This study examines the JUNIOR SUMMIT online community, which consisted of 3,062 adolescents representing 139 countries, varying SES, and a range of experience with computers. The online forum culminated in the election of 100 delegates. By analyzing the messages posted before results of the election were announced, we explore whether language use predicts who was elected as a leader, as well as gender differences in leadership style. Results indicate that the young online leaders do not adhere to adult leadership styles of contributing many ideas, sticking to task, and using powerful language. On the contrary, while the young people elected as delegates do contribute more, their linguistic style is likely to keep the goals and needs of the group as central--by referring to the group rather than to themselves, and by synthesizing the posts of others rather than solely contributing their own ideas. Furthermore, both boy and girl leaders follow this pattern of interpersonal language use. These results reassure us that young people can be civically engaged and community minded, while indicating that these concepts themselves may change through contact with the next generation. Epidemiological studies have demonstrated that levels of plasma fibrinogen, von Willebrand factor (vWf), plasminogen activator inhibitor-1 (PAI-1) and tissue-type plasminogen activator (tPA) are associated with the incidence of vascular disease. Since oncostatin M dramatically induces fibrinogen biosynthesis by hepatocytes and could be implicated in vascular injury leading to atherosclerosis, we have analyzed the effect of oncostatin M on PAI-1, vWf and tPA secretion by endothelial cells. A 2-h incubation of human umbilical vein endothelial cells with oncostatin M increases thrombin-induced secretion of vWf to the same extent as tumour necrosis factor-alpha or interleukin-1 (137+/-26% of control for 5 ng/ml oncostatin M, P < 0.001, n=5). The effects on tPA and PAI-1 secretion were different depending on the type of endothelial cells tested. On human umbilical vein endothelial cells, oncostatin M induced an increase in PAI-1 and a decrease in tPA secretion, which could explain the thrombogenicity of oncostatin M on large vessels. On a human microvasculature endothelial cell line, oncostatin M did not modify PAI-1 but induced an increase in tPA secretion. This observation of the effects of oncostatin M on both macro- and microvasculature could explain the increased levels of vWf, PAI-1 and tPA in the plasma of atherosclerotic subjects identified in epidemiological studies, suggesting that oncostatin M could play a key role in the development of atherosclerotic lesions.  相似文献

A novel method for the determination of hepatotoxins; microcystins (MCs), and nodularin (Nod) in lake water and domestic chlorinated tap water has been developed using liquid chromatography hyphenated with electrospray ionization triple quadrupole mass spectrometry (LC-ESI-MS/MS). Optimization of the mass spectrometer parameters and mobile-phase composition was performed to maximize the sensitivity and reproducibility of the method. Detection of the hepatotoxins was carried out using multiple reaction monitoring experiments, thus improving the selectivity of the method. A total ion chromatogram and a precursor ion scan on ion m/z 135 was also applied to all samples to detect unknown microcystins or microcystins for which there are no standards available. A comprehensive validation of the LC-ESI-MS/MS method was completed that took into account matrix effects, specificity, linearity, accuracy, and precision. Good linear calibrations were obtained for MC-LR (1-200 microg/L; R2=0.9994) in spiked lake and tap water samples (1-50 microg/L; R2=0.9974). Acceptable interday repeatability was achieved for MC-LR in lake water with RSD values (n=9) ranging from 9.9 (10 microg/L) to 5.1% (100 microg/L). Excellent limits of detection (LOD) and limits of quantitation (LOQ) were achieved with spiked MCs and Nod samples; LOD=0.27 microg/L and LOQ=0.90 microg/L for MC-LR in the "normal linear range" and LOD=0.08 microg/L and LOQ=0.25 microg/L in the "low linear range" in both lake and chlorinated tap water. Similar results were obtained for a suite of microcystins and nodularin. This sensitive and rapid method does not require any sample preconcentration, including the elimination of solid-phase extraction (SPE) for the effective screening of hepatotoxins in water below the 1 microg/L WHO provisional guideline limit for MC-LR. Furthermore, SPE techniques are time-consuming, nonreproducible at trace levels, and offer poor recoveries with chlorinated water. The application of this LC-ESI-MS/MS method for routine screening of hepatotoxins in lake and chlorinated tap water (average Cl2=0.23 mg/L) is achieved and this study represents the first direct method for the screening of hepatotoxins in chlorinated tap water.  相似文献

