Lactobacillus helveticus is a lactic acid bacterium very used in fermented milks and cheese. The rapid growth of L. helveticus in milk is supported by an efficient cell envelope proteinase (CEP) activity, due to subtilisin-like serine proteases. These enzymes play also crucial roles in texture and flavor formation in dairy products as well as in generating in situ bioactive peptides. In L. helveticus, several genes encoding putative CEPs were detected and characterized by a large intraspecific diversity; little is known about regulation of expression of CEP-encoding genes. Anchored at the bacterial surface, CEPs are large-sized enzymes (> 150 kDa) hydrolyzing β- and αs1-casein as well. Substrate cleavages occur after almost all types of amino acids residues, but mass spectrometry analysis revealed L. helveticus strains with specific profiles of substrate hydrolysis, which could explain identification of strains associated with interesting technological properties. In this review, the most recent data regarding CEP-encoding genes, CEP activities toward caseins and L. helveticus strain diversity are discussed. 相似文献

Piperine is the natural bioactive component of black pepper (Piper nigrum L.) with several astounding therapeutic properties. In this study, sequential microwave-ultrasound-assisted extraction approach was used for isolation of piperine from black pepper. The effect of various factors such as extraction solvent, particle size of pepper, solvent to solid ratio, microwave power and time and ultrasound temperature and time on the extraction yield of piperine was considered. The maximum extraction yield was 46.6 mg piperine/g pepper which was obtained using ethanol as solvent at the particle size of 0.15 mm, solvent to solid ratio of 20:1, microwave power of 100 W for 1 min, and ultrasound temperature of 50 ° C for 30 min. This extraction yield was higher than those obtained by Soxhlet (39.1 mg/g), microwave-assisted (38.8 mg/g) and ultrasound-assisted (37.0 mg/g) extractions. The purity of the extracted piperine was 81.4% as determined by HPLC analysis. The FTIR and UV-vis analyses confirmed that the structure of piperine remained intact after extraction and purification which is very important for medicinal applications. 相似文献

