The aim of this study was to evaluate the antimicrobial activity, pH level, calcium ion release, and radiopacity of calcium hydroxide pastes associated with three radiopacifying agents (iodoform, zinc oxide, and barium sulfate). For the pH and calcium release tests, 45 acrylic teeth were utilized and immersed in ultrapure water. After 24 h, 72 h, and 7 days the solution was analyzed by using a pH meter and an atomic absorption spectrophotometer. Polyethylene tubes filled with the pastes were used to perform the radiopacity test. For the antimicrobial test, 25 dentin specimens were infected intraorally in order to induce the biofilm colonization and treated with the pastes for 7 days. The Live/Dead technique and a confocal microscope were used to obtain the ratio of live cells. Parametric and nonparametric statistical tests were performed to show differences among the groups (P < 0.05). The pH analysis at 7 days showed significant differences (P < 0.05) among the groups. No differences among the pastes were found in the calcium release test on the 7th day (P > 0.05). The calcium hydroxide/iodoform samples had the highest radiopacity and antimicrobial activity against the biofilm‐infected dentin in comparison to the other pastes (P < 0.05). Calcium hydroxide mixed with 17% iodoform and 35% propylene glycol into a paste had the highest pH, calcium ion release, radiopacity, and the greatest antimicrobial action versus similar samples mixed with BaSO4 or ZnO. The present study evaluated whether the changes in the labeling pattern of cytoskeletal proteins in osteogenic cells cultured on bioactive glass‐based materials are due to altered mRNA and protein levels. Primary rat‐derived osteogenic cells were plated on Bioglass® 45S5, Biosilicate®, and borosilicate (bioinert control). The following parameters were assayed: (i) qualitative epifluorescence analysis of actin and tubulin; (ii) quantitative mRNA and protein expression for actin and tubulin by real‐time PCR and ELISA, respectively, and (iii) qualitative analysis of cell morphology by scanning electron microscopy (SEM). At days 3 and 7, the cells grown on borosilicate showed typical actin and tubulin labeling patterns, whereas those on the bioactive materials showed roundish areas devoid of fluorescence signals. The cultures grown on bioactive materials showed significant changes in actin and tubulin mRNA expression that were not reflected in the corresponding protein levels. A positive correlation between the mRNA and protein as well as an association between epifluorescence imaging and quantitative data were only detected for the borosilicate. SEM imaging of the cultures on the bioactive surfaces revealed cells partly or totally coated with material aggregates, whose characteristics resembled the substrate topography. The culturing of osteogenic cells on Bioglass® 45S5 and Biosilicate® affect actin and tubulin mRNA expression but not the corresponding protein levels. Changes in the labeling pattern of these proteins should then be attributed, at least in part, to the presence of a physical barrier on the cell surface as a result of the material surface reactions, thus limiting fluorescence signals. The classification of colorectal cancers (CRC) is currently based largely on histologically determined tumour characteristics, such as differentiation status and tumour stage, i.e., depth of tumour invasion, involvement of regional lymph nodes and the occurrence of metastatic spread to other organs. These are the conventional prognostic factors for patient survival and often determine the requirement for adjuvant therapy after surgical resection of the primary tumour. However, patients with the same CRC stage can have very different disease-related outcomes. For some, surgical removal of early-stage tumours leads to full recovery, while for others, disease recurrence and metastasis may occur regardless of adjuvant therapy. It is therefore important to understand the molecular processes that lead to disease progression and metastasis and to find more reliable prognostic markers and novel targets for therapy. This review focuses on cell surface proteins that correlate with tumour progression, metastasis and patient outcome, and discusses some of the challenges in finding prognostic protein markers in CRC. 相似文献

This work shows that radio-frequency (RF) fields can simultaneously align carbon nanotubes (CNTs) dispersed in a resin and induce Joule heating to cure the resin. The timescales of alignment and curing using RF heating are numerically computed and compared at different field strengths in order to determine a temperature where alignment happens before the matrix crosslinks. Composites are experimentally fabricated at the desired target temperature and are optically analyzed and quantified; the CNT network is successfully aligned in the direction of the applied electric field. This methodology can be used to create composites where the local alignment can be varied across the sample. Composites fabricated using RF fields have higher electrical conductivity in the direction of the aligned CNTs than an oven-cured, randomly aligned sample. Also, RF-cured nanocomposites exhibit higher tensile strength and modulus in the direction of alignment compared to an oven-cured sample. Finally, it is further demonstrated how this methodology can be coupled with a direct ink writing additive manufacturing process to induce alignment in any desired direction, even orthogonal to the shear forces in the extrusion direction. 相似文献

