Reactive oxygen species (ROS) are increasingly being implicated in the regulation of cellular signaling cascades. Intracellular ROS fluxes are associated with cellular function ranging from proliferation to cell death. Moreover, the importance of subtle, spatio-temporal shifts in ROS during localized cellular signaling events is being realized. Understanding the biochemical nature of the ROS involved will enhance our knowledge of redox-signaling. An ideal intracellular sensor should therefore resolve real-time, localized ROS changes, be highly sensitive to physiologically relevant shifts in ROS and provide specificity towards a particular molecule. For in vivo applications issues such as bioavailability of the probe, tissue penetrance of the signal and signal-to-noise ratio also need to be considered. In the past researchers have heavily relied on the use of ROS-sensitive fluorescent probes and, more recently, genetically engineered ROS sensors. However, there is a great need to improve on current methods to address the above issues. Recently, the field of molecular sensing and imaging has begun to take advantage of the unique physico-chemical properties of nanoparticles and nanotubes. Here we discuss the recent advances in the use of these nanostructures as alternative platforms for ROS sensing, with particular emphasis on intracellular and in vivo ROS detection and quantification.  相似文献

The aim of this work is to study the effect of thermal aging on the mechanical, dynamic‐mechanical, and chemical properties of SBR (styrene‐butadiene rubber) composites filled with SBR industrial rubber scraps. Eight composites with varying proportions (10–80 phr) of the SBR ground scraps (SBR‐r) were prepared and subjected to accelerated aging in an air‐oven. The composites were evaluated, and the results were compared with a control sample (base formulation with 0 phr of SBR‐r), before and after thermal aging. The accelerated aging led to a decrease in the mechanical properties as a result of an increase in the stiffness of the material, related to an increase in the crosslink density. However, these properties were not affected by the addition of rubber scraps up to 50 phr, either before or after aging. The increase in the glass transition temperature of the composites after aging, measured using dynamic mechanical analysis, confirmed the occurrence of a postcrosslinking process. Fourier transform infrared spectroscopy and crosslink density revealed that the aging mechanism was dependent on the SBR‐r content. © 2011 Wiley Periodicals, Inc. J Appl Polym Sci, 2011  相似文献

A 22 central composite design with five center points was performed to estimate the effects of temperature (120, 130 and 140 °C) and acid loading (100, 150 and 200 mg g?1) on the yield of monomeric xylose recovery from wheat straw hemicellulose (YS/RM). Under the best hydrolysis condition (140 °C and 200 mg g?1), a YS/RM of 0.26 g g?1 was achieved. After vacuum concentration and detoxification by pH alteration and active charcoal adsorption, the hydrolyzate was used as source of xylose for xylitol bioproduction in a stirred tank reactor. A xylitol production of 30.8 g L?1 was achieved after 54 h?1 of fermentation, resulting in a productivity (QP) of 0.57 g L?1 h?1 and bioconversion yield (YP/S) of 0.88 g g?1. The aim of the study was to evaluate the overall hygienic–sanitary conditions of university hospitals foodservices and assess food safety knowledge and self‐reported practices of food handlers. A self‐administered questionnaire based on sanitary standards for foodservices was given to 123 food handlers from three public hospitals. Microbiological analysis of food handling surfaces and food hygiene inspections were also performed. Food handlers' self‐reported knowledge and practices reached satisfactory levels (77.2% and 78.1%, respectively), and showed a significant positive intercorrelation (p < .05). Food hygiene inspections in the hospitals foodservices presented a 60.9% average of adequacy and microbiological analysis results showed that 66.7% of the evaluated surfaces presented significant contamination by mesophilic bacteria. The presence of coliforms at 45°C above the safety limit was identified in 47.9% of the surfaces. The results showed a need for motivating hospital food handlers to practice the safe handling knowledge that they have demonstrated in theory.  相似文献

Objective: Hematopoietic stem cell transplant (HSCT) survivors who are 1 to 3 years posttransplant are challenged by the need to resume valued social roles and activities—a task that may be complicated by enduring transplant-related psychological distress common in this patient population. The present study investigated whether transplant survivors who receive adequate social support from their spouse or intimate partner experience lower distress. Method: Effects of receiving a greater quantity of partner support (a common approach to studying enacted support) were compared with effects of receiving more effective partner support (i.e., support that more closely matches their needs in terms of its quantity and quality). Men and women (N = 230) who were 1 to 3 years posttransplant completed measures of partner support quantity (Manne & Schnoll, 2001), partner social support effectiveness (Rini & Dunkel Schetter, 2010), and psychological distress (Brief Symptom Inventory; Derogatis & Spencer, 1982). The practical constraints of conducting rigorous research in applied settings often limit the ability to include well-validated, but lengthy, measures in survey designs. In the current study we tested the utility of using a strategic item selection process to alleviate this issue. We evaluated the measurement and structural model fit of items selected using a strategic item selection format and assessed the predictive validity of those abbreviated scales. We also compared fit and predictive validity to the full version item sets to assess the comparability of the abbreviated scales. Results indicated that measurement models and structural models for the a priori items produced similar fit when compared with full version items across both measures. When comparing the proportion of variance explained in the criterion, there were no differences between the strategically selected items and the full versions of the scales. We conclude that the a priori selection of items based on previous research may be a useful technique for developing short measures of psychological constructs that mitigate practical, analytical, and methodological issues associated with lengthy surveys. Total cyanide analysis by distillation is used most commonly to assess cyanide content of water samples. This manual method is robust but slow and provides no information about cyanide speciation, a significant limitation in that cyanide species have substantially different toxicity characteristics. Seven alternative methods for the analysis of cyanide species or groups of species were evaluated in reagent water and five different contaminated water matrices, including five species-specific methods--weak acid dissociable (WAD) cyanide, free cyanide by microdiffusion, available cyanide, automated WAD cyanide by thin film distillation, metal cyanides by ion chromatography--and two automated techniques for total cyanide--total cyanide bythin film distillation and total cyanide by low-power UV digestion. The species-specific cyanide analytical techniques achieved low, ppb-level detection limits and exhibited satisfactory accuracy and precision for most contaminated waters. Analysis of low concentrations of cyanide species in raw wastewater was problematical for the available cyanide and ion chromatography methods, which experienced significant interference problems and/or low recoveries. There was recovery of significant diffusible cyanide in microdiffusion tests with nickel-cyanide-spiked samples, reflecting dissociation of this weak metal-cyanide complex during the test and demonstrating that the test can recover some fraction of WAD cyanide in addition to free cyanide. The automated total cyanide methods, which involve UV digestion, achieved low detection limits for most waters but exhibited low recoveries for some waters.  相似文献