In this work, nanocomposites of styrene butadiene rubber (SBR), cellulose II, and clay were prepared by cocoagulation of SBR latex, cellulose xanthate, and clay aqueous suspension mixtures. The incorporated amount of cellulose II was 15 phr, and the clay varied from 0 to 7 phr. The influence of cellulose II and clay was investigated by rheometric, mechanical, physicochemical, and morphological properties. From the analysis of transmission electron microscopy (TEM), dispersion in nanometric scale (below 100nm) of the cellulosic and mineral components throughout the elastomeric matrix was observed. XRD analysis suggested that fully exfoliated structure could be obtained by this method when low loading of silicate layers (up to 5 phr) is used. The results from mechanical tests showed that the nanocomposites presented better mechanical properties than SBR gum vulcanizate. In this work, sodium montmorillonite clay was added, as filler, to nanocomposites of natural rubber (NR) and cellulose II (regenerated cellulose) in amounts varying from 0 to 5 phr (per hundred resin). Natural rubber (NR)/cellulose II/montmorillonite nanocomposites were prepared by co‐coagulating NR latex, montmorillonite aqueous suspension and cellulose xanthate. The clay was previously exfoliated in water, and the resulting suspension was then added to the mixture of NR latex with cellulose xanthate. Morphological, rheometric, mechanical, and dynamic mechanical properties were evaluated, and an increase in these properties was observed upon the addition of cellulose and clay nanomaterials to the rubber matrix. The design of the coaxial mixers depends on many interrelated parameters including the geometry and dimensions of the mixing vessel, the location and type of the impellers, speed ratio, impeller diameter, rotation mode, and fluid rheology. No study has been reported in the literature regarding the mixing performance of the coaxial mixers in the agitation of yield-pseudoplastic fluids. Thus, the main objective of this study was to evaluate the performance of a Scaba–anchor coaxial mixer (a novel configuration) in the mixing of xanthan gum solutions (yield-pseudoplastic fluids). The Herschel–Bulkley model was used to describe the rheological behavior of the xanthan gum solutions. To develop new correlations for the generalized Reynolds and power numbers of the coaxial mixers employed in the agitation of this class of non-Newtonian fluids, we utilized numerous experimental and computational fluid dynamics (CFD) data. Polyaniline (PANI) films were deposited under cyclic voltammetric conditions on 304L stainless steel by aniline electropolymerisation from acidic and slightly basic solutions containing respectively the followed support-electrolytes: H2C2O4 and KNO3.It was found that the film produced in oxalic acidic medium was more conductive than that obtained in potassium nitrate one.The PANI coatings corrosion performances in 0.5 M NaCl were investigated and compared using standard electrochemical methods, electrochemical impedance spectroscopy (EIS) and SEM analysis.The highest corrosion resisting efficiency was obtained for PANInitrate which exhibited a significant physical barrier property against the attack of corrosive products. Infants born after intrauterine growth restriction (IUGR) are at risk of developing arterial hypertension at adulthood. The endothelium plays a major role in the pathogenesis of hypertension. Endothelial colony-forming cells (ECFCs), critical circulating components of the endothelium, are involved in vasculo-and angiogenesis and in endothelium repair. We previously described impaired functionality of ECFCs in cord blood of low-birth-weight newborns. However, whether early ECFC alterations persist thereafter and could be associated with hypertension in individuals born after IUGR remains unknown. A rat model of IUGR was induced by a maternal low-protein diet during gestation versus a control (CTRL) diet. In six-month-old offspring, only IUGR males have increased systolic blood pressure (tail-cuff plethysmography) and microvascular rarefaction (immunofluorescence). ECFCs isolated from bone marrow of IUGR versus CTRL males displayed a decreased proportion of CD31+ versus CD146+ staining on CD45− cells, CD34 expression (flow cytometry, immunofluorescence), reduced proliferation (BrdU incorporation), and an impaired capacity to form capillary-like structures (Matrigel test), associated with an impaired angiogenic profile (immunofluorescence). These dysfunctions were associated with oxidative stress (increased superoxide anion levels (fluorescent dye), decreased superoxide dismutase protein expression, increased DNA damage (immunofluorescence), and stress-induced premature senescence (SIPS; increased beta-galactosidase activity, increased p16INK4a, and decreased sirtuin-1 protein expression). This study demonstrated an impaired functionality of ECFCs at adulthood associated with arterial hypertension in individuals born after IUGR. 相似文献

The rapidly increasing solar conversion efficiency (PCE) of hybrid organic–inorganic perovskite (HOIP) thin-film semiconductors has triggered interest in their use for direct solar-driven water splitting to produce hydrogen. However, application of these low-cost, electronic-structure-tunable HOIP tandem photoabsorbers has been hindered by the instability of the photovoltaic-catalyst-electrolyte (PV+E) interfaces. Here, photolytic water splitting is demonstrated using an integrated configuration consisting of an HOIP/n+silicon single junction photoabsorber and a platinum (Pt) thin film catalyst. An extended electrochemical (EC) lifetime in alkaline media is achieved using titanium nitride on both sides of the Si support to eliminate formation of insulating silicon oxide, and as an effective diffusion barrier to allow high-temperature annealing of the catalyst/TiO2-protected-n+silicon interface necessary to retard electrolytic corrosion. Halide composition is examined in the (FA1-xCsx)PbI3 system with a bandgap suitable for tandem operation. A fill factor of 72.5% is achieved using a Spiro-OMeTAD-hole-transport-layer (HTL)-based HOIP/n+Si solar cell, and a high photocurrent density of −15.9 mA cm−2 (at 0 V vs reversible hydrogen electrode) is attained for the HOIP/n+Si/Pt photocathode in 1 m NaOH under simulated 1-sun illumination. While this thin-film design creates stable interfaces, the intrinsic photo- and electro-degradation of the HOIP photoabsorber remains the main obstacle for future HOIP/Si tandem PEC devices. 相似文献