Campylobacter isolates (n = 297; 202 C. jejuni and 95 C. coli isolates) recovered from 2,513 retail meat samples (chicken breasts, ground turkey, ground beef, and pork chops) were examined for antimicrobial susceptibility. The isolates were further analyzed for genetic relatedness by pulsed-field gel electrophoresis (PFGE) using SmaI and KpnI restriction enzymes, and a subset of isolates (n = 174) were subtyped by multilocus sequence typing (MLST). The resistance most frequently observed was that to doxycycline (27.6%), followed by ciprofloxacin (13.8%) and erythromycin (6.4%). All isolates were susceptible to gentamicin and meropenem. C. coli showed higher resistance to doxycycline than did C. jejuni (42.1 versus 20.8%) and lower resistance to ciprofloxacin than did C. jejuni (10.5 versus 15.3%). Erythromycin resistance was only observed in C. coli. PFGE using SmaI plus KpnI digestion generated 168 clusters from 297 isolates: 115 from C. jejuni and 53 from C. coli. MLST revealed 44 sequence types (STs) under 10 clonal complexes from 120 C. jejuni and 27 STs under two clonal complexes from 54 C. coli. There was a positive association between PFGE and STs; however, PFGE showed greater discriminatory power than MLST. Subtyping data did not correlate with antimicrobial resistance phenotypes. 相似文献

We constructed a fuel‐flexible fuel cell consisting of an alkaline anion exchange membrane, palladium anode, and platinum cathode. When an alcohol fuel was used with potassium hydroxide added to the fuel stream and oxygen was the oxidant, the following maximum power densities were achieved at 60 °C: ethanol (128 mW cm−2), 1‐propanol (101 mW cm−2), 2‐propanol (40 mW cm−2), ethylene glycol (117 mW cm−2), glycerol (78 mW cm−2), and propylene glycol (75 mW cm−2). We also observed a maximum power density of 302 mW cm−2 when potassium formate was used as the fuel under the same conditions. However, when potassium hydroxide was removed from the fuel stream, the maximum power density with ethanol decreased to 9 mW cm−2 (using oxygen as oxidant), while with formate it only decreased to 120 mW cm−2 (using air as the oxidant). Variations in the performance of each fuel are discussed. This fuel‐flexible fuel cell configuration is promising for a number of alcohol fuels. It is especially promising with potassium formate, since it does not require hydroxide added to the fuel stream for efficient operation. 相似文献

While recent research has provided valuable information as to the composition of laser printer particles, their formation mechanisms, and explained why some printers are emitters while others are low emitters, questions relating to the potential exposure of office workers remained unanswered. In particular, (i) what impact does the operation of laser printers have on the background particle number concentration (PNC) of an office environment over the duration of a typical working day? (ii) What is the airborne particle exposure to office workers in the vicinity of laser printers? (iii) What influence does the office ventilation have upon the transport and concentration of particles? (iv) Is there a need to control the generation of, and/or transport of particles arising from the operation of laser printers within an office environment? (v) What instrumentation and methodology is relevant for characterizing such particles within an office location? We present experimental evidence on printer temporal and spatial PNC during the operation of 107 laser printers within open plan offices of five buildings. The 8 h time-weighted average printer particle exposure is significantly less than the 8 h time-weighted local background particle exposure, but that peak printer particle exposure can be greater than 2 orders of magnitude higher than local background particle exposure. The particle size range is predominantly ultrafine (<100 nm diameter). In addition we have established that office workers are constantly exposed to nonprinter derived particle concentrations, with up to an order of magnitude difference in such exposure among offices, and propose that such exposure be controlled along with exposure to printer derived particles. We also propose, for the first time, that peak particle reference values be calculated for each office area analogous to the criteria used in Australia and elsewhere for evaluating exposure excursion above occupational hazardous chemical exposure standards. A universal peak particle reference value of 2.0 × 10(4) particles cm(-3) has been proposed. 相似文献

The present paper deals with the computational simulation of a pile of thin sheets. The sheets are not laminated or glued, but they interact by frictional contact. In general, it is not possible to perform a full-scale finite element contact computation for piles containing thousands of sheets; the problem size becomes too large, and numerical solution methods suffer from severe convergence problems due to the large number of strongly coupled contact conditions. In this paper, a macroscopic material model is presented for the two-dimensional case. The pile of sheets is homogenized by introducing an effective anisotropic constitutive law, which is motivated by formulations of the theory of elasto-plasticity. This macroscopic material law models the behavior of a pile of sheets, allowing for no tensile stresses in the direction normal to the sheets and obeying Coulomb??s law of friction in the tangential contact plane. Applying this macroscopic material model, an equivalent homogeneous body can be treated using much coarser discretizations. Computational results for the problems are provided, and a comparison with simplified contact computations is done. 相似文献