Using an atomic force microscope (AFM) at a controlled contact force, we report the electrical signal response of multi-walled carbon nanotubes (MWCNTs) disposed on a golden thin film. In this investigation, we highlight first the theoretical calculation of the contact resistance between two types of conductive tips (metal-coated and doped diamond-coated), individual MWCNTs and golden substrate. We also propose a circuit analysis model to schematize the «tip-CNT-substrate» junction by means of a series-parallel resistance network. We estimate the contact resistance R of each contribution of the junction such as Rtip-CNT, RCNT-substrate and Rtip-substrate by using the Sharvin resistance model. Our final objective is thus to deduce the CNT intrinsic radial resistance taking into account the calculated electrical resistance values with the global resistance measured experimentally. An unwished electrochemical phenomenon at the tip apex has also been evidenced by performing measurements at different bias voltages with diamond tips. For negative tip-substrate bias, a systematic degradation in color and contrast of the electrical cartography occurs, consisting of an important and non-reversible increase of the measured resistance. This effect is attributed to the oxidation of some amorphous carbon areas scattered over the diamond layer covering the tip. For a direct polarization, the CNT and substrate surface can in turn be modified by an oxidation mechanism.  相似文献

The custom-built Portable Fine Particle Concentrator (PFPC) is evaluated for the measurement of ice nucleating particles (INPs) in the atmosphere. The concentrations of INPs in remote regions of the atmosphere are very low, often close to instrumental detection limits. The PFPC is a dual slit-nozzle virtual impactor where particles are concentrated from an input flow of 250 LPM (litres per minute) into an output flow of 10 LPM. The enrichment factors (EFs) for ambient particles with diameters between 0.4 and 2.5?µm were found to be 21?±?5 at sea level and 18?±?2 at a field station 3580 meters above sea level for the PFPC operated in horizontal configuration. Similar enhancement factors (16?±?5) in the concentrations of INPs measured by the Horizontal Ice Nucleation Chamber at the high-altitude station were observed when the air mass was characterized by high numbers of particles larger than 0.5?µm. When the number size distribution was dominated by particles smaller than 0.5?µm, the INP EF was considerably lower. Corroborating short-term measurements were provided by additional INP-measuring instruments, the Fast Ice Nucleus CHamber and the Frankfurt Ice Deposition Freezing Experiment. Results from two aerosol mass spectrometers also indicate significant particle enhancement using the PFPC. These results indicate that the PFPC can be usefully deployed to improve the detection efficiency of ambient INP measurements.

The objective was to characterize and associate the receptor reactivities of fibroblastic growth factor (FGF)‐2, FGF‐7, FGF‐8, epidermal growth factor (EGF), α‐actin and vimentin in relation to the androgen receptor (AR), α and β estrogen receptors (ERα and ERβ), and prolactin receptor in the prostate of elderly men showing low‐ and high‐grade adenocarcinoma. Thirty prostatic samples were taken from 60‐ to 90‐year‐old patients without prostatic lesions and with low‐grade cancer and high‐grade cancer, from the University Hospital, School of Medicine, the State University of Campinas. The results showed that increased FGF‐2, FGF‐7, and FGF‐8 receptor reactivities and decreased AR reactivity were verified in both high‐ and low‐grade cancer. However, the FGF‐8 receptor showed greater involvement at the beginning of the malignancy alterations. Increased EGF receptor (EGFR) reactivity and diminished α‐actin immunohistochemistry were identified in both cancer groups. Also, increased ERα, PR, and vimentin receptors were verified in both cancer groups. To conclude, the ERα involvement in the reactive stroma activation led to a microenvironment, which was favorable to cancer progression, due to maximizing stromal imbalance. The prolactin could be related to cancer progression due to its interaction with ERα action, indicating that this hormone could be a relevant target to prevent the estrogenic effects in the prostatic lesions. Both FGF receptor (FGFR)‐2 and FGFR‐8 play a fundamental role in the early stages of prostate cancer, suggesting that these molecules could be a promising therapeutic target. The differential localization of the fibroblastic factors between the prostatic epithelium and stroma of elderly men, who presented prostate cancer, could indicate a favorable distinction for tumoral progression.