Poly (ADP-ribose) polymerase inhibitors (PARPi) are targeted therapies that inhibit PARP proteins which are involved in a variety of cell functions. PARPi may act as modulators of angiogenesis; however, the relationship between PARPi and the vasculogenic mimicry (VM) in breast cancer remains unclear. To determine whether PARPi regulate the vascular channel formation, we assessed whether the treatment with olaparib, talazoparib and veliparib inhibits the vascular channel formation by breast cancer cell lines. Here, we found that PARPi act as potent inhibitors of the VM formation in triple negative breast cancer cells, independently of the BRCA status. Mechanistically, we find that PARPi trigger and inhibit the NF-κB signaling, leading to the inhibition of the VM. We further show that PARPi decrease the expression of the angiogenic factor PTX3. Moreover, PTX3 rescued the PARPi-inhibited VM inhibition. In conclusion, our results indicate that PARPi, by targeting the VM, may provide a new therapeutic approach for triple negative breast cancer.  相似文献

Seven groups of 12 Wistar rats were fed during two experimental periods. During the first period of 4 weeks, rats were fed diets containing 100 g kg?1 of casein or soya-protein. In the case of casein, diets contained casein, casein + 3.5 g kg?1 of sodium carrageenan and casein + 3.5 g kg?1 of sodium carrageenan + 1 g kg?1 of DL-methionine. In the case of soya protein, diets contained soya protein, soya protein + 6.2 g kg?1 of sodium carrageenan, soya protein + 6.2 g kg?1 of sodium carrageenan + 1 g kg?1 of DL-methionine and soya protein + 1 g kg?1 of DL-methionine. During the second period of 8 weeks, all rats were fed laboratory pellets containing 170 g kg?1 of standard protein mixture. Methionine supplementation of heated soya bean meal containing carrageenan prevented the decrease of growth observed with unsupplemented diet. Methionine supplementation of casein improved protein efficiency ratio and had no effect on long-term growth of rats because carrageenan did not cause growth retardation. At the end of second period, carrageenan ingested during the first period had no effect on plasma cholesterol. When carrageenans added to casein were ingested during the first period, plasma triglycerides were significantly lower after the second period. It would seem that carrageenans induce protein malnutrition by a lack of methionine when they are present in low protein diets with low methionine content, and that a sufficient content of methionine in diets prevents the carrageenan effect.  相似文献

A new class of heterogeneous catalysts for asymmetric hydrogenation of enamides was synthesized using molecular imprinting technology. These new catalysts are molecularly imprinted polymers (MIPs) made from rhodium (I) and copper (II) complexes with the bis(oxazoline) chiral ligands. One of the Rh-MIPs showed 87% ee toward L-enantiomeric product while the Cu-MIP showed 82% ee toward D-enantiomeric product. Both MIPs are easy to separate and reusable.  相似文献

Sepsis in the young population, which is particularly at risk, is rarely studied. O-GlcNAcylation is a post-translational modification involved in cell survival, stress response and metabolic regulation. O-GlcNAc stimulation is beneficial in adult septic rats. This modification is physiologically higher in the young rat, potentially limiting the therapeutic potential of O-GlcNAc stimulation in young septic rats. The aim is to evaluate whether O-GlcNAc stimulation can improve sepsis outcome in young rats. Endotoxemic challenge was induced in 28-day-old rats by lipopolysaccharide injection (E. Coli O111:B4, 20 mg·kg−1) and compared to control rats (NaCl 0.9%). One hour after lipopolysaccharide injection, rats were randomly assigned to no therapy, fluidotherapy (NaCl 0.9%, 10 mL·kg−1) ± NButGT (10 mg·kg−1) to increase O-GlcNAcylation levels. Physiological parameters and plasmatic markers were evaluated 2h later. Finally, untargeted mass spectrometry was performed to map cardiac O-GlcNAcylated proteins. Lipopolysaccharide injection induced shock with a decrease in mean arterial pressure and alteration of biological parameters (p < 0.05). NButGT, contrary to fluidotherapy, was associated with an improvement of arterial pressure (p < 0.05). ATP citrate lyase was identified among the O-GlcNAcylated proteins. In conclusion, O-GlcNAc stimulation improves outcomes in young septic rats. Interestingly, identified O-GlcNAcylated proteins are mainly involved in cellular metabolism.  相似文献
