6‐h window, may fail to achieve the expected outcome. This is due to the hydroxyl radical‐induced modification of fibrin(ogen) molecules rendering them refractory to fibrinolytic degradation, but no cause of the increased free radical generation in stroke was offered. Here, we show by means of electron microscopy that iron ions added to human blood dramatically enhances fibrin fibers formation with thrombin, and significantly delays fibrinolysis during spontaneous clotting of native blood. Iron ions caused the appearance dense matted fibrin deposits, similar, if not identical, to those observed in plasma of patients with stroke. These results may explain a known relationship between thrombotic diseases and the increased body concentrations of free iron and/or hemoglobin derivatives. We conclude that any action resulting in the inhibition of hemostatic abnormalities, as well as in the reduction of body free iron and scavenging of hydroxyl radicals (e.g., by polyphenols) can potentially prevent pathological reactions associated with consequences of stroke. The hepatitis C virus (HCV) is a single‐stranded (ss) RNA virus that is responsible for chronic liver diseases, such as cirrhosis, end‐stage liver disease, and hepatocellular carcinoma. Driven by the need to detect the presence of the HCV viral sequence, herein it is demonstrated for the first time that the nonlinear optical (NLO) properties of gold nanoparticles can be used for screening and quantifying HCV RNA without any modification, with excellent detection limit (80 pM ) and selectivity (single base‐pair mismatch). The hyper‐Rayleigh scattering (HRS) intensity increases 25 times when label‐free, 145‐mer, HCV ss‐RNA is hybridized with 400 pM target RNA. The mechanism of HRS intensity change is discussed with experimental evidence for a higher multipolar contribution to the NLO response of gold nanoparticles.  相似文献

Thin films of Cu(In,Ga)Se2 (CIGS) absorber layers for thin film solar cells have been manufactured on polyimide foil in a low temperature, ion beam assisted co-evaporation process.In the present work a set of CIGS thin films was produced with varying selenium ion energy. Solar cell devices have been manufactured from the films and characterized via admittance spectroscopy and capacitance-voltage profiling to determine the influence of the selenium ion energy on the electric parameters of the solar cells. It is shown that the impact of energetic selenium ions in the CIGS deposition process leads to a change in the activation energy and defect density and also in the spatial distribution of electrically active defects.For the interpretation of the results two defect models are taken into account.  相似文献
